INTRODUCTION
One of complications which obesity can. cause is ovarian dysfunction with the resultant infertility. Wolguk Hwan(越鞠丸) has been commonly used to treat infertility due to obesity. This paper investigated the effect of Wolguk Hwan(越鞠丸) on the improvement of ovarian dysfunction and infertility in obesity-induced female mice. The indicators in this experiment were weight change, the rate of ovulation, in vitro fertilization and embryonic development of oocytes, the frequency of implantation and delivery, and levels of FSH, LH, E2 in serum.
METHOD & MATERIALS
The obesity group included sample 1 which was fed high density protein, high density lipid and chow in high density and weighed over 27g, and control 1 which was fed normal diet and chow in low density and weighed over 23g.
The normal group included, sample 2 which was fed high density protein, high density lipid and chow in high density and weighed less than 22, and control 2 which was fed normal diet and chow in low density and weighed less than 22g. All samples were administered with 0.3ml of Wolguk Hwan(越鞠丸) once a day for 4 days and all control groups were administered with 0.3m] of normal saline water in the same way as the
samples.
we used PMSG and HCG to cause multiovulation after administration of Wolguk Hwan(越鞠丸) and counted the ratio of ovulated oocytes and normal oocytes and examined samples' fertilization and embryonic development of oocytes.
Some of the mice were induced to fertilization and on the 1st day we examined the ratio of implantation with Chicago blue dye. Other mice were led to delivery and the ratio of the number of offspring and gender was observed.
RESULTS
Compared with normal group, the body weight was significantly (P<0.01) decreased in obesity group. Compared with control group, mean number of oocytes per mice ovulated was significantly (P<0.05) increased totally in the sample group of obesity and normal group. But Compared with control group, mean number of normal oocytes per mice ovulated was significantly (P<0.05) increased only in the sample group of obesity group. Compared with control group, the rate of in vitro embryonic development of oocytes was significantly (P<0.05) increased totally in the sample group of obesity and normal group. Implantation rate and delivery rate were significantly ineffective totally in obesity and normal group. The levels of serum FSH, LH, E2 in the mice were significantly ineffective totally in obesity and normal group.
CONCLUSION
According to these results, Wolguk Hwan(越鞠丸) is presumed to reduce body weight and improve ovarian functions by regulating both endocrine and metabolic activities.
