PHOSPHOLIPASE C β4b ISOZYME REGULATES Gaq SUBUNITS AND βγCOMPLEX OF G PROTEIN-MEDIATED PHOSPHOINOSITIDE HYDROLYSIS

Phospholipase C β 4b isozyme has been characterized recently, showing that it is a cytosolic splice variant of ribonucleotides-inhibitable phospholipase C β 4a and the deduced amino acid sequence of them contains G₁, G₂, and G₃ motifs responsible for guanine nucleotides binding. In order to investigate molecular interaction of phospholipase C β 4b isozyme with heterotirmeric G protein subunits, high efficiency transient transfection was used to introduce cDNA corresponding to various Gαq subunits, βγ complex of G proteins, and phospholipase C β 4b isozyme into Cos-7 cells. Cells that were cotransfected with Gαq subfamily(Gαq, 11, 14, 16) of G protein or various combination of βγ complex(β₁γ₁, β₁γ₂, β₁γ₃, β₂γ₂) with phospholipase C β 4b isozyme and prelabelled with the [³H]myoinositol showed marked decreases in formation of [³H]inositol phosphates as compared with cells cotransfected with Gαq subunits of G protein or βγ complex, respectively, either after stimulation with aluminium fluoride or not. Cotransfection with cDNAs corresponding to the activated mutant of Gαq subunits and phospholipase C β 4b isozyme also results in similar decreases in the formation of [³H]inositol phosphates. The role of phospholipase C β 4b isozyme in signal transduction has not been elucidated yet. Here, we propose that phospholipase C β 4b may act as negative regulator of phospholipase C-mediated signal transduction system where Gαq subunits and βγ dimer stimulate phospholipase C β isozymes, respectively.
