실뱀장어(Anguilla japonica)의 내인성, 외인성 산소소비 리듬에 대한 수온 및 염분 영향 = Effects of water temperature and salinity on the endogenous and exogenous rhythms of oxygen consumption in glass eels Anguilla japonica
실뱀장어(Anguilla japonica)의 수산·경제적 중요성 때문에 주 소상요인인 조석(또는 조류의 흐름), 수온, 염분 등이 소상량에 미치는 영향에 대해서는 많은 연구가 수행되었다. 그러나 대부분 하구에서 채집한 실뱀장어의 소상량과 소상요인과의 관계를 추정하였을 뿐, 생리 기작에 대한 연구는 미비한 편이다. 따라서 본 연구에서는 주 소상요인과 관련된 생리 기작을 규명하기 위하여 자동호흡측정기(automatic intermittent-flow respirometer: AIFR)를 이용한 산소소비율 분석을 통해 하구역에서 채집한 실뱀장어의 내인성 산소소비 리듬을 관찰하였다. 또한 실뱀장어의 내인성, 외인성 산소소비 리듬에 대한 수온 및 염분 영향을 관찰하였으며, 담수 노출에 따른 염분 적응기작에 관하여 논의하였다. 실뱀장어 시료는 한국 서해 연안의 강화도, 금강 하구와 남해의 제주도 천제연 하구에서 수집하였다.
실뱀장어의 내인성 생체리듬은 일정한 수온과 어두운 상태에서 먹이공급 없이 관찰하였다. 이러한 조건에서 실뱀장어의 산소소비 패턴은 강한 내인성 산소소비 리듬(12.1~12.7시간)이 나타났으며, 산소소비율의 정점(peak)은 채집지역의 고조(high tide)와 거의 동일한 시기에 관찰되었는데, 이는 실뱀장어의 호흡 패턴이 채집지역의 조석주기에 의한 12.4시간의 내인성 생체시계(endogenous biological clock)에 의해 조절되는 것으로 판단된다.
채집된 하구역과 같은 염분조건에서 수온상승에 따른 실뱀장어의 산소소비 반응을 관찰한 결과, 초기 색소발달단계인 VA~VIA1 단계 실뱀장어의 산소소비율은 13℃ 이하 수온에서 뚜렷한 변화 없이 낮은 값을 유지하였으며, 13~21℃ 수온구간에서는 계속 증가하였고, 21℃ 보다 높은 수온에는 다시 감소하였다. 그러나 후기 색소발달단계인 VIA1~VIA3 단계 실뱀장어의 산소소비율은 수온을 상승시킴에 따라 계속 증가하였다. 이러한 결과들은 실뱀장어의 수온에 대한 반응이 성장단계에 따라 다르다는 것을 의미한다. 또한 13℃ 이하 수온구간에서 실뱀장어의 산소소비율은 낮은 값이 측정되었는데, 이는 실뱀장어의 담수 소상과 관련된 수온인 것으로 추정된다.
임의로 선정한 14시간 주기로 2, 1, 0.7, 0.5, 0.4℃씩 수온을 상승 또는 하강시키면서 관찰한 실뱀장어의 산소소비 패턴은 수온변화 주기와 유사한 호흡리듬을 나타났다. 이러한 결과는 실뱀장어의 생리리듬이 내인성 조석주기 리듬뿐만 아니라 수온변화와 같은 외인성 리듬에 의해서도 조절된다는 것을 의미한다. 특히, 0.4℃의 미세한 수온변화에도 호흡반응이 관찰된다는 것은 본 종이 미세한 환경변화를 감지할 수 있는 능력이 있다는 것을 의미하며, 이러한 능력은 하구역이나 담수의 불안정한 환경을 피해 성장에 알맞은 서식지를 선정하는 과정에서 중요한 역할을 담당할 것으로 추정된다.
일정수온에서 염분을 실뱀장어를 채집한 하구역의 염분에서 저 염분(15.0, 10.0, 5.0 psu)에 노출시켰을 때 실뱀장어의 호흡 패턴은 저 염분에 노출된 후에도 뚜렷한 차이가 없었다. 그러나 직접 담수에 노출된 실뱀장어의 산소소비율은 노출 전보다 6.3~22.2% 감소하였으며, 산소소비 패턴은 27~55시간 동안 교란되었다. 교란과정을 거친 후 실뱀장어는 정상적인 호흡 패턴을 회복하였다. 이러한 결과들에 의하면 실뱀장어는 해수에서 직접 담수로 소상할 경우, 약 2일 가량의 심한 염분 스트레스를 받는 다는 것을 알 수 있다.
