Bone is a dynamic tissue that maintains bone mass throughout life through a process of bone remodeling by an active balance between osteoblast-mediated bone formation and osteoclast-mediated bone resorption. Bone remodeling becomes increasingly imbalanced by stress factors including aging, hormones, and dietary habits. The continuation of imbalance causes bone diseases such as osteoporosis, a fatal geriatric and metabolic disease in an aging society. Current osteoporosis treatment methods are limited to the use of anti-resorptive agents such as bisphosphonate, and are not suitable for long-term treatment due to fatal side effects. Therefore, it is important to discover natural materials that have fewer side effects and can be used for a long time. This study conducted a study on osteoblastic differentiation and bone formation using eucalyptol, a natural organic compound extracted from plants. Eucalyptol is a main component of natural essential oils extracted from aromatic plants such as Eucalyptus globules and is a monoterpenoid organic compound. Eucalyptol has therapeutic effects such as antibacterial, anti-inflammatory, and antioxidant in chronic diseases such as inflammatory disorders, respiratory diseases, and diabetic diseases. However, its function on osteoblast differentiation and bone diseases such as osteoporosis has not been studied. Since oxidative stress and inflammation act as major causes of bone disease, we hypothesized that eucalyptol would play an advantage in bone metabolism, and aimed to investigate the new function of eucalyptol for osteoblastic differentiation and bone formation. In this study, the efficacy of eucalyptol on osteoblast differentiation was confirmed in MC3T3-E1, an osteoblastic progenitor cell, and primary mouse calvarial cells. As a result, eucalyptol significantly increased the mRNA and protein expression levels of osteoblast differentiation marker genes. Also, it was confirmed that eucalyptol significantly promotes alkaline phosphatase (ALP) activity and mineralization. Next, the correlation between eucalyptol and ERK signaling in osteoblasts was confirmed. As a result, the osteoblast differentiation effect of eucalyptol was reversed by treatment with PD98059, an ERK inhibitor, and it was shown that the osteoblast differentiation effect of eucalyptol was mediated by ERK phosphorylation. We investigated the in vivo effect of eucalyptol using an animal model of surgical bone loss induction to confirm that the results at the cellular level also appear at the animal level. First, as a result of examining the efficacy of eucalyptol in zebrafish, it was confirmed that the amputated caudal fin bone rays of zebrafish were regenerated by eucalyptol in a dose-dependent manner. In the case of ovariectomized mice, eucalyptol (10 mg/kg, 50 mg/kg) was orally administered to mice induced osteoporosis by ovariectomy for 8 weeks, and the effectiveness was confirmed through micro-CT imaging. As a result, it was confirmed that bone loss of the distal femur was improved in mice in the eucalyptol 50 mg/kg administration group. Histological and immunohistochemical evaluations of distal femoral tissue sections from mice were confirmed by H&E and IHC staining. As a result, the ovariectomized mouse group administered with eucalyptol had fewer adipocytes and more bone marrow and the phosphorylation of ERK increased compared to the untreated mouse group. Taken together, these findings indicate that eucalyptol ameliorates bone loss by promoting osteoblast differentiation through ERK phosphorylation. We present the availability of eucalyptol as a natural organic compound that may be useful in preventing bone loss.
더보기뼈는 조골세포(Osteoblast) 매개 골 형성과 파골세포(Osteoclast) 매개 골 흡수 사이의 활성 균형에 의한 골 재형성(Bone remodeling) 과정을 통해 일생동안 골량을 유지하는 역동적인 조직이다. 골의 재형성 과정은 노화, 호르몬, 식습관 등을 포함한 스트레스 인자에 의해 점차 불균형해진다. 불균형의 지속은 골다공증(Osteoporosis)과 같은 골 질환을 유발하며, 고령화 사회에서는 치명적인 노인성 및 대사성 질환으로 자리 잡고 있다. 현재 골다공증 치료방법은 Bisphosphonate와 같은 항 흡수제(Anti- resorptive agent) 사용으로 국한되어 있으며, 치명적인 부작용 때문에 장기간 치료용으로 적합하지 않다. 따라서 부작용이 적고 오래 사용 가능한 천연물 소재를 발굴하는 것이 중요하다. 본 연구는 식물에서 추출한 천연 유기화합물인 eucalyptol을 사용하여 조골세포 분화 및 골 형성 관련 연구를 진행하였다. Eucalyptol은 Eucalyptus globules와 같은 방향성 식물에서 추출한 천연 에센셜오일의 주성분으로 monoterpenoid 계열의 유기화합물이다. Eucalyptol는 염증 장애, 호흡기 질환, 당뇨성 질환 등 만성 질환에서 항균, 항염증, 항산화 등의 치료 효과가 있다. 그러나 조골세포 분화 및 골다공증과 같은 뼈 질환에 대한 기능은 연구되지 않았다. 골 질환에서 산화 스트레스 및 염증은 주요 병인으로 작용하기 때문에 eucalyptol이 골 대사에서 유리한 역할을 할 것이라 가설을 설정하였으며, 조골세포 분화와 골 형성에 대한 eucalyptol의 새로운 기능을 조사하는 것을 목표로 하였다. 본 연구에서 조골세포 분화에 대한 eucalyptol의 효능은 조골전구세포인 MC3T3-E1과 primary mouse calvarial 세포에서 확인하였다. 그 결과, eucalyptol이 조골세포 분화 표지 유전자의 mRNA 및 단백질 발현 수준을 유의하게 증가시켰다. 또한, eucalyptol이 alkaline phosphatase(ALP) 활성 및 무기질화(Mineralization)를 유의하게 촉진하는 것을 확인하였다. 다음으로 조골세포에서 eucalyptol과 ERK 신호전달의 상관관계를 확인하였다. 그 결과, eucalyptol의 조골세포 분화 효과는 ERK의 억제제(inhibitor)인 PD98059 처리로 역전되었으며, eucalyptol의 조골세포 분화 효과가 ERK 인산화에 의해 매개됨을 보여주었다. 우리는 세포 수준의 결과가 동물 수준에서도 적용되는지 확인하기 위해 외과적 골 소실 유발 동물모델을 사용하여 eucalyptol의 생체 내 효과를 조사하였다. 먼저, zebrafish에서 eucalyptol의 효능을 조사한 결과, zebrafish의 절단된 꼬리지느러미 뼈 광선이 eucalyptol에 의해 용량 의존적으로 재생된 것을 확인하였다. 난소절제 마우스의 경우, 난소적출로 골다공증을 유도한 마우스에 eucalyptol(10 mg/kg, 50 mg/kg)을 8주간 경구 투여하였으며 micro-CT 촬영을 통해 유효성을 확인하였다. 그 결과, eucalyptol 50 mg/kg 투여군 마우스의 원위 대퇴골 뼈 손실이 개선되는 것을 확인하였다. 마우스 원위 대퇴골의 조직학적 및 면역조직화학적 평가는 hematoxylin and eosin(H&E) 및 immunohistochemistry(IHC) 염색을 통해 확인하였다. 그 결과, eucalyptol을 투여한 난소절제 마우스 그룹이 미투여 마우스 그룹과 대비하여 대퇴골 조직의 지방세포가 적고 골수가 많았으며, ERK의 인산화 발현이 증가하였다. 이상의 연구 결과를 종합하면, eucalyptol이 ERK의 인산화를 통해 조골세포 분화를 촉진하여 골 손실을 개선함을 나타낸다. 우리는 뼈 손실을 예방하는데 유용할 수 있는 천연 유기 화합물로서 eucalyptol의 이용가능성을 제시한다.
