산화적 스트레스는 퇴행성 뇌 질환을 유발하는 주요 원인 중 하나로 여겨지고 있으며, 세포 내 활성산소종 (reactive oxygen species: ROS)을 생성하여 생체고분자인 단백질, 핵산, 지질을 파괴 또는 변형시켜 세포를 손상시킨다. 지속적으로 활성산소종에 노출이 되면 뇌 허혈을 포함한 다양한 인간 질환이 유발되기 때문에 활성산소종은 세포사멸에서 중요한 역할을 한다. 특히 뇌 질환 치료에서는 blood-brain barrier (BBB) 때문에 약물 전달의 큰 어려움을 가지고 있다. 이러한 약물전달의 문제점을 해결하기 위해 단백질 침투기술이 개발되었다. 단백질 침투
기술은 protein transduction domain(PTD)를 통해 세포 나 조직 내로 외인성 치료용 단백질을 전달하기 위해 일반적으로 사용되는 방법이다. PTD 융합단백질은 세포또는 뇌 조직으로 단백질을 전달할 수 있기 때문에 현재 다양한 질환에 응용하고 있는 상황이다. 본 연구에서는 3가지 치료용 타겟 단백질인 heat shock protein 22 (HSP22), phosphoprotein enriched in astrocytes 15 (PEA15), NAD(P)H: quinone oxidoreductase1 (NQO1) 단백질을 이용하였다. 그러나, 정확한 생물학적 기능과 허혈성 신경 손상에서의 이러한 단백질의 정확한 메커니즘은 제대로 이해되지 않았다. 따라서, 우리는 Tat-HSP22, PEP-1-PEA15, Tat-NQO1을 제작 및 정제하여 산화 스트레스로 유도 된 해마 신경(HT-22) 세포에서의 세포 사멸 및 뇌 허혈 동물
모델에서의 이 3가지 단백질의 효과를 연구하였다. Heat shock protein 22 (HSP22)은 모든 조직에서 발견되며 스트레스로 유도되는 단백질로 알려져있다. 이것은 small HSP family의 하나로 활성 산소종 및 세포 사멸 신호 경로의 조절을 통해 산화 스트레스에 대한 세포 손상에 대한 보호 효과를 가지고 있다. 우리는 HT-22세포에서 세포 내 ROS 생성 및 DNA 손상의 억제를 통해 산화 스트레스에 의한 침투성 Tat-HSP22의 보호 효과를 확인하였다. 또한 Tat-HSP22 단백질은 미토콘드리아 손상과 세포 사멸과 관련 된 단백질인 cytochrome c, Bax, Bcl-2, cleaved caspase-3의 발현 조절을 통해 세포 사멸에 대한 보호 효과를 확인하였다. 또한, 뇌 허혈 동물 모델에서도 침투성 Tat-HSP22 단백질은 해마 신경 세포 사멸과 관련 된 성상 세포와 미세아교 세포 그리고 cleaved caspase-3의 활성을 감소시킴으로써 뇌 허혈에 대한 보호 효과가 있음을 확인하였다.
Phosphoprotein enriched in astrocytes 15 (PEA15) 단백질은 신경계 특히 성상세포에서 풍부하게 발현되는 15kDa의 작은 인단백질이다. 이것은 다양한 세포 과정에서 세포 생존, 증식, 세포 사멸, 포도당 신진 대사에 관여하는 다기능 단백질이다. 또한, PEA 15 단백질은 in vitro와 in vivo에서 산화 스트레스로 유발된 도파민 성 신경 세포 사멸에 대한 보호에 중요한 역할을 한다고 알려져 있다. 세포 침투성 PEP-1-PEA15 단백질은 HT-22 세포내로 침투되어 산화적 스트레스로 유도한 세포내 활성산소종과 DNA 손상을 감소시켰으며, 활성화된 MAPK, Akt, 및 NF-kappaB (NF-kB)를 억제시켜 세포사멸을 억제시켰다. 또한 apoptosis 관련 단백질인 Bax, Bcl2, caspase-3의 발현을 조절하여 세포사멸에 대한 보호 효과가 있음을 확인하였다. 뇌 허혈 동물 모델에서도 PEP-1-PEA15 단백질은 뇌신경 세포사멸을 억제하여 뇌 허혈에 대한 보호 효과가 있음을 확인하였다.
NAD(P)H: quinone oxidoreductase1 (NQO1)은 항산화 기능을 포함하는 다기능 단백질이다. 이 단백질은 산화 스트레스에 대한 세포 생존에 중요한 역할을 한다. NQO1은 Keap1/Nrf2/ARE 기작의 조절에 의해 높게 유도되는 단백질로 알려져 있다. 또한 NQO1 단백질은 암, 당뇨등을 포함하여 세포 사멸뿐만 아니라 AD 및 PD를 비롯한 신경 퇴행성 질환의 억제에 중요한 역할을 한다. Tat-NQO1 단백질은 HT-22 세포내로 침투되어 산화적 스트레스로 유도한 세포 사멸, ROS 레벨과 DNA 손상에 대해 효과적으로 억제시켰다. 또한, 산화적 스트레스에 의한 세포 내에서 활성화된 MAPK와 Akt를 억제시켰으며, caspase-3 발현을 조절하여 세포사멸에 대한 보호 효과가 있음을 확인하였다. 또한, 뇌 허혈 동물 모델에서도 침투성 Tat-NQO1 단백질은 동물의 뇌의 해마 신경 세포 사멸과 관련 된 성
상 세포와 미세아교 세포 대한 보호 효과가 있음을 확인하였다.