채집지역의 주, 야 수온 차이를 고려하여 25시간 주기로 2℃씩 수온을 변동시킨 상태에서 직접 담수에 노출된 실뱀장어의 산소소비율은 노출 전과 뚜렷한 차이가 없었다. 또한 실뱀장어의 호흡 패턴은 수온변화 주기와 유사한 산소소비 리듬(24.9~25.8시간)이 관찰되었는데, 이는 실뱀장어의 경우 담수 소상 시기 동안 염분변화에 따른 스트레스보다 수온변화에 의한 생리 스트레스를 더 강하게 받는다는 것을 제시하고 있다. 이러한 결과는 보(dam)로 차단된 하구환경에서 실뱀장어가 염분적응 과정을 거치지 않고 해수에서 수로(channel)나 어도(fish ladder)를 통하여 직접 민물로 소상할 수 있는 생리 적응 기작을 제시하는 중요한 결과이다.
Numerous studies have explored the effects of tide (or tidal current), temperature, and salinity as major factors in the upstream migration of glass eels, owing to their economic importance in aquaculture. These studies have only reported the relationships between catch data and environmental factors.
However, the physiological mechanisms of upstream migration have rarely been studied. This study investigated the endogenous rhythm of glass eels collected at the estuary to examine a physiological mechanisms affecting their upstream migration by analyzing of the oxygen consumption rate (OCR) using an automatic intermittent flow respriometer (AIFR). This study focused on the effects of water temperature and salinity on the endogenous and exogenous rhythms of oxygen consumption in glass eels, Anguilla japonica and salinity adaptation mechanisms after freshwater exposure. Glass eels were collected from the Kanghwa, Keum River estuary on the western Korean coast and from Cheonjeyeon estuary at the southern tip of Korea.
The OCRs of glass eels were measured in the laboratory without food under constant temperature and darkness. Under these conditions, the glass eels' oxygen consumption patterns exhibited a strong endogenous circatidal rhythm (12.1 - 12.7 h), and peaks of OCR were observed concurrent with times of high tide at the collection sites. These results suggest that the oxygen consumption rhythms of glass eels are influenced by an endogenous biological clock that is synchronized to the local tidal cycle (12.4 h).
The oxygen consumption responses of glass eels differed across pigmentation stages. In the early pigmentation stage (VA - VIA1 stages), the glass eels maintained a low OCR value under 13℃, which gradually increased as temperatures rose from 13 to 21℃, but then decreased. However, the OCR in the late pigmentation stage (VIA1 - VIA3 stages) continued to increase during experiments. These results indicate that the response to temperature in glass eels changes according to the pigmentation stage. The glass eels maintained a relatively low OCR until the temperature reached 13℃. This value may be interpreted as a main temperature related to freshwater migration of the glass eels.
When the glass eels were exposed to a cyclic increase or decrease in water temperature (2, 1, 0.7, 0.5, 0.4℃ per 14 h), OCR peaks exhibited a clear rhythmicity similar to cycles of temperature change. These results indicate that the oxygen consumption of glass eels is controlled not only by an endogenous circatidal rhythm but also by exogenous rhythms related to small environmental changes, such as temperature shifts as small as 0.4℃. The ability of glass eels to respond precisely to environmental changes in estuary may help them find a suitable habitat for growth in continental waters.
When the salinity was reduced from that of seawater to a low salinity concentration (15.0, 10.0, and 5.0 psu) under a constant temperature, the oxygen consumption patterns of glass eels were similar to those observed before the lower salinity exposure. However, after direct freshwater exposure, the OCRs of glass eels decreased by 6.3 - 22.2%, and the oxygen consumption patterns were disturbed during about 27 - 55 h. After these periods, the glass eels resumed normal oxygen consumption pattern. These results suggest that direct migration from seawater to freshwater results in a severe physiological stress for about 2 days.
When the temperature periodically changed by 2℃ per 25 h and glass eels were directly exposed to freshwater, the OCRs of the glass eels did not differ from the values observed before and after freshwater exposure. In addition, the oxygen consumption patterns (24.9 - 25.8 h) corresponded to the cyclic change in water temperature after freshwater exposure. These results suggest that the stronger stress associated with temperature dominates the physiological response and masks the response to the less significant stress caused by the salinity change. The results of the present study indicate that there is a physiological mechanism that allows glass eels to migrate directly into the freshwater through the fish ladder or channel at estuaries blocked by dams.
