With the progress of farming, orcharding and gardening, the consumption of agricultural pesticides has been remarkably increased, and its intoxication has been widely publicized. Among many pesticides, organophosphorus chemicals have been believed to be almost nontoxic because of their character of non-accumulativeness in tissue or organs and they have been used most frequently. However, some studies have reported quite many cases of farmers, orchardists and horticulurists who have been suffered from the adverse effect of organophosphorus pesticides. Thus, it has been gradually recognized that the intoxication from organophosphorus pesticides needs to be reviewed.
This study was in this relevance, attempted to examine the effects of
organophosphorus pesticides upon the blood and liver and four most commonly used
organophosphorus pesticides (phosphamidon, malathion, diazinon and monocrotophos) in Korea were selected for the study. For the experiment, the Sprague-Dawley rats weighing around 200 gm were divided into 1) basic diet group (control) and pesticide administered groups of 2) phosphamidon 3) malathion 4) diazinon and 5) monocrotophos. The basic diet composed of adequate solid neutrients was given to each rat orally 15-20 gm per day. A half and a quarter of LD?? of organophosphorus pesticides were daily dosed to each rat in the experimental group one time in the former and three times in the latter. The liver has been examined grossly, light and electron microscopically, and
serum cholinesterase, GOT, GPT, alkaline phosphatase, hemoglobin and WBC differential count have also been measured.
The results obtained from this experiment are summarized as follows:
1. Hemoglobin is decreased in 24, 72, 120 hr's groups of phosphamidon diazinon,
malathion and monocrotophos administered in order.
2. In white cell series, neutrophils are decreased but lymphocytes increased in groups of phosphamidon, diazinon, malathion and monocrotophos in order.
3. GOT is increased in groups of phosphamidon, malathion, diazinon and monocrotophos in order, especially in 24 hrs group.
4. GPT is slightly increased in groups of diazinon, phosphamidon, malathion and
monocrotophos in order.
5. Alkaline phosphatase is increased in groups of phosphamidon, malathion, diazinon and monocrotophos in order, especially in phosphamidon and malathion groups.
6. Cholinesterase is significantly suppressed in groups of phosphamidon, malathion, diazinon and monocrotophos in order.
7. Light microscopically, congestion, partial stenosis of sinusoids, swelling and vacuolar changes of hepatic cord cells are observed in all experimental groups. These changes are progressed and necrosis of hepatic cord cell is observed in order of phosphamidon, malathion and diazinon.
8. On electron microscopy, lipid droplets, diminution of ER, elongation of mitochondria in the hepatic cord cells observed in groups of phosphamidon, malathion and diazinon are more apparent than that in the group of monocrotophos. Additionally a shrunken or dissolved nucleus, altered electron dense mitochondria and electron lucent glycogen
particles are visible in groups of phosphamidon, malathion and diazinon.
