Interpretable semantic segmentation and semi-supervised learning for whole slide pathological image analysis

최근 조기 암 진단의 중요성 증대로 인해 인공지능 분야에서 다양한 연구 주제로 다뤄지고 있다. 기존의 병리 진단 과정은 정확한 판정 및 치료를 위해 전문가의 판단을 필요로 한다. 이로 인해, 많은 시간과 비용이 발생하게 되며 병리 의사의 업무 피로도가 증가한다. 본 연구는 유방암의 조기 진단 작업 개선에 초점을 두고 있다. 이를 위해 정확한 암 영역 검출 방법과 의미 분할의 해석 방법, 병리 이미지 데이터 레이블링 문제를 해결하는 방법을 제안한다. 의사의 병리 진단은 WSI 를 기반으로 이루어지기 때문에 모델의 예측 결과 및 설명도 WSI 수준에서 이루어져야 하지만 이에 대한 이론적 방법론이 부족하다. 이 문제를 극복하기 위해 디지털 병리 영상(Whole Slide Image; WSI)에서 악성 종양 영역을 정확하게 검출하고 해석하기 위한 ResNeSt 와 U-Net 기반의 분할 방법과 기울기 가중치 클래스 활성화 매핑(Seg-Grad-CAM)을 통한 의미 분할 해석 방법을 제안한다. 제안 방법은 보다 정확하고 신뢰할 수 있는 악성 종양 진단 결과를 도출하기 위해 픽셀 수준에서 의미 분할을 수행한다. 또한 결과의 해석력을 증가시키기 위해 patch 가 아닌 WSI 기반의 결과 해석 방법을 제안한다. 우리는 병리 이미지 데이터를 수집하여 픽셀 수준에서의 의미 분할 성능 비교 실험을 수행했으며 경쟁 알고리즘보다 높은 성능을 보여 제안된 방법의 효과를 확인했다. 병리학 이미지 데이터에서 레이블링이 서로 상이하다는 문제가 존재한다. 이 문제는 병변의 위치를 레이블링하는 과정과 병변의 스펙트럼성 특징에서 야기되어 모델 학습 과정에서 혼동을 발생시킨다. 이 문제를 극복하기 위해 준지도학습 기법을 적용했다. 우리는 제한된 레이블링이 된 데이터와 많은 양의 레이블링이 되어 있는 데이터를 활용하는 준지도학습 방법론을 제안한다. 우리는 레이블링 된 데이터의 비율을 비교 실험하여 최적의 비율로 구성하였다. 제안 방법은 적은 레이블링 데이터를 활용하여 레이블링 비용을 줄일 수 있다. 우리는 병리 이미지 데이터를 수집하여 지도학습과 준지도학습의 의미분할 비교 실험을 수행했으며 준지도학습의 성능이 전체 레이블 데이터로 학습한 지도 학습만큼의 성능과 유사하게 도출될 수 있음을 보였다.
더보기Due to the escalating importance of early cancer detection, various research topics are being addressed in the field of artificial intelligence. AI the conventional pathological diagnosis process requires the judgment of experts for accurate diagnosis and treatment. This leads to significant time and cost implications, as well as increased fatigue for pathologists. This study focuses on improving the early detection process for breast cancer. Thus, it proposes methods for accurate cancer region detection, interpretation of semantic segmentation, and addressing the issue of pathological image data labeling. Whole slide images (WSIs), which are high-resolution digital representations of entire pathology slides, are used for pathological diagnosis. When applying AI model digital pathology the model's predictive results and explanations should also occur at the WSI level, yet there is a lack of theoretical methodology for this. To overcome this issue, we propose a Split- Attention Networks (ResNeSt) and U-Net-based method for accurate malignant tumor detection from WSI, along with a semantic segmentation interpretation using Semantic Segmentation via Gradient-weighted Class Activation Mapping (Seg-Grad-CAM). The proposed method performs semantic segmentation at the pixel level to derive accurate and reliable malignant tumor diagnosis results. Furthermore, we propose a WSI-based interpretation method instead of patch- based to enhance the prediction interpretability. We collected pathological image data performed experiments and showed our method outperforms others in semantic segmentation. The inconsistent labeling problem in pathological label data is the most significant issue. This is caused by the process of labeling the location of the lesions and the spectral characteristics of the lesions, leads to confusion resulting in reducing model accuracy, decreasing trustworthiness and increasing clinical misdiagnoses. We here propose applying a semi-supervised learning that utilizes both labeled and unlabeled data to train models and make predictions. We conducted comparative experiments on the proportion of labeled data and configured it at the optimum ratio. The proposed method allows for reduction in labeling costs by utilizing data with fewer labels. We collected WSIs and conducted comparative experiments on semantic segmentation of supervised learning and semi-supervised learning. The results demonstrated that the performance of semi-supervised learning was similar to that of supervised learning trained solely on the entire labeled dataset.
