Enzymatic Synthesis and Characterization of Lauroyl Tripeptides as Multi-functional Food Additives with Antibacterial and Antioxidative Activities

In emulsion foods, lipid oxidation and microbial contamination by foodborne pathogens, diminishing the food quality and safety, have been considered as major risks. Under the strategy to control the risks with a single compound, the use of a multi-functional food additive with antibacterial and antioxidative activities has been proposed. In this study, lipase-catalyzed acylation of antioxidative peptides with a fatty acid was performed to produce a novel multi-functional food additive.
Firstly, multi-functional lauroyl tripeptide-KHA (TPL-KHA) with amino acid sequence of Lys-His-Ala was produced via the lipase-catalyzed acylation. Among diverse antioxidative peptides, tripeptides-KHA was selected by considering its antioxidative properties and simplicity of chemical structure. Additionally, a general methodology for the enzymatic synthesis of the lauroyl peptides were established by determining effects of reaction parameters on conversion yield and chemo-selectivity of the reaction.
As a result, the reaction parameters were optimized as follows: organic solvent of 2-methyl-2-butanol, reaction temperature at 55℃, substrate molar ratio (lauric acid:tripeptide-KHA) of 4.0, and reaction time for 72 h. The conversion yield of 74.73% was obtained under the optimum reaction conditions. The chemo-selectivity of the reaction was determined from structural analysis by LC-ESI-MS and 1H-NMR. The spectra from LC-ESI-MS showed that tripeptide-KHA was acylated with one equivalent of lauric acid. Furthermore, 1H-NMR analysis identified that Nε-lauroyl tripeptide-KHA was chemo-selectively produced by the lipase-catalyzed acylation.
The multi-functionalities of TPL-KHA, including surface-active, antibacterial, and antioxidative properties, were assessed. TPL-KHA showed the surface activity at the air-water interface with critical micelle concentration (CMC) of 2.71 mM and γCMC of 30.44 mN/m. Moreover, TPL-KHA exhibited bacteriostatic and bactericidal effects on Gram-positive (Staphylococcus aureus, Bacillus cereus) and Gram-negative (Escherichia coli, Salmonella Typhimurium) foodborne pathogens. The minimum inhibitory concentrations (MICs) of TPL-KHA ranged from 2.83-4.00 mM and the minimum bactericidal concentrations (MBCs) were approximately 1.3-fold higher than those of MICs. Moreover, it was identified that TPL-KHA had ABTS+ radical scavenging activity of a Trolox equivalents antioxidant capacity (TEAC) value of 0.206 μmol TE/μmol. Additionally, TPL-KHA of 100 μM showed the inhibitory activity of 80.99% against linoleic acid peroxidation at 9 days after incubation at 40℃.
Secondly, lauroyl tripeptide (TPL) model was designed in order to understand a correlation between physicochemical properties and antibacterial activity of lauric acid derivatives. At once, to develop a novel candidate for the multi-functional food additive, antioxidative tripeptides with different physicochemical properties were screened and employed.
Among candidates for the tripeptides, tripeptides of Pro-His-Lys (PHK), Asn-His-Lys (NHK), Ala-His-Lys (AHK), Arg-His-Lys (RHK), and Leu-Trp-Lys (LWK) showed the antioxidative activities against linoleic acid peroxidation similar to (p > 0.05) that of α-tocopherol. The five TPLs with different physicochemical properties, including lauroyl tripeptide-PHK (TPL-PHK), -NHK (TPL-NHK), -AHK (TPL-AHK), -RHK (TPL-RHK), and -LWK (TPL-LWK), were produced via the lipase-catalyzed acylation.
MIC and MBC of TPLs against two Gram-positive (S. aureus, B. cereus) and two Gram-negative bacteria (E. coli, S. Typhimurium) were determined to compare the antibacterial activities. TPL-AHK, TPL-PHK, and TPL-NHK showed similar antibacterial activity at MICs ranging from 2.50 to 5.17 mM, but TPL-LWK showed no inhibitory activity against the bacterial growth. Interestingly, the MICs of TPL-RHK, ranging from 0.30-0.58 mM, were much lower (i.e., much stronger activity) than those of the others. MBCs of TPLs except for TPL-LWK were approximately 1.0 to 1.8-fold higher than those of MICs, indicating TPLs exhibited both bacteriostatic and bactericidal effects.
