고혈압은 심혈관 질환, 뇌졸중 및 신장 질환 등을 초래하는 만성 질환이며, 조기사망 및 장애의 원인이 된다. 전세계적으로 성인의 약 30%가 고혈압을 갖고 있으며, 저 소득수준 및 중간 소득수준의 국가에서는 고혈압을 갖는 인구의 수가 계속 증가하고 있다. 따라서 고혈압의 치료는 뇌졸중, 장기손상 및 조기사망과 같은 질병을 예방하는데 매우 중요하다. 합성(화학)약물로 고혈압을 치료하는 것은 발기부전, 얼굴과 다리의 부종, 피부 발진 등의 부작용을 유발한다. 혈압을 조절하는 주요한 메커니즘 중 하나는 angiotensin-1 converting enzyme (ACE)에 의해 조절되는 Renin-Angiotensin-Aldosterone 시스템이다. 따라서 ACE는 고혈압을 줄이기 위한 중요한 마커로 사용된다. 최근에는 고혈압과 장내 미생물과의 강한 연관성이 제기되었다. 이 이론은 장내 미생물의 불균형은 병리학적 변화에 영향을 미치며, 식이 요법으로 쉽게 변화될 수 있다. 이러한 이유로 건강증진 및 질병치료를 위한 식품 개발에 있어, 장내 미생물을 dysbiosis에서 eubiosis로 바꿀 수 있는 식품은 중요하다. 하지만 음식으로부터 유래한 항고혈압성 펩타이드가 건강한 장내 미생물조건으로의 변화에 대한 연구는 아직 보고 된 바가 없다. 따라서 본 논문은 유청단백질과 분리 대두 단백질에서 항 고혈압 성 펩타이드를 개발하고, 작용 매커니즘을 연구하였으며, 동물 모델의 장내 미생물에 미치는 영향을 연구하였다.
먼저, 유청단백질에서 Lactic acid bacteria (LAB)의 ACE억제 펩타이드 생성능을 평가하였다. 유청단백질은 총 34개의 LAB와 37℃에서 48시간동안 발효되었다. 34개의 LAB중 7개가 강한 ACE억제 펩타이드 생성능을 보였으며, Pediococcus acidilactici SDL1414는 가장 높은 생성능을 보였다. 질량 분석을 통해 분석한 결과 P. acidilactici SDL1414로 발효된 시료에서 저 분자량 펩타이드(< 7KDa)의 약 60%가 ACE의 생성을 억제하는 펩타이드였다. 고혈압 환자 중 유당분해효소결핍증 있기 때문에 콩팥에 항 고혈압 펩타이드를 개발하기로 결정하였다.
유청단백질에서 ACE억제 펩타이드 생성능이 높은 7개의 LAB를 대두 단백질과 발효시켜 최적 LAB를 선별하였다. 그 결과, Pediococcus pentosaceus SDL1409가 가장 높은 ACE 억제효능을 보였다. 저 분자량 펩타이드는 liquid chromatography-electrospray ionization quantitative time-of-flight mass spectrometry를 통해 분석 되었으며, 가장 많고 일반적인 구조적 특징을 갖는 펩타이드 (EDEVSFSP, SRPFNL, RSPFNL, ENPFNL)가 선택되고 합성되었다. 이 펩타이드들은 0.1-0.5 mg/mL의 농도에서 강한 IC50 값을 나타냈으며, P/FNL, N-말단 서열 및 펩타이드 내의 아미노산의 위치는 ACE 억제에 필수적인 것으로 나타났다.
바이오컨버전스를 이용하여 펩타이드의 ACE 저해능을 향상 시켰으며 BPH/2J 마우스(고혈압 모델)에서 항 고혈압 효과를 평가하였다. 본 연구에서는 효소 가수분해와 LAB발효를 통해 시료를 생산하였다. 효소 처리 및 발효 후, in vitro ACE 저해 활성(P-SPI)은 60.8±2.0%에서 88.24±3.2 %로 증가하였다. 질량 분석실험을 통해 P-SPI에서 3가지 강력하고 풍부한 ACE 억제 펩타이드 PPNNNPASPSFSSSS, GPKALPII 및 IIRCTGC의 존재를 밝혀냈다. BPH/2J 마우스에 대한 P-SPI (10mg / kg 및100 mg / kg 체중)의 단일 경구투여는 대조군에 비해 수축기 및 이완기 혈압을 유의하게 낮추었다. P-SPI는 혈청 ACE의 활성을 억제하고 혈청 산화질소 수치를 높이며 산화 스트레스를 줄임으로써 고혈압을 감소시켰다. 또한 자연발생고혈압쥐(SHR)에서 P-SPI의 장기 섭취와 장내 미생물과의 관계를 확인하였다. P-SPI (10 mg/kg, 100 mg/kg)를 경구투여한 군에서 섭취 후 4주째에 수축기 혈압이 각각 10.8 mmHg 및 19.5 mmHg 감소되었으며, 체중 증가도 억제되었다. 분변 미생물을 통해 얻은 16S rRNA의 new generation 시퀀싱 분석은 SHR 장내 미생물의 풍부성과 다양성을 현저하게 감소시키는 것으로 나타났지만, 섭취후 6주째에는 P-SPI를 먹인 SHR은 장내 미생물의 풍부 성과 다양성을 향상 시켰습니다. P-SPI 급여는 SHR의 Firmicutes : Bacteroidetes 비율을 82:8에서 50:47으로 크게 감소시켰다. 프로피오네이트와 H2S를 생산하는 박테리아는 P-SPI 경구투여 후 Streptococcaceae와 Erysipelotrichales와 같은 잠재적인 병원성 박테리아의 개체군이 감소하는 동안 유의하게 증가하였다. SHR의 장내 미생물은 에너지대사에 관여하는 효소가 풍부하고 신진대사 속도가 빠르지만 P-SPI 처리 된 SHR에서는 급격히 감소했다. 또한 수용성 비타민생산은 P-SPI 급여 된 SHR의 장내 미생물에서 유의하게 증가 하였다. 우리의 결과는 P-SPI의 장기간섭취는 고혈압을 감소시키고 장내 미생물을 건강한 상태로 조절하는 것으로 나타났다.
