Protein arrays and ultrastructural imaging with atomic force micrscopy (AFM) play a critical role for proteome research and understanding ultrastructures of various biomolecules. First, we presented fluorescence-based enzyme activity assay on well-type protein arrays. Transglutaminase (TG), a calcium-dependent enzyme that catalyzes transamidation reactions, is involved in several functions in cells such as apoptosis, cell adhesion, morphology change and inflammation. Thus, it is essential to determine in situ TG activity to investigate its functions in cells. In situ TG activity has been determined by conventional methods such as ELISA plate assay, confocal microscopy and Western blotting. In this study, we describe a novel chip-based approach to determine in situ TG activity in a high-throughput manner. The arrays were fabricated by immobilizing 3-aminopropyltrimethoxysilane of well-type arrays. The assay reproducibility was evaluated by analyzing the inter-array/-reaction reproducibility (a correlation coefficient of 0.99, 0.98 respectively). The new assay was successfully applied to the rapid analysis of in situ TG activity stimulated by maitotoxin. The activation of in situ TG activity by MTX was significantly inhibited by TG inhibitors, cystamine and monodensylcadaverine (MDC) in a dose-dependent manner.
Second, AFM has emerged as a tool for studying the architecture and dynamic properties of biomolecular structures under near-physiological conditions since AFM provided real height profile of samples with sub-nanometer resolution. In this study, we newly observed the ultra-thin fibers, which were not observed by optical microscopy, by ultrastructural imaging with AFM. The ultrastructural imaging of ultra-thin fiber revealed that height of ultra-thin fibers is 2-4 nm. In addition, we investigated to identify ultra-thin fibers and to reveal regulation mechanism for formation of ultra-thin fibers in cells. AFM imaging of ultra-thin fibers demonstrated that ultra-thin fibers distributed at adjacent region of neighboring cells. In addition, the generation of ultra-thin fibers was regulated by intracellular ROS.
Thus, the array-based in situ TG activity assay is a powerful system in the rapid and high-throughput analysis of transglutaminase functions in cells. Moreover, ultrastructural imaging with atomic force microscopy revealed extracellular ultra-thin fibers.
단백질 연구와 다양한 생체분자의 초 미세 구조를 이해하기 위해 protein array와 atomic force microscopy (AFM) 를 이용한 연구는 중요한 역할을 한다. 먼저, 단백질 칩을 이용하여 in situ transglutaminase (TG) activity를 분석하는 방법을 개발하기 위하여 본 실험에서는 well-type array를 만들어 연구를 수행하였다. 정제된 TG의 in vitro activity를 측정해 본 결과 TG의 농도별로 activity가 증가하였으며, array 간의 재현성은 0.998, 반응 간/ array 간의 재현성은 0.976으로 나타났다. in situ TG를 활성화 시키는 물질로 알려진 maitotoxin (MTX) 을 농도별로 처리하여 in situ TG activity가 증가하는 것을 확인하였다. 그리고, MTX에 의해 증가된 in situ TG activity는 TG의 inhibitor인 cystamine과 MDC의 농도에 따라 in situ TG activity가 감소하였다.
다음으로, 광학현미경으로 관찰하지 못했던 새로운 ultra-thin fiber를 AFM을 이용하여 발견하였다. AFM은 수 나노미터 수준의 해상도로 시료를 관찰할 수 있어, 생체분자 구조를 연구하는데 중요한 기술이다. 본 실험에서는 NIH 3T3 fibroblast cell에 PMA를 처리하였을 때 ultra-thin fiber가 형성되는 것을 확인하였다. Ultra-thin fiber의 굵기는 2-4 nm 정도 이며, 세포 내 활성산소에 의해 조절됨을 확인하였다.
그러므로 단백질 칩을 이용한 in situ TG activity 분석 방법 개발은 세포 내 TG를 high-throughput으로 분석 할 수 있으며, 게다가 AFM을 이용한 초미세구조 이미징으로 새로운 ultra-thin fiber를 발견 할 수 있었다.