본 연구에서는 세포 침투성 Tat-HSP22와 PEP-1-PEA15, Tat-NQO1 융합단백질이 효율적으로 세포 및 조직 내로 침투되어 산화적 스트레스로 유도한 HT-22 세포사멸과 뇌 허혈에 대한 뛰어난 보호 효과가 있음을 확인하였다. 이러한 결과를 토대로, 세포 침투성 Tat-HSP22와 PEP-1-PEA15, Tat-NQO1 융합단백질은 뇌 허혈을 포함한 산화적 스트레스와 관련된 다양한 질환에서 단백질 치료제로서의 가능성이 있음을 제시하고 있다.
Oxidative stress is a major risk factors in eurodegenerative diseases via the generation of intracellular reactive oxygen species (ROS) which lead to the destruction of macromolecules including proteins, DNA, and lipids eventually resulting in cell death. Oxidative stress also plays an important role in the pathological processes of various human diseases as well as ischemic injury.
Drug therapy for brain diseases is complicated by the difficulty of traversing the blood-brain barrier (BBB). Protein transduction technology is a commonly used method for delivering exogenous therapeutic target proteins into cells and tissues via protein transduction domains (PTDs). PTDs are well known for their ability to deliver proteins into cells and have been used in various clinical applications. In this study, we used three target proteins: heat shock protein 22 (HSP22), phosphoprotein enriched in astrocytes 15 (PEA15), and NAD(P)H: quinone oxidoreductase1 (NQO1).
These proteins are known to have antioxidant functions and protective effects against oxidative stress. However, the exact biological functions and precise mechanisms of these proteins in ischemic neuronal damage remains poorly understood. Therefore, we constructed and purified cell permeable Tat-HSP22, PEP-1-PEA15, and Tat-NQO1
proteins to examine the protective effects of these proteins against oxidative stress-induced hippocampal neuronal (HT-22) cell death and in animal models of ischemia.
HSP22 protein is known to be a stress-inducible protein found in all organisms. It is a member of the family of small HSPs and has protective effects against various oxidative stress-induced diseases via regulation of intracellular ROS levels and apoptosis signaling pathways. We showed thatTat-HSP22 protein transduced into HT-22 cells and significantly inhibited HT-22 cell death caused by oxidative stress via inhibition of intracellular ROS generation and DNA fragmentation. In addition, Tat-HSP22 protein protected against apoptosis through regulation of mitochondrial
dysfunction and apoptosis related protein expression including cytochrome c release, cleaved caspase-3, Bax, and Bcl-2. Further, Tat-HSP22 protein transduced into animal brain and markedly attenuated hippocampal neuronal death via inhibition of astrocyte, and microglial activation as well as activation of caspase-3 expression in an ischemic animal model. PEA15 is a small (15 kDa) phosphoprotein prominently expressed in the brain,
particularly in astrocytes. It is a multifunctional protein which is highly involved in various cellular processes related to cell survival, proliferation, apoptosis, and glucose
metabolism. In addition, PEA15 protein is known play a crucial role in protection against oxidative stress-induced dopaminergic neuronal cell death in vitro and in vivo. We demonstrated that PEP-1-PEA15 protein transduced into HT-22 cells and protected against oxidative stress-induced HT-22 cell death by inhibition of cellular toxicities including elevated ROS levels, DNA damage and activation of MAPK, Akt, and NF-kB. Transduced PEP-1-PEA15 protein protected against oxidative stress-induced HT-22 cell death via regulation of Bcl-2, Bax, and caspase-3 expression levels. In an ischemic animal model, PEP-1-PEA15 protein markedly inhibited hippocampal neuronal cell death via inhibition of astrocyte and microglial activation. NQO1 is a multifunctional protein the functions of which include antioxidant functions. It also plays an important role to cell survival against oxidative stress. NQO1 is known as a highly inducible protein regulated by the Keap1/Nrf2/ARE
pathways. In addition, NQO1 protein plays an important role to the inhibition of various forms of cell death including cancer, diabetes, as well as neurodegenerative disorders including AD and PD. We showed that Tat-NQO1 protein transduced into HT-22 cells and significantly promoted cell survival via inhibition of intracellular ROS production and levels of DNA damage induced by oxidative stress. Also,
Tat-NQO1 protein inhibited the activation of Akt, MAPK, and caspase-3 signaling pathways under oxidative stress conditions. In addition, Tat-NQO1 protein markedly
enhanced hippocampal neuronal cell survival via inhibition of astrocytes and microglial activation in an ischemic animal model. In conclusion, we have shown that constructed and purified Tat-HSP22, PEP-1-PEA15, and Tat-NQO1 proteins efficiently transduced into HT-22 cells and drastically protected against oxidative stress-induced HT-22 cell death. Furthermore these cell permeable proteins efficiently transduced into the hippocampus and
significantly increased neuronal cell survival in an ischemic animal model. Therefore, Tat-HSP22, PEP-1-PEA15, and Tat-NQO1 proteins may potentially provide a
therapeutic strategy for the treatment of various oxidative stress-related diseases including ischemia.
