Kisspeptin에 의한 조골세포 분화의 분자적 조절 기전 = Molecular Mechanism of Osteoblast Differentiation by Kisspeptin

Kiss1 유전자에서 생성되는 단백질인 Kisspeptin은 세포막 수용체인 GPR54(G protein coupled receptor 54)를 통하여 인간 피부암(Melanoma) 및 유방암(Breast cancer)에서 종양 전이 억제(Tumor metastasis suppressor)를 하는 것으로 알려진 인자이며, 사춘기 발달(Pubertal development)과 성선자극호르몬성 성선부전증(Hypogonadotropic hypogonadism) 조절에 중요하다고 보고 되어있다. 뼈 형성(Bone formation)은 뼈를 생성하는 조골세포(Osteoblast)와 뼈를 흡수하는 파골세포(Osteoclast) 사이의 균형을 통해 분화 조절을 한다고 알려져 있다. 조골세포는 만능 중간엽 줄기세포(Mesenchymal Stem Cells; MSCs)로 부터 유래되었으며, 이 MSCs는 조골세포를 비롯하여 근세포(Myocytes), 지방세포(Adipocytes), 연골세포(Chondrocyte) 등으로 분화 할 수 있는 능력을 가진다. 현재까지 조골세포 분화(Osteoblast differentiation)에서 종양 전이 억제와 사춘기 조절 인자인 Kisspeptin에 대한 연구는 전무한 실정이다. 본 연구는 조골세포 분화에서 Kisspeptin의 역할과 그 분자적 기전을 알아보기 위한 실험을 진행하였다. 먼저 조골세포 분화 마커 유전자들이 Kisspeptin에 의하여 발현이 조절되는 것을 확인하였다. 그 결과, Dlx5(Distal-less homeobox 5), Runx2(Runt-related transcription factor 2), ALP(Alkaline phosphatase) 등의 유전자들이 Kisspeptin 처리에 의하여 mRNA 발현이 증가하였으며, 특이하게도 강력한 조골세포 분화 개시 유전자에 해당하는 BMP2(Bone morphology proteins 2) 유전자의 발현 또한 증가됨을 RT-PCR과 western blot 분석으로 확인하였다. 이 결과를 바탕으로 Kisspeptin에 의하여 증가된 BMP2 유전자 및 단백질이 분화 유도 기능을 가지고 있는지에 대한 연구를 수행하였다. Kisspeptin에 의한 BMP2 발현 증가가 BMP2 신호전달의 초기 단계인 Smad1/5/9의 인산화 수준을 증가시킴을 western blot 분석을 통하여 증명하였으며, 이 신호전달이 조골세포 분화 마커 유전자들의 mRNA 발현을 증가시킴을 증명하였다. 다음으로는 Kisspeptin의 분자적 기전을 확인하기 위하여 BMP2 Promoter가 cloning된 reporter vector(BMP2-luc)을 이용하였다. Kisspeptin은 C3H10T1/2 세포에서 BMP2-luc의 활성을 현저히 증가시킴을 확인하였다. 이전 보고에서 NFATs(Nuclear factors of activated T cell)가 직접적으로 BMP2 유전자의 전사활성을 증가시킨다고 보고되어 있었으며, NFATs는 최근 신장 세포에서 Kisspeptin/GPR54 신호전달에서 매개체로 작용하여 BMP7의 발현을 조절한다는 연구 결과가 보고되었다. 이런 사실을 바탕으로 NFATc4 유전자 과발현 벡터(over-expression vector)를 구축하여 세포 배양 실험한 결과, Kisspeptin 처리 없이 NFATc4 유전자 과발현 만으로도 BMP2-luc 활성이 현저하게 증가하는 것을 확인하였다. 이 결과는 Kisspeptin이 GPR54를 통하여 NFATc4를 자극함으로써 BMP2 유전자의 전사활성을 증가시키는 것으로 이해할 수 있다. 마지막으로 이상의 기전을 재확인하기 위하여 GPR54 Knock out 세포를 사용하여 Kisspeptin에 의한 세포내 신호전달 과정을 최종적으로 확인하였다. GPR54 Knock out 세포에서는 Kisspeptin 처리에도 불구하고 Smad1/5/9의 인산화가 증가되지 않았고, BMP2 및 분화 마커 유전자의 발현 변화가 없는 것을 확인하였다. 이상의 연구 결과를 요약하면, 사춘기 조절 인자의 하나인 Kisspeptin은 BMP2의 발현을 증가시킴으로써 조골세포 분화를 촉진하며 그 분자적 기전은 GPR54를 통하여 NFATc4 활성을 증가시켜 BMP2 유전자의 전사를 조절하는 것임을 증명한 것이다.
더보기Kisspeptin, which is producted from Kiss1 gene, has been reported to tumor metastasis suppressor in melanoma and breast cancer via its receptor, GPR54. The Kiss1/GPR54 signaling is playing an important role in the puberty development. Bone formation is a tightly regulated process of lineage-specific differentiation events, and bone homeostasis is maintained by a balance between bone resorption by osteoclast and bone formation by osteoblast. Osteoblast is derived from pluripotent mesenchymal stem cells with the capacity to also differentiate into myocytes, adipocytes and chondrocytes which are the necessary components to form bone matrix and allow subsequent mineralization. So far, the study about Osteoblast differentiation has never been reported, so this paper is specially about revealing the roles of Kisspeptin and its molecular biological effects. Osteoblast differentiation is controlled by a range of hormones and cytokines, such as bone morphogenetic proteins (BMPs), and multiple transcription factors, such as Runx2(Runt-related transcription factor 2), ALP(Alkaline phosphatase) and Dlx5(Distal-less homeobox 5). However, It is not known about their function in osteoblast differentiation. First is the effect of Kisspeptin on osteoblast differentiation. Treatment with Kisspeptin increased osteogenic gene expression in quantitative RT-PCR and Real-time PCR. RT-PCR showed that Kisspeptin significantly increased expression of BMP2 mRNA. Western blot analysis also showed that Kisspeptin increased the level of BMP2. Based on the result above, An experiment to check whether Kisseptin-induced BMP2 shows the functions of protein differentiation. Kisspeptin-induced BMP2 increased BMP2 signaling in an early stage phosphorylation, which is Smad1/5/9. The signaling was demonstrated that increasing the osteoblast differentiation marker genes mRNA expression. Finally, To confirmed molecular mechanisms of Kisspeptin on osteoblast, we used BMP2-luc which was cloned through BMP2 promoter. In the previous report NFATs (Nuclear factors f activated T cell) are reported directly increases the transcriptional activity of BMP2 gene. This NFATc4 mRNA levels was increased by Kisspeptin on osteoblast. Also, the cell culture experiment says that NFATc4 over-expression alone was able to confirm that BMP2 levels increase. By using GPR54 Knock out cells, we confirmed that Kisspeptin’ signaling through the receptor GPR54. This results that the wild type cells is the time-dependent manner BMP2 protein levels increased by the Kisspeptin treatment. However, GPR54 knock out cells were not changed by the treatment Kisspeptin. This study was Kisspeptin increased BMP2 and osteogenic marker genes mRNA level were analyzed RT-PCR and real-time PCR. These results are Kisspeptin induced osteoblast differentiation via NFATc4-mediated BMP2 signaling
