New Genetic and Biochemical Approaches on Mushroom Taxonomy in Genus Pleurotus

Fungal taxonomy has been developed based on morphological characteristics, which is laborious and time-consuming. Fungal taxonomy is associated with striking disadvantages of the lack of standardized and stable terminology as well as its high subjectivity. Typically, 18S and ITS rRNA analyses are the mainstream genetic methods. Nonetheless, such methods can not provide sufficient information for diagnosis of fungal species due to the few variable domains in 18S rRNA and the low PCR success rate of ITS sequences. To offset the above shortcomings, genetic and biochemical approaches have been proposed to replace the current identification methods and re-establish a new taxonomic system.
The genetic and biochemical characteristics were characterized by quantitative analyses, which can thereby enforce fungal taxonomy. Classic morphological approaches were carried out along with antagonistic and growth model analyses. Phylogenetic analysis was investigated based on ITS and LSU sequences. Meanwhile, DNA polymorphism was analyzed by inter-simple sequence repeat (ISSR) analysis, as well as DNA G+C content and DNA-DNA hybridization (DDH) tests. In addition, the biochemical characteristics were also explored, such as sugar, amino acid, polar lipid, fatty acid, quinone and mycolic acid.
In this study, all strains belonging to 43 different species were characterized and visualized through line drawing in accordance with their antagonistic phenomena. As could be observed, genus Pleurotus exhibited a linear growth rate through the four characteristic growth phases at about 1-5.2 mm/day. The similarities between Pleurotus species were over 99.0% in ITS analysis and 95.0-99.0% in LSU analysis. Both the phylogenetic trees had displayed a significant similarity in different species locations. Moreover, ISSR results had shown differences in the polymorphism among Pleurotus species. Genus Pleurotus was high DNA G+C content mushroom (50-70 mol%). The delimitation value of Pleurotus species was 80% in DDH test. In addition, the major biochemical compositions of Pleurotus species included glucose and arabinose for sugars; aspartic acid, arginine, lysine, tyrosine and alanine for amino acids; phosphatidyl-N-methylethanolamine, diphosphatidylglycerol, phosphatidylcholine and phosphatidylethanolamine for polar lipids; anteiso-C14:0, C16:0, C18:2 cis-9,12, C18:0, C18:3 cis-6,12,14 and summed feature 8 as major fatty acids and mycolic acid. MK-6 was sole quinone.
Results can be concluded in this study as follows: Firstly, morphological results that visualized by line drawing picture, was used to correct incorrect data and supply new morphological data for genus Pleurotus. Pleurotus species that linear growth model with four growth phases had notable antagonistic effect. Secondly, LSU analysis was more effective genetic approach than ITS analysis based on homological sequence similarity values. Phylogenetic trees displayed different genetic relationship between Pleurotus speies. Thirdly, ISSR analysis showed significant differences in polymorphic level of Pleurotus species, as well as measurements of DNA G+C content and DDH tests were also effective biomarkers for genus Pleurotus identification. Fourthly, Biochemical approaches could distinguish each Pleurotus species at species level based on various biochemical taxonomic characteristics. Finally, in conclusion, Pleurotus species were associated with the genetic and biochemical taxonomic characteristics. Fungal taxonomy was re-established through the genetic and biochemical approaches. These new taxonomic approaches provided a new taxonomic system, which can be used as new taxonomic principles of fungal taxonomy.