The physicochemical properties, including net charge and log P values (hydrophilicity), of TPLs were determined. TPL-RHK with the strongest antibacterial activity possessed a positive net charge of +1.1. It was identified that TPL-AHK, TPL-PHK, and TPL-NHK had the same positive net charge of +0.1, but TPL-LWK had no positive net charge. Therefore, it was obvious that the positive net charge was one of the significant factors determining the antibacterial activity of the lauric acid derivatives. A certain correlation between hydrophilicity and antibacterial activity was not determined. However, based on the results that TPL-NHK had stronger antibacterial activity than TPL-PHK overall, it was thought that antibacterial activity of lauric acid derivatives might be enhanced as their hydrophilicity increased. Propidium iodide (PI) uptake assay and the fluorescence microscopy with dual staining by SYTO 9 and PI probes suggested that the antibacterial activities of TPLs could be attributed to alterations in permeability of the cell membrane. Accordingly, it is inferred that the physicochemical properties, especially positive net charge, of the lauric acid derivatives are likely to influence the interaction with the cell membrane, which is considered as the crucial determinant for the antibacterial activity.
Finally, TPL-RHK with the strongest antibacterial activity among TPLs was characterized as a promising multi-functional food additive. Since the one promising applications is in emulsion foods, the emulsifying, antibacterial, and antioxidative properties of TPL-RHK were assessed in oil-in-water emulsion (O/W emulsion) model system.
The emulsifying properties of TPL-RHK were evaluated by visual observations and measurement of ζ-potential of TPL-RHK-stabilized emulsions at different pH from 3.0 to 7.0. TPL-RHK-stabilized emulsions had higher stability at pH 3.0 and 4.0, compared to those at pH 5.0-7.0. The measurement of ζ-potential showed that the instability of the TPL-RHK-stabilized emulsions at the neutral pH could be due to the reduced electrostatic repulsions. Meanwhile, creaming or phase separation were observed in all the TPL-RHK-stabilized emulsions at pH from 3.0 to 7.0 within 3 days after storage at 25℃. The low colloidal stability of TPL-RHK-stabilized emulsions could be attributed to the high hydrophilicity of TPL-RHK. By contrast, emulsions stabilized by TPL-RHK with Tween 20 were stable during the storage for 9 days, similar to Tween 20-stabilized emulsions. In addition, Z-average diameters and polydispersity index of the emulsions stabilized by TPL-RHK with Tween 20 were similar to those of Tween 20-stabilized emulsions. Based on the results, it was plausible that TPL-RHK could be applied to O/W emulsion foods without affecting emulsion properties at pH ranging from 3.0 to 7.0.
Bactericidal effects of TPL-RHK on both Gram-positive (S. aureus, B. cereus, Listeria monocytogenes) and Gram-negative (E. coli, S. Typhimurium, Pseudomonas aeruginosa) foodborne pathogens were assessed in Tween 20-stabilized O/W emulsions. MBCs of TPL-RHK against the bacterial strains were determined at concentrations ranging from 0.20 to 0.80 mM. In addition, time-kill assay indicated that TPL-RHK killed S. aureus and E. coli in time- and concentration-dependent manners. Treatment of TPL-RHK on S. aureus for 12 h at the concentration of equivalent to 1.0×MBC resulted in 3.68 log reductions in the viable cells and the treatment for 15 h completely killed the cells. Moreover, decreases in 4.54 and 5.10 log of the viable cells of E. coli were observed in treatment of TPL-RHK at concentration of equivalent to 1.0×MBC for 18 and 24 h, respectively. The results suggested that TPL-RHK could control the microbial contamination by foodborne pathogens in O/W emulsion foods.
TPL-RHK showed ABTS+ radical scavenging activity with a TEAC value of 0.475±0.007 μmol TE/μmol, similar to that of tripeptide-RHK (0.502±0.022 μmol TE/μmol), indicating the antioxidative activity of tripeptide- RHK was retained after the acylation. However, TPL-RHK and tripeptide-RHK had no reducing power on ferric ion (Fe3+). Furthermore, TPL-RHK significantly inhibited the formation of lipid hydroperoxides and thiobarbituric acid-reactive substances (TBARS) from O/W emulsions with thermal acceleration at 50℃ during 12 days.
In conclusion, this study suggests that lauroyl tripeptides, produced via lipase-catalyzed acylation, have a potential as the multi-functional food additive to simultaneously control the lipid oxidation and foodborne pathogens in emulsion foods.
유화 식품(emulsion foods)의 지방산화와 식품 병원균에 의한 미생물 오염은 식품 품질 및 안전성을 저하시키는 주요 위해 요인이다. 이러한 위해 요인을 단일 화합물로 제어하기 위해, 항균 및 항산화 활성을 갖는 다기능성 식품첨가물의 사용이 제안되었다. 본 연구에서는 항산화 펩타이드(antioxidative peptide)와 로우르산(lauric acid)을 기질로 하는 라이페이스 촉매-아실화 반응(lipase-catalyzed acylation)을 통해 신규 다기능성 식품첨가물 소재를 개발하고자 하였다.
