NLRP3 inflammasome plays an important role in the initiation of inflammatory process through producing the active forms of the inflammatory cytokines including interleukin (IL)-1β. The mitotic Ser/Thr kinase NEK7 has been recently reported as one of pivotal players in the assembly and activation of NLRP3 in flammasome. NLRP3 inflammasome is well-known to contribute to a variety of inflammatory conditions of the respiratory system. However, little information exists so far regarding the role of NEK7-licensed NLRP3 inflammasome activation in the development of nasal polyps (NPs) in the sinonasal cavity. We aim to define the implication of NEK7 and NLRP3 inflammasome in NPs from patients suffering from chronic rhinosinusitis (CRS). NPs were obtained from CRS patients during the endoscopic sinus surgery and the inferior turbinate tissues were obtained from control subjects during the septoplasty or skull base surgery as healthy control. Results showed that protein expressions of NLRP3, caspase-1, and pro- and mature IL- 1β were remarkably increased in NPs compared to those in the control. In addition, expression of NEK7 protein was notably increased in NPs, which was well correlated with the more severe CRS verified by endoscopic, radiologic, and symptomatic scoring systems of CRS. We further subdivided the NPs into eosinophilic NPs (EPs) and non-EPs (NEPs) on the basis of blood eosinophil counts and, interestingly, we observed significant cytoplasmic co-localization and physical interaction between NEK7 and NLRP3 particularly in EPs compared to those in controls and NEPs. These findings suggest that NEK7-licensed assembly and activation of NLRP3 inflammasome may be crucially implicated in the development of NP, particularly EP. Therapeutic targeting the NEK7-NLRP3 interaction may have potential in CRS patients possessing EP that is frequently associated with the more severe clinical presentations and radiological features.
