Therapeutic activities and mechanisms of action of compound K and hydrazinobenzoylcurcumin in glioblastoma

Glioblastoma is one of the most aggressive and deadliest cancers and has poor prognosis due to resistance and resistance to conventional therapy.
Recently, stem cells of glioblastoma have been implicated as the main cause of resistance to recurrence and treatment of Glioblastoma. Therefore, glioblastoma stem cell (GSCs) target therapy has attracted attention as a powerful strategy for the treatment of brain tumors. In the first chapter of this study, describes that the activity of compound K, a metabolite of ginsenoside for human glioblastoma cells, U87MG and U373MG and GSCs derived from U87MG and U373MG. In both human glioblastoma cells, U87MG and U373MG, CK inhibited not only cell growth and clonogenicity through cell cycle G0 / G1 arrest but also metastasis ability such as migration and invasion. CK also induced nuclear condensation, cleavage, ROS, and mitochondrial membrane potential, thereby increasing apoptosis cell death. In addition, we demonstrated that CK induced anti-proliferative activity by inducing phosphorylation of PI3K, Akt and mTOR, and beside induced the activation of cleaved PARP, caspase-3 and caspase-9, thereby inducing apoptosis cell death. Furthermore, CK has effect on cancer stem cell-like phenotypes in GSCs derived from U87MG and U373MG. CK significantly inhibited not only the self-renewal capacity, but also the metastatic potential of glioblastoma stem-like cells. Notably, CK inhibited the expression of key stemness markers for GSCs, such as CD133, Nanog, Oct4 and Sox2, which contribute to self-renewal, multilineage capabilities and heterogeneity in GSCs. In the present study, our findings suggest that CK may be a new indicator into the molecular mechanisms involved in the anticancer effect on both cancer cells and cancer stem cells in GBM.
As an independent study, Chapter II describes the evaluation of anticancer activity of curcumin-derivative HBCs in glioblastoma stem-like cells. In this study, we demonstrated that HBC known as Ca2 + / CaM antagonists have effect on cancer stem like-cell phenotypes in human glioblastoma stem cells, U87MG and U373MG. Calcium Calmodulin (CaM) is a major cellular Calcium binding protein, implicated in a variety of cellular functions, including cell proliferation, growth. We observed the effect of HBC on the activity of CaMKII, which interacts with Ca2 + / CaM and causes enzyme activity. HBC reduced the phosphorylation of CaMKII and suppressed not only the self-renewal capacity, but also the metastatic potential in U87MG GSCs and U373MG GSCs. HBC also reduced the phosphorylation of c-Met and its downstream effectors such as STAT3, AKT, and ERK1/2, thereby suppressing the expression levels of glioblastoma stem cells markers such as CD133, Nanog, OCT4, and Sox2. We next examined the activity of si-CaMKII-gamma knockdown U87MG GSCs to investigate the relationship between CaMKII and c-met signaling. As a result, CaMKII-gamma depletion significantly inhibited the proliferation, neurosphere formation, invasion as well as the expression of c-Met signaling and stemness markers. These results suggest that GSC can be selectively targeted by inhibiting the activity of Calmodulin and CaMKII, suggesting that HBC will be a new indicator of GSC treatment through the mechanism of inhibiting the activity of Calmodulin and CaMKII.