첫 번째로, 화학적 구조 및 항산화 특성을 고려하여 선정된 트라이펩타이드-KHA(tripeptide-KHA, TP-KHA)와 로우르산 간의 라이페이스 촉매-아실화 반응을 통해 다기능성 로우로일 트라이펩타이드-KHA(lauroyl tripeptide-KHA, TPL-KHA)를 제조하였다. 또한 아실화 반응조건의 최적화 및 기질 선택성 평가를 통해 로우로일 펩타이드의 효소적 합성 방법론을 확립하였다.
아실화 반응조건의 최적화 결과, 2-메틸-2-부탄올(2-methyl-2-butanol)의 유기용매, 55℃의 반응온도, 4.0의 기질비(로우르산:트라이펩타이드-KHA), 72시간의 반응시간이 최적 반응조건으로 설정되었으며, 본 조건에서 74.73%의 TPL-KHA 전환수율이 도출되었다. 라이페이스 촉매-아실화 반응의 기질 선택성을 확인하기 위해 LC-ESI-MS 및 1H-NMR을 통한 구조 분석을 수행하였다. 그 결과, TP-KHA는 1당량의 로우르산과 결합되는 것으로 확인되었고, 이 때 로우르산은 입실론(epsilon) 위치에 있는 아미노기에 선택적으로 아실화 되는 것으로 확인되었다.
TPL-KHA의 표면특성 평가 결과, TPL-KHA의 임계 미셀 농도(critical micelle concentration, CMC)는 2.71 mM, CMC에서의 표면장력(surface tension)은 30.44 mN/m로 공기-물 계면에서 표면장력 감소효과가 관찰되었다. 또한 TPL-KHA는 그람 양성(Staphylococcus aureus, Bacillus cereus) 및 그람 음성(Escherichia coli, Salmonella Typhimurium) 식품 유해균에 대한 정균 및 살균 효과를 나타내었다. TPL-KHA의 최소억제농도(minimum inhibitory concentration, MIC)는 2.83-4.00 mM 범위에서 도출되었으며, 최소살균농도(minimum bactericidal concentration, MBC)는 MIC의 약 1.3배 높은 농도에서 결정되었다. 더불어, TPL-KHA는 ABTS+ 라디칼에 대한 소거능을 나타내었으며, 100 μM의 TPL-KHA는 리놀레산의 열산화에 대해 80.99%의 항산화력을 나타내었다.
두 번째로, 로우르산 유도체의 항균력과 이화학적 특성 사이의 상관관계를 규명하기 위해 로우로일 트라이펩타이드(lauroyl tripeptide, TPL) 모델을 구축하였다. 동시에, 다기능성 식품첨가물 신규 소재를 탐색하기 위해 각기 다른 이화학적 특성을 갖는 항산화 트라이펩타이드를 선별하여 본 연구에 활용하였다. 리놀레산 열산화에 대해 알파-토코페롤과 유사한 항산화력을 나타낸 트라이펩타이드를 기질로 하여 각기 다른 이화학적 특성을 가진 5가지(TPL-Pro-His-Lys (TPL-PHK), TPL-Asn-His-Lys (TPL-NHK), TPL-Ala-His-Lys (TPL-AHK), TPL-Arg-His-Lys (TPL-RHK), TPL-Leu-Trp-Lys (TPL-LWK))의 TPL을 라이페이스-촉매 아실화 반응을 통해 제조하였다.
TPL들의 항균력 비교를 위해 그람 양성 2종(S. aureus, B. cereus) 및 그람 음성 2종(E. coli, S. Typhimurium) 식품 유해균에 대한 각각의 MIC와 MBC를 도출하였다. 그람 양성 및 음성 식품 유해균에 대해 2.50-5.17 mM 범위에서 MIC를 나타낸 TPL-AHK, TPL-PHK, TPL-NHK의 항균력 간에는 유의적 차이가 나타나지 않았으며, TPL-LWK는 항균력을 전혀 나타내지 않았다. 흥미롭게도 TPL-RHK의 MIC는 0.30-0.58 mM로 다른 TPL보다 약 10배 가량 강한 항균력을 나타내었다. TPL-LWK를 제외한 4가지 TPL의 MBC는 MIC보다 1.0-1.8 배 가량 높았으며, 이를 통해 TPL들은 그람 양성 및 음성 식품 유해균에 대해 정균 및 살균효과를 가진 것으로 판단하였다.
