Bisphenol-A(BPA) is classified as an endocrine disruptors that interferes with the humans endocrine system and produces adverse developmental, reproductive, neurological, and immunological effects in both humans and wildlife. To study the reproductive and toxicological effects of BPA in aquatic ecosystem, we employed Daphnia magna (D. magna) as a test animal model. D. magna is one of the most widely used water species employed in ecotoxicology testing all over the world. As a result, reproduction, based on the number of offspring produced, was determined to be significantly reduced in 10 mg/L of BPA compared with the control for 21 days. Gene expression profiles using DNA microarray analysis was also carried out in response to BPA exposure. Our data showed that numerous putative genes which have homologous sequences in other animal models were altered including cuticular protein, vitellogenin, protease, and the ribosomal protein family. These genes, which are critically reported in other animal systems, could be used as novel biomarkers of BPA exposure using DNA microarray analysis of D. magna.
더보기Nanoparticles are generally defined as particles less than 100nm. Among them, silver nanoparticle (AgNP) has been widely utilized for the medical approach due to anti-bacterial properties. However, it has been shown to have toxicological effects such as lethality and reproductive activity, on many species, including a great many fish species and, drosophila. It also has been reported that AgNP induce oxidative stress. However, the molecular mechanism of AgNP toxicity regarding oxidative stress has not been fully elucidated yet. In this study, we employed Daphnia magna and Cyprinus carpio as bio-indicator organisms to compare the molecular gene expression level of AgNPs exposure. Our microarray data showed that numerous putative genes including hemoglobin, heme oxygenase and alpha-globin were significantly altered in response to AgNPs exposure in both D. magna and C. carpio. The genes discovered from microarray can be valuable for risk assessment of AgNP in aquatic system and probably might be applied to comparative toxicogenomic database.
더보기Nickel and chromium, classified as heavy metals, can come into contact with the human population through air, food, and water sources, by which they may produce abnormal changes in organs, including kidneys, liver, cardiovascular, immune, and reproductive systems. Recognition of heavy metal biomarkers for toxicity assessments is of prime importance. Caenorhabditis elegans (C. elegans) is a well-known animal model for genetic studies, and serves as a living biosensor in ecotoxicological researches. Our study demonstrates the changes of genomic expression in C. elegans in response to heavy metal using nematode-specific microarray. We identified a number of genes as heavy metal responsive genes which were differentially expressed (>2 fold) following 24 h exposure to each heavy metal. The analysis of gene ontology revealed molecular components involved in several biological processes in C. elegans exposed to heavy metals. In addition, using the comparative toxicogenomics database, we have deduced the heavy metal-affected molecular mechanisms and symptoms of exposure. In conclusion, our investigation would be able to presents a precise route for understanding important roles of several heavy metal responsive genes in heavy metal toxicity. Furthermore, our study also identifies significant biomarkers in response to heavy metals. This might be of virtue for monitoring and evaluating hazard risk in the field of ecotoxicology.
더보기급격한 산업 발달로 인해 다양한 종류의 환경 호르몬과 같은 내분비계 장애물질이 생성되었고, 이들은 수질, 토양뿐만 아니라 음식물섭취 등 여러 경로를 통해 인체로 유입될 가능성이 점차 높아지고 있는 실정이다. 내분비계 장애물질로 분류된 Bisphenol-A(BPA)는 발달과정 및 생식능, 신경계, 면역계 등에 부작용을 유발하는 것으로 알려지고 있다. BPA의 생식능 및 독성영향을 평가하기 위해, 물벼룩 종인 Daphnia magna 종을 시험동물모델로 선정하였다. 물벼룩(D. magna)은 환경독극물에 대한 감수성이 높아 OECD guideline의 공시 생물종으로 추천되고 있으며, 주로 수질오염 평가를 위해 많이 이용되고 있다. OECD guideline에서 제시하는 시험법에 따라 3주간의 실험에서 생식능 측정 결과 10mg/L의 BPA농도에서 Offspring 생산수가 대조군 대비 현저하게 줄어든 것으로 나타났다. 또한 BPA에 노출된 물벼룩에서의 유전자 발현패턴을 분석하기 위하여 DNA 마이크로어레이 시스템을 도입하여 분석하였다. 그 결과 다른종과의 sequence 유사성을 가진 cuticle 단백질, vitellogenin, protease, ribosomal protein 등의 유전자가 발현변화를 보였다. 이들의 유전자는 BPA독성평가에 있어서 바이오 마커로서 활용될 수 있을 것으로 사료된다.
더보기최근 나노기술이 발전 및 대두됨에 따라 많은 산업에 응용되고 있으며, 특히 의학분야에서는 신약개발을 위한 분자적 도구로서 이용되고 있다. 그 중 은나노 물질은 세탁기, 화장품 등 소비자가 쉽게 접하는 곳에서도 이용되어 인체에 대한 노출량이 많은 것으로 알려져 있다. 은나노 사용량이 늘어나면서 잠재적인 위험성이 커지는 것으로 예상되는 가운데, 독성중심의 연구도 활발히 진행이 되고 있다. 기존연구에 의하면 은나노는 fish, drosophila등에서 치사 및 생식능에 독성영향을 미치는 것으로 알려져 있으나, 구체적인 분자적 기전은 아직 연구된 바가 없는 실정이다. 은나노 독성연구를 위해 대체 동물로서 D. magna 및 C. carpio를 대체동물로서 이용하였고, RNA level에서의 유전자 발현변화 확인을 위해 DNA 마이크로어레이 실험을 수행하였다. 그 결과, 산화적 스트레스와 연관된 Heme oxygenase, Hemoglobin 등의 유전자들이 발현변화를 보였다. 이러한 유전자들은 은나노 독성의 산화적 스트레스 관점에서 중요한 바이오 마커로서 활용될 수 있을 것으로 사료된다.
더보기급격한 산업의 발달로 인한 환경오염의 심화로 인해 환경오염에 대한 관심이 커지게 되었다. 중금속인 니켈, 크롬 등의 물질의 사용량이 증가하면서 이에 대한 독성연구는 필요하다고 볼 수 있다. 본 시험에서는 대표적 중금속인 니켈, 크롬에 노출된 Caenorhabditis elegans내에서의 유전자 발현 패턴을 DNA 칩 마이크로어레이 시험을 통해 분석하였다 (>2-fold, p-value<0.05). 본 시험에서 중금속에 의해 증가된 유전자는 glutathione S-tansferase, superoxide dismutase, choline kinase 등 산화적 스트레스 관련 혹은 발암관련 유전자들이 발현변화를 보였다. 이러한 유전자들은 각각의 중금속류에 대한 바이오 마커로서 이용될 수 있을 것으로 사료된다. 결론적으로 본 시험에서 얻어진 독성자료는 중금속에 의한 생체 내 반응 기작 해석에 기초를 제공할 것이며, 그 외에도 선충을 사용함으로써 수질에 비해 상대적으로 취약한 토양 및 발암성에 대한 모니터링 기법으로서의 가능성을 기대하고, 아직까지 선충을 대상으로 시험한 보고가 없기 때문에 더더욱 의미가 있다.
