Development of Wearable Glucose Monitoring Sensor for the Healthcare of Diabetic Patients

Objective: In this paper we report on a novel method for enhancing repeatability and stability of continuous glucose sensor. Background: The most widely used blood glucose monitoring systems are disposable devices based on blood collection by pricking fingers. Diabetic patients are simply able to check blood glucose level with the finger prick tests, but this gives the value of glucose level only at the particular time when the test is done. Therefore, it is difficult that the patients completely check changes of glucose level during the day or night when the blood sugars may go very high or low. Additionally, finger prick tests can induce significant pain and infection problems. Method: The electrode surface is initialized by sequential negative and positive pulse to reduce the factors influencing stability, such as non-faradaic current by residual products. Secondly for evaluation of the continuous glucose sensor, the micro-needle is connected to a reusable transmitter that continuously sends the measured glucose values to a mobile phone via Bluetooth. The insights and knowledge from this study provide a solution to recover the sensor to the same initial state before beginning each trial, which allow the sensor to have reliable results of in-vivo testing. Results: The results shown that the proposed continuous glucose sensor had excellent linear response at glucose concentration between 0 and 400 ㎎/dL with sensitivity of 12.1 nA/㎎·dL-1 (CV = 1.7%) and coefficient of determination, R² = 0.99. In addition, the sequential pulse allowed the sensor to have high repeatability of CV = 1.1% (n=10) and long-term stability of CV = 5.56% for 72 hours. Conclusion: For in-vivo long-term testing, the COC-PPy/GOx-pTTCA/Nafion based micro-needle was combined with the transmitter and penetrated the human skin. Comparing with conventional glucose sensor, it was confirmed that the proposed continuous glucose sensor could measure the glucose in the ISF and maintain its sensitivity under the aggressive biological environment. Application: The proposed continuous glucose sensor does not require a complicated and sophisticated process. Despite the simple structure and short fabrication process, the sensor could have enhanced repeatability and stability. The COC-PPy/GOx-pTTCA/Nafion based continuous glucose sensor has the great potential for evolving into future CGMS for proactive managing diabetes.