TPL의 이화학적 특성 중 순전하(net charge) 및 친수성(hydrophilicity)을 확인한 결과, 가장 강한 항균력을 나타낸 TPL-RHK는 +1.1의 양전하(positive net charge)를 가졌으며, 유사한 항균력을 나타낸 TPL-AHK, TPL-PHK 및 TPL-NHK는 +0.1의 동일한 양전하를 가진 반면, 항균력을 나타내지 않은 TPL-LWK는 양전하를 갖지 않는 것으로 확인되었다. 따라서, 양전하는 로우르산 유도체의 항균력을 결정하는 주요 인자로 판단되었다. TPL의 친수성과 항균력 사이의 특정한 상관관계는 파악되지 않았으나, 동일한 전하도를 가짐에도 불구하고 TPL-NHK가 전반적으로 TPL-PHK보다 더 강한 항균력을 갖는다는 결과에 근거하여 친수성이 증가함에 따라 TPL의 항균력이 향상 될 수 있음을 확인하였다. Propidium iodide (PI) uptake 시험 및 SYTO 9 및 PI의 이중염색 처리 후 형광 현미경 분석을 통해 TPL의 항균력은 세포막의 투과성(permeability) 변화에 기인하는 것으로 판단하였다. 따라서, 로우르산 유도체의 이화학적 특성은 세포막과의 상호작용에 대한 영향을 통해 항균력을 결정하는 것으로 사료된다.
마지막으로, 가장 강한 항균력을 나타낸 TPL-RHK의 다기능성 식품첨가물 소재로서의 활용 가능성을 확인하기 위해 유화, 항균, 항산화 특성을 수중유적형 유화계(oil-in-water emulsion system)에서 평가하였다. TPL-RHK의 유화 특성은 pH 3.0-7.0 범위에서 유화액의 시각적 관찰 및 제타 전위 측정을 통해 평가되었다. TPL-RHK 유화액은 pH 3.0 및 4.0에서 pH 5.0-7.0에서 보다 더 높은 안정성을 나타내었다. 제타 전위 측정을 통해 중성 pH 조건에서 TPL-RHK 유화액은 정전기적 반발력 감소로 인해 불안정한 것으로 판단되었다. TPL-RHK 유화액은 pH 3.0-7.0 범위에서 25℃에 저장 중 3일 이내에 크리밍(creaming) 또는 상 분리(phase separation) 되는 낮은 유화 안정성을 나타내었으며, 이는 TPL-RHK의 높은 친수성에 의한 것으로 해석된다. 대조적으로, TPL-RHK와 Tween 20 병용에 의해 제조된 유화액은 pH와 상관없이 9일의 저장기간 동안 Tween 20 유화액과 유사한 안정성을 나타내었으며, 두 에멀젼의 평균 직경(Z-average diameter) 및 다분산성(polydispersity index) 또한 차이가 없었다. 이에 따라, pH 3.0-7.0 범위에서 유화 특성에 영향을 미치지 않는 TPL-RHK는 수중유적형 유화 식품에서 식품첨가물 소재로 활용 가능함을 확인하였다.
수중유적형 유화계에서 TPL-RHK는 그람 양성(S. aureus, B. cereus, Listeria monocytogenes) 및 그람 음성(E. coli, S. Typhimurium, Pseudomonas aeruginosa) 식품 병원균에 대해 0.20-0.80 mM 범위에서 살균효과를 나타내었다. MBC 농도의 TPL-RHK를 S. aureus에 12 시간 처리한 결과, 3.68 log의 생균수 감소가 관찰되었고, 15 시간 처리 시 S. aureus가 완전히 사멸하는 것을 확인하였다. 또한 MBC 농도의 TPL-RHK를 E. coli에 18 및 24 시간 처리하였을 때, 각각 4.54 및 5.10 log의 생균수가 감소 됨을 확인하였다. 이에 따라 TPL-RHK는 유화 식품에서 식품 유해균에 의한 미생물 오염을 제어할 수 있음을 확인하였다.
TPL-RHK는 TP-RHK와 유사한 ABTS+ 라디칼 소거능을 나타내었으며, 이는 TP-RHK의 항산화력이 아실화 반응 후에도 유지됨을 의미한다. 반면, TPL-RHK 및 TP-RHK는 Fe3+에 대한 환원력을 나타내지 않았다. TPL-RHK는 50℃에서 12일 동안의 열 가속화(thermal acceleration)를 통해 유도된 수중유적형 유화액의 지방 열산화에 대해 항산화력을 나타내었다.
결론적으로, 본 연구에서 라이페이스-촉매 아실화 반응에 의해 제조된 TPL은 유화 식품의 지방산화 및 식품 유해균에 의한 미생물 오염을 동시에 제어할 수 있는 다기능성 식품첨가물 소재로 활용될 수 있을 것이라 사료된다.
