Blood brain barrier (BBB) plays major roles of preserving brain homeostasis and preventing toxic substances to enter the brain. In addition, tight junctions (TJ) between endothelial cells in the BBB play a role in selectively preventing the entry of most blood-borne substances into the brain. Development of treatment for degenerative brain diseases is a challenge as TJs in the BBB frequently prevent the entry of most medications into the brain. Furthermore, difficulties in performing in-vivo studies and high cost involved have decelerated the development of drugs. Therefore, developing in-vitro BBB models have gained significance. For these reasons, development of in-vitro BBB models has gained importance and recently-developed models include the Transwell system, DIV-BBB, and the latest, microfluidic BBB models. However, these have not satisfied requirements for an ideal in-vitro BBB model. We fabricated in-vitro microfluidic device to mimic the unique characteristics of BBB. The BBB chip is fabricated using micro electro mechanical systems (MEMS) technology. The BBB chip which includes a porous membrane is integrated with both Si-based multi-layered microfludic device and poly-dimethylsiloxane (PDMS) part. The microfluidic channel of silicon chip is composed porous membrane of polyimide. The porous membrane is transparent and arranged a pore size of 2 μm, a thickness of 3 μm. The top PDMS layer has a thickness of 3 mm and through holes, defining the inlets and outlets, were punched using a 1 mm punch. Fabricated silicon chip was deposited on front sides of the SiO2 3000 ?. Then top PDMS and silicon chip were assembled together using O2 plasma treatment and the BBB chip was autoclave treatment for three times. With this device, capillary endothelial cell (bEnd3, RBMEC) and primary astrocyte (primary rat astrocyte, primary mouse astrocyte) were co-cultured across the porous flexible membrane. Then the shear stress was applied on capillary endothelial cell by continuous flow of media through the microfluidic channel. Expression of the tight junction was acquired by immunostaining, permeability assay and transendothelial electrical resistance (TEER) measurement at the BBB chip to confirm the effects of co-culture and shear stress condition on capillary endothelial cell, respectively. FITC-Dextrans 4 kDa and 70 kDa fluorescent tracer diffusion was measured to evaluate barrier permeability in the microfluidic BBB device in the capillary endothelial cell mono-culture condition and the capillary endothelial cell-primary astrocyte co-culture condition. A comparison of the capillary endothelial cell mono-culture condition and the capillary endothelial cell-primary astrocyte co-culture condition showed that the permeability value (P value) for FITC-Dextrans 4 kDa in the co-culture decreased 18-32% more than that in the capillary endothelial cell mono-culture. In addition, a comparison of the capillary endothelial cell mono-culture condition and the capillary endothelial cell-primary astrocyte co-culture condition showed that the P value for FITC-Dextrans 70 kDa in the co-culture decreased 39-46% more than that in the capillary endothelial cell mono-culture. A comparison of the capillary endothelial cell mono-culture condition and the capillary endothelial cell mono-flow condition showed that the P value for FITC-Dextrans 4 kDa in the capillary endothelial cell mono-flow decreased 10-12% more than that in the capillary endothelial cell mono-culture. In addition, a comparison of the capillary endothelial cell mono-culture condition and the capillary endothelial cell mono-flow condition showed that the P value for FITC-Dextrans 70 kDa in the capillary endothelial cell mono-flow decreased 20-44% more than that in the capillary endothelial cell mono-culture. TEER was measured to evaluate barrier tightness in the BBB microfluidic device, under conditions that include capillary endothelial cell mono-culture as well as capillary endothelial cell-primary astrocyte co-culture and the capillary endothelial cell mono-flow condition. A comparison of the capillary endothelial cell mono-culture condition and the capillary endothelial cell-primary astrocyte co-culture condition showed that the TEER value in the capillary endothelial cell co-culture increased 20-30% more than that in the capillary endothelial cell mono-culture. In addition, a comparison of the capillary endothelial cell mono-culture condition and the capillary endothelial cell mono-flow condition showed that the TEER value in the capillary endothelial cell mono-flow increased 10-20% more than that in the capillary endothelial cell mono-culture. Therefore, the environment was considered to have improved barrier tightness between capillary endothelial cells. In-vitro flow BBB model system should be useful for studying the interactions between endothelial cell, astrocyte cell as well as the role of shear stress in the barrier property of endothelial cell. Also, in-vitro flow BBB model is likely to significantly contribute to future in-vitro studies of the BBB biochemistry, physiology, pharmacology and pathology. In this study, in-vitro BBB microfluidic device will be suited for study of tight junction barrier function in detail and evaluation of drug passage to gain further insight into the treatment of central nervous system diseases.
