인간유래 암세포주에서 아데노 부속 바이러스를 매개로한 전사 표적화 유전자 발현연구 = Transcriptional targeting of adeno-associated virus(AAV)-mediated gene expression in various human cancer cells
To establish gene therapeutic medicines for cancer treatments, a safe and tumor-targeting gene delivery vector with a cancer-specific strong expression needs to be developed. To develop such an ideal gene delivery vector for cancer gene therapy, several viral vectors including retrovirus, adenovirus, vaccinia virus, etc have been intensively studied. However, few studies using adeno-associated virus (AAV) vector have been done for cancers, so we investigated how efficiently a therapeutic gene can be delivered specifically to cancers via AAV-mediated delivery systems. As reported elsewhere, our previous results suggested that AAV serotype 2 (AAV2) with the highest transduction efficiency in human cancer cells can be further developed as an ideal cancer gene deliver vector. Here, we tried to develop a transcriptional targeting vector by testing several tumor-specific promoters in AAV2-mediated gene delivery systems. Based on bioinformatics, we selected putative cancer-specific promoters of protein regulator of cytokinesis 1 (PRC1), ribonuclease reductase 2 (RRM2) and Baculoviral IAP repeat-containing 5 (BIRC5) genes over-expressed in cancer cells. We first compared each promoter’s cancer specific activity in the breast or lung normal and cancer cell lines using a green fluorescent protein (GFP) gene as a reporter and confirmed that both promoters from PRC1 and RRM2 showed strong cancer-specific expression and their expressions were quite comparable to the CMV promoter in cancer cell lines. We then determined their in vivo promoter activities using a Xenogen IVIS 200 bioluminescent imaging (BLI) system after the nude mice implanted with HeLa were intravenously (IV) or intratumorally (IT) injected with rAAV2-CMV-Luc, rAAV2-PRC1-Luc, rAAV2-RRM2-Luc or rAAV2-BIRC5-Luc vectors. The gene expression in the IV- or IT-treated mice with rAAV2-PRC1-Luc was stronger and faster than that with rAAV2-CMV-Luc 48 hours post injection (day 2). Later, the gene expressions in the rAAV2-PRC1-Luc-injected mice became as high and persistent as those in the rAAV2-CMV-Luc-treated mice. Interestingly, the gene expressions in the rAAV2-PRC1-Luc-injected mice were mainly localized to tumor and liver as those in the rAAV2-CMV-Luc-treated mice were seen. Our present study demonstrated that the promoters of PRC1 and RRM2 showed in vitro cancer-specific strong expression, but the PRC1 promoter was also able to be active in liver of the treated nude mice.
