일반적으로 유기물질에 할로젠 원소가 치환되어있는 물질은 독성이 있는 것으로 알려져 있고 물질의 독성을 제거하기 위한 다양한 할로젠이탈 방법들이 제시되고 있다. 할로젠이탈 반응을 위해 hydride를 포함하고 있는 시약이 사용되는데 독성이 있는 tin hydride의 사용이 가장 잘 알려져있고 silane이나 alcohol을 hydride source로 사용한 방법들도 제시되었다. 지금까지 aryl halide의 할로젠이탈 반응을 통한 환원반응은 많이 개발되었지만 alkyl halide의 환원반응은 예시가 적고, 특히 alkyl chloride의 경우 할로젠이탈 반응 연구가 거의 진행되지 않았다.
이번 연구에서는 구리 촉매 촉진 하에 hydride source로 값이 싸고 다루기 쉬운 diisobutylaluminum hydride (DIBAL-H)를 사용한 alkyl chloride의 환원성 할로젠이탈 반응을 개발하였다. 온화한 조건 하에서 5 mol %의 N-헤테로사이클릭카벤 (NHC)를 기반으로 한 구리 촉매 촉진 하에 1차와 2차 alkyl chloride가 환원된 생성물을 합성할 수 있었다.
아이소시아네이트를 사용한 아마이드 합성 방법 개발
아마이드는 생리 활성 분자나 기능성 물질의 기본 골격을 이루고 있을 뿐 아니라 다양한 유기 합성의 유용한 중간체로 사용될 수 있다. 따라서 아마이드를 합성하기 위한 다양한 방법이 개발되었다. 특히 유기금속시약과 아이소시아네이트를 사용한 반응이 활발하게 연구되었는데 그 중 organoaluminum은 다른 유기금속시약에 비해 aluminum 자체가 가지고 있는 반응성 때문에 다양하게 합성에 응용할 수 있다는 장점이 있다. 하지만 organoaluminum 시약과 아이소시아네이트를 사용해서 아마이드를 합성하는 방법은 많이 알려져 있지 않다.
본 연구에서는 알렌으로부터 구리 촉매 촉진 하에 DIBAL-H를 사용하여 organoaluminum 중간체를 합성한 후 아이소시아네이트를 사용하여 4차 탄소를 가지는 호모 알릴릭 아마이드를 합성하는 반응을 개발하였다. 또한, 알카인으로부터 trimethylaluminum과 촉매량의 triethylamine을 사용하여 alumination을 통해 얻은 alkynylaluminum을 아이소시아네이트와 반응시켜 알카인 아마이드를 합성하는 방법을 개발하였다.
Copper-Catalyzed Reductive Dehalogenation of Alkyl Chlorides Using DIBAL-H
In general, halogen substituted compounds are known to be toxic. Therefore, various dehalogenation reactions have been developed to eliminate the toxicity of halogen-containing compounds. The most well-known method is the reductive dehalogenation of aryl or alkyl chlorides using reducing reagents such as toxic tin hydride. Methods using silanes or alcohols as hydride sources in the presence of transition metal catalysts have also been presented. To date, compared to reductive dehalogenation of aryl halides, a few examples of reduction reactions with alkyl halides have been studied.
In this study, we developed a copper-catalyzed reductive dehalogenation reaction of alkyl chlorides using inexpensive and readily avaliable diisobutylaluminum hydride (DIBAL-H) as a hydride source. A broad range of primary and secondary alkyl chlorides was successfully dehalogenated in the presence of 5 mol % N-heterocyclic carbene (NHC) based copper catalyst under mild reaction conditions.
Development of Methods for Synthesis of Amides Using Isocyanates
Amides are important building blocks for preparing potential drugs and functional materials as well as useful intermediates in synthetic organic chemistry. Due to their significance and synthetic utility, various methods for the construction of amides have been developed. One attractive approach to amides is amidation using a variety of organometallic reagents and isocyanates. Organoaluminums possess strong dual characters, Lewis acidity due to Al, and nucleophilicity because of the carbon atom bound to Al. This intrinsic reactivity affords various synthetic advantages to organoaluminums over other organometallic reagents, leading to their wide application in organic synthesis. However, amidation with organoaluminum reagents for the construction of amides has been rarely studied.
In this study, we described a new method for the synthesis of amides through a copper-catalyzed hydroalumination of allenes with DIBAL-H, followed by nucleophilic addition of in situ generated allylic aluminum reagent with isocyanates. A variety of homoallylic amides with quaternary carbon centers were synthesized with high efficiency. In addition, straightforward amidation from terminal alkynes through the in situ generation of aluminum acetylides obtained from Et3N-catalyzed alumination of terminal alkynes with trimethylaluminum and their successive nucleophilic addition to isocyanates was developed.
