Atopic dermatitis (AD) is known to be the result of dry hypersensitive skin condition, which hinders the protective barrier function of the horney layer. The comprised skin barrier makes the skin readily develop inflammation in response to external stimuli including microorganisms. In terms of fungus, lipophilic yaests of the genes Malassezia has actively been studied. According to previous reports, the colonization level of Malassezia and the composition of Malassezia species are associated with the severity of AD. Most of the studies have been conducted based on traditional culture-dependent methods because genomic methodologies available for fungus have been limited compared with tools for bacteria. The efficiency of culture-dependent methods is, however, prone to vary with the isolation techniques and the culturing conditions. Here, we sequenced skin fungal communities, so-called `mycobiome` of 10 patients with AD and compare with those of 10 healthy controls. Skin swab samples from antecubital fossae of all subjects were obtained. The used phylogenic marker was internal transcribed spacer 2 (ITS2) regions of DNA. Taxonomic identification at various levels, from phylum to species, with alpha diversity analysis revealed more diverse fungal genera in AD samples. Malassezia was the most dominant genus among 680 genera identified in both subject groups. Species-level analysis identified nine different Malassezia species and revealed Malassezia globosa to be the most abundant species except in two AD patients and one healthy control, where Malassezia restricta was dominant. Although interpersonal variation was observed, the diversity assessed by the Shannon diversity index (SDI) indicated that AD samples had more diverse fungal communities than healthy controls at both genus and species level. Although this study was limited by its small number of cases and lack of precise clinical correlation, our data provide useful insights for further investigation.
