Development of a random genomic DNA microarray for the detection of Listeria monocytogenes and Yersinia enterocolitica in food

A DNA microarray containing non-sequenced random genomic
DNA fragments of Listeria monocytogenes and Yersinia enterocolitica was
developed. As a first step, diagnostic abilities of this microarray were
verified using cells grown in a laboratory medium. To confirm the diagnostic
ability of the microarray, genomic DNAs extracted from other L.
monocytogenes or Y. enterocolitica strains, other species of genus Listeria or
Yersinia and other bacteria belonging to different genus were hybridized with
this microarray. Hierarchical clustering and principal components analysis
indicates that the microarray was able to detect L. monocytogenes or Y.
enterocolitica strains and clearly distinguished them from other speices in
Listeria or Yersinia genus and other bacteria in different genus. As a next
step, the diagnostic abilities of these microarrays in milk were determined.
When hybridized with genomic DNAs extracted from milk containing L.
monocytogenes or Y. enterocolitica strains without other microorganisms,
detection limits of this microarray were 6 log CFU/ml. However, when other
types of bacterial strains (B. cereus, S. Montevideo, Bacillus spp. [isolated
from milk], P. aeruginosa) were present in high number in milk, the
detection limits of the microarray were 8 and 7 log CFU/ml for L.
monocytogenes and Y. enterocolitica, respectively. Because of high detection
limits of this microarray, sample preparation protocols to increase the populations of L. monocytogenes and Y. enterocolitica in milk were
investigated. Enrichment conditions for rapid growth of L. monocytogenes or
Y. enterocolitica with other bacterial strains (B. cereus, S. Montevideo,
Bacillus spp. [isolated from milk], and P. aeruginosa) in various media
(TSBY, BHIB, and UVMB) and temperatures (25 and 37°C) were investigated. When 2.2 and 2.7 log CFU/ml of L. monocytogenes and Y.
enterocolitica, respectively, in milk were enriched in UVMB at 37°C for 24 h
and in TSBY at 25°C for 16 h, populations of L. monocytogenes and Y.
enterocolitica increased to 7.7 and 7.3 log CFU/ml, respectively. When the
microarray hybridized with the genomic DNAs extracted from these
enriched cultures, 100% positive hybridization signal were shown on the
microarray. Therefore it was concluded that the microarrays containing
random genomic DNA fragments can detect L. monocytogenes and Y.
enterocolitica in milk by a combination with culture-based methods.
유전자의 정보 없이 Listeria monocytogenes와 Yersinia enterocolitica의 지노믹 DNA 단편을 이용하여 유전자 칩을 개발하였다. 첫째 단계로, 실험 배지에서 배양한 균을 이용하여 칩의 진단성능을 확인하였다. 칩 제작에 사용하지 않은 다른 L. monocytogenes와 Y. enterocolitica, Listeria와 Yersinia 속 (genus)의 다른 종 (species), 그리고 Listeria와 Yersinia의 다른 속에 속하는 균주의 지노믹 DNA가 칩의 DNA 단편과 혼성화 되었다. 주성분 분석법과 크러스터링을 통한 통계분석결과 이 칩은 L. monocytogenes와 Y. enterocolitica만 검출하고 다른 종과 다른 속에 속하는 균주는 분명하게 구별하였다. 다음 단계로 이 칩이 우유에 오염된 균을 검출할 수 있는지를 확인해 보았다. 먼저, 우유에 L. monocytogenes와 Y. enterocolitica가 단독으로 오염되어 있을 때와 다른 미생물이 함께 오염되어 있을 때 이 칩의 검출한계를 조사해 보았다. L. monocytogenes와 Y. enterocolitica가 단독으로 오염되어 있을 때 이 칩의 검출한계는 6 log CFU/ml이었다. 하지만 우유에 다른 미생물 (Bacillus cereus, Salmonella Montevideo, Bacillus spp. [isolated from milk], Pseudomonas aeruginosa)과 함께 오염되어 있을 때 이 칩의 L. monocytogenes와 Y. enterocolitica의 검출한계는 각각 8 그리고 7 log CFU/ml 이었다. 이 칩의 높은 검출한계 때문에 균수를 증가시킬수 있는 샘플준비 절차로 증균배양방법이 선택되었다. 다양한 종류의 증균배양용 배지 (TSBY, BHIB, 그리고 UVMB)와 온도 (25와 37°C)에 따라 L. monocytogenes와 Y. enterocolitica가 다른 균주 (B. cereus, S. Montevideo, Bacillus spp. [isolated from milk], P. aeruginosa)와 같이 오염된 우유 속에서 개체수가 빠르게 증가되는 증균배양조건이 조사되었다. 2.2 log CFU/ml의 L. monocytogenes와, 2.7 log CFU/ml의 Y. enterocolitica는 UVMB배지와 37°C 에서 24시간 그리고 TSBY배지와 25°C에서 16시간 증균배양 했을 때, 각각 7.7과 7.3 log CFU/ml로 증가하였다. 이 증균 배양액에서 지노믹 DNA 를 추출하여 칩과 혼성화한 결과 L. monocytogenes와 Y. enterocolitica의 DNA 단편부분에서 각각 100% 양성시그널이 나타나는 것이 관찰되었다. 이 결과는 유전자 정보없이 L. monocytogenes와 Y. enterocolitica의 DNA 단편을 이용해서 제작된 이 칩이 일반배양법과 조합처리 되었을 때 다른균에 영향없이 우유에 오염된 L. monocytogenes와 Y. enterocolitica를 검출할수 있다는 것을 보여준다.
