Macrophages play an important role in defending against foreign pathogens, healing wounds, and regulating the inflammatory response, depending on their polarity. Modulating macrophage polarity is a key challenge to treat various diseases, such as chronic/acute inflammation and cancers. The macrophage polarity may be modulated by delivering nucleotide into macrophages, but the transfection efficiency of macrophages still remains low. Nanoparticles have been widely used in biomedicine, diagnosis, biological imaging, drug and nucleotide carriers. Quantum dots (QDs), among nanoparticles, have luminescent properties, which are useful for intracellular tracking and/or biomedical imaging. This study was designed to investigate whether QD complex can be used as a safe and efficient nucleotide carrier to macrophages that are resistant to transfection, using a murine macrophage cell line, RAW 264.7 cells. RAW 264.7 cells were treated with QDs with amine (Amine-QDs), carboxyl (Carboxyl-QDs), or polyethylene glycol (PEG) (PEG-QDs) at various concentrations (5, 10, 20, and 40 pmol). Carboxyl- and PEG-QDs were efficiently internalized into the cells from 79.09 to 89.67 % and from 71.69 to 91.45 %, respectively, whereas Amine-QDs were poorly delivered (less than 20 %). Both Carboxyl- and Amine-QDs showed cell toxicity, starting at 10 pmol, that made cell viability below 46.7 and 39.3 % at 40 pmol, respectively, but PEG-QDs did not cause any cytotoxic effects. Interestingly, PEG-QDs significantly increased the cell viability from 132.4 % at 5 pmol to 227.8 % at 20 pmol. Carboxyl-QDs were modified with polyethylenimine (PEI) (QD/PEI), and further conjugated with either plasmid DNA (pDNA) or small RNAs, small interfering RNA (siRNA) and microRNA, to produce QD complex. The size of Carboxyl-QDs, QD/PEI, pDNA-QD complex, and small RNAs-QD complex was 15.1±0.4, 79.0±0.2, 152.6±0.4, and 115.5±0.1 nm, respectively. Their surface charges were -29.53, 28.62, 20.63, and 21.45 mV, respectively. While the expression efficiency of the GFP pDNA-QD complex was not efficient compared to a commercial reagent, the GAPDH silencing efficiency of the small RNAs-QD complex showed significantly higher efficiency. Whereas Carboxyl-QDs decreased cell viability in a dose-dependent manner, pDNA-QD complex and small RNAs-QD complex did not cause any cytotoxic effects even at 120 pmol. Internalization mechanisms of Carboxyl-, PEG-QDs, and small RNAs-QD complex were evaluated by using endocytosis inhibitors. While Carboxyl- and PEG-QDs were mainly internalized via Clathrin-mediated endocytosis, small RNAs-QD complex utilized not only Clathrin- but also Caveolin-mediated endocytosis, possibly due to the larger size of QD complex. Immunofluorescence staining for early (EEA1) and late (LAMP-1) endosome markers showed that internalized small RNAs-QD complex were found in early endosomes as early as within 5 min and escaped at 30 min, and found in late endosomes (LAMP-1) at 2 and 4 h. The signals of FAM-labeled siRNA or microRNA free from QDs were escaped from late endosomes and released into the cytoplasm at 4 h. QD complex conjugated with miR-10a (QD complex/miR-10a) reduced the expression of Lipopolysaccharides (LPS)-induced M1 markers. Furthermore, QD complex/miR-10a treatment without LPS increased expression levels of M2 markers. QD complex/miR-10a inhibited the phosphorylation of NF-B, MAPK, and AKT signaling pathways. Collectively, QDs could be used as efficient and less toxic nanocarriers with dual modes for delivery and tracking of small RNAs to target macrophages for modulating their polarization. QD complex/miR-10a could inhibit M1 and promote M2 polarization to resolve inflammation diseases.