Hypertension is a chronic disease that results in cardiovascular diseases, stroke and kidney diseases. It is one of the preventable risks of premature deaths and disabilities. Earlier studies have indicated that about 30% of adults globally are hypertensive and the number keeps increasing in low and middle income countries. Treatment of hypertension is therefore very important to prevent consequent disease conditions such as stroke, organ damage and premature death. Treatment of hypertension with synthetic (chemical) drugs has been associated with many undesirable side effects such as erection problems, lack of energy, swollen face and feet and skin rashes. One of the key mechanisms that regulate blood pressure is the Renin-Angiotensin-Aldosterone System mediated by angiotensin-1 converting enzyme (ACE). This enzyme has therefore become a good drug target for reducing high blood pressure. In recent years, a strong association between hypertension and changes in the gut microbiota has been suggested. It is believed that imbalance in the gut microbiota could have pathological effects on the host. Interestingly, the gut microbiota is plastic and can be modulated by diet. For this reason, designing foods that can shift the gut microbiota profile from dysbiosis towards eubiosis could be important for promoting health and treating diseases. To the best of our knowledge, very limited or no studies have investigated how antihypertensive peptides from food directly shift the gut microbiota of hypertensive patients towards healthy states. Therefore, the objectives of this thesis were to develop antihypertensive peptides in whey and isolated soy proteins, investigate their mechanism(s) of action and study their impact on the gut microbiota of animal models. Firstly, the ability of Lactic acid bacteria (LAB) to generate ACE inhibitory peptides from whey proteins was investigated. The protein was fermented with 34 LAB for 48 h at 37 ºC. Pediococcus acidilactici SDL1414 was the best of seven LABs whose fermentates showed strong ACE inhibitory abilities. Mass spectrometry revealed that about 60% of the low molecular weight peptides (˂ 7 KDa) in the P. acidilactici SDL1414 fermented samples were ACE inhibitory peptides. Since some hypertensive patients may be lactose intolerant, we decided to develop antihypertensive peptides in soybean, which would be acceptable for the general population. Therefore, we investigated the abilities of the best LAB strains obtained from the whey study to ferment isolated soy proteins to generate ACE inhibitory peptides. In this study, Pediococcus pentosaceus SDL1409 was found to yield the strongest ACE inhibitory fermentate. Therefore the strain was used to ferment soybean protein isolates for 48 h at 37 °C. The low molecular weight peptides were identified using liquid chromatography-electrospray ionization quantitative time-of-flight mass spectrometry analysis. On the basis of peptide abundance and common structural features, peptides EDEVSFSP, SRPFNL, RSPFNL and ENPFNL were selected and synthesized. The peptides showed strong IC50 values between 0.1 and 0.5 mg/mL. The presence of P/FNL, the N-terminal sequence as well as the position of amino acids in the peptides were found to be essential for ACE inhibition.
Also, the ACE inhibitory ability of the peptides was improved using bioconvergence technology and their antihypertensive effects were tested in BPH/2J mice (hypertensive model). In the study, we combined enzymatic hydrolysis and LAB fermentation to generate an antihypertensive product. After enzyme treatment and fermentation, the in vitro ACE inhibitory activity of the product (P-SPI) increased from 60.8 ± 2.0 % to 88.24 ± 3.2 % while Captopril (a positive control) had an inhibitory activity of 94.20 ± 5.4 %. Mass spectrometry revealed the presence of three potent and abundant ACE inhibitory peptides PPNNNPASPSFSSSS, GPKALPII and IIRCTGC in P-SPI. Single oral administration of P-SPI (10mg/kg and 100 mg/kg body weight) to BPH/2J mice significantly lowered systolic and diastolic blood pressure relative to control group. P-SPI reduced high blood pressure by inhibiting serum ACE activity, improving serum nitric oxide levels and reducing oxidative stress. Lastly, we ascertained the impact of long term (six weeks) consumption of the P-SPI on blood pressure reduction in spontaneous hypertensive rats (SHR) and the effects on the gut microbiota. Oral administration of 10 mg/kg and 100 mg/kg body weight of P-SPI significantly reduced systolic blood pressure by 10.8 mmHg and 19.5 mmHg respectively at the 4th week of feeding. P-SPI consumption also strongly suppressed weight gain in SHR. Next generation sequencing and analysis of fecal microbial 16S rRNA revealed significant reduction in microbial richness and diversity in SHR gut. However, SHR that were fed with P-SPI for 6 weeks had improved gut microbial richness and diversity. P-SPI feeding significantly reduced the Firmicutes: Bacteroidetes ratio of SHR from 82: 8 to 50: 47. The levels of propionate and H2S producing bacteria significantly increased while the populations of potential pathogenic bacteria such as Streptococcaceae and Erysipelotrichales decreased after P-SPI feeding. The gut microbiota of SHR were found to be enriched with enzymes involved in energy metabolism and their rate of metabolism were high but they were drastically reduced in P-SPI treated SHR. Meanwhile, water-soluble vitamin production was significantly increased in the gut microbiota in the P-SPI group relative to untreated SHRs. Our results show that, long-term consumption of P-SPI reduces high blood pressure and modulates the gut microbiota towards a healthy state.