더보기뇌혈관장벽 (blood-brain barrier, BBB)는 확산장벽으로 중추신경계 (central nervous system, CNS)의 모세혈관에 있는 독특한 조직구조이며, 모세혈관내피세포 (capillary endothelial cell)과 성상세포 (astrocyte)와 혈관주위세포 (pericyte) 등의 신경-혈관단위체 (neurovascular unit)으로 구성되어있다. 뇌혈관장벽은 뇌로 유해한 물질이 유입되는 것을 차단하고, 뇌의 항상성을 유지하는 기능을 수행한다. 또한, 모세혈관내피세포 사이에 존재하는 밀착연접 (tight junction)이 뇌로 들어오는 대부분의 혈관유래물질을 선택적으로 차단하는 역할을 한다. 성상세포와 혈관주위세포 등의 신경-혈관단위체는 모세혈관내피세포와 상호작용을 통해 밀착연접을 향상시키는 것으로 알려져 있다. 그러나 뇌혈관장벽의 밀착연접이 퇴행성 뇌 질환을 치료하기 위한 대다수의 약물이 뇌 안으로 유입되는 것을 차단하기 때문에 치료약물의 개발에 어려움을 겪고 있다. 이러한 이유로 생체 밖 (in-vitro) 뇌혈관장벽 모델의 필요성이 중요하게 부각되고 있다. 현재까지 transwell system, DIV-BBB와 최근에 개발된 미세유체 뇌혈관장벽모델 등이 있지만 아직까지는 이상적인 생체 밖 뇌혈관장벽 모델의 요구사항을 충족시키지 못하였다. 본 연구에서는 마이크로 미세가공기술 (MEMS)을 이용하여 투명한 다공성 막이 소자에 일체화 되어있는 구조를 갖는 뇌혈관장벽 미세유체소자를 제작하였다. BBB chip은 다공성 막이 일체화 되어 있는 실리콘 기반의 미세유체소자와 poly-dimethylsiloxane (PDMS)로 구성되어 있다. 실리콘 칩의 다공성 막은 polyimide로 제작하였으며, 다공성 막의 홀 사이즈는 2 μm, 두께는 3 μm이다. PDMS 층의 두께는 3 mm이고, 채널과 만나는 양 쪽에는 주입구와 방출구가 뚫려 있다. 제작된 뇌혈관장벽 미세유체 소자는 기존의 다른 세체 밖 뇌혈관장벽 모델에 비해서 얇고 투명한 다공성 막을 지니며, 공배양과 유체환경 모사가 동시에 가능하다는 장점을 갖는다. 생체 안 환경과 유사하도록 뇌혈관장벽 미세유체소자의 다공성 막에 모세혈관내피세포와 1차 성상세포(primary astrocyte)를 공배양 한 후 혈액을 대체한 배지로 유체환경을 모사하였다. 이러한 소자에서 TEER 측정과 밀착연접단백질 (tight junction protein)의 염색, 투과성분석 (permeability assay)을 통하여 밀착연접 형성을 확인하였다. 뇌혈관장벽모사 미세유체소자에서 형광염색을 시행하여 밀착연접단백질 (Claudin-5, ZO-1)의 발현을 확인하였다. 또한 제작한 미세유체소자에서 투과성분석과 TEER 측정을 통하여 형성된 밀착연접을 비교하였다. 투과성 분석에서 투과성 값 (P value)을 비교해 본 결과 성상세포와의 공배양 환경과 유체환경에서 투과성 값이 모두 감소함을 확인하였다. 성상세포와의 공배양 환경에서는 FITC-Dextrans 4 kDa 투과성 값은 약 18-32% 감소하였고, FITC-Dextrans 70 kDa 투과성 값은 약 39-46% 감소하였다. 또한 유체환경에서는 FITC-Dextrans 4 kDa 투과성 값은 약 10-12% 감소하였고, FITC-Dextrans 70 kDa 투과성 값은 약 20-44% 감소하였다. 이것은 성상세포와의 공배양 환경과 유체환경이 모세혈관내피세포의 밀착연접을 향상시키는 것을 나타낸다. 또한, TEER 측정에서 TEER 값을 비교해 본 결과 성상세포와의 공배양 환경과 유체환경에서 TEER 값이 모두 증가함을 확인하였다. 공배양 환경과 유체환경에서 투과성 값이 모두 감소함을 확인하였다. 성상세포와의 공배양 환경에서 TEER 값은 약 20-30% 증가하였고, 유체환경에서 TEER 값은 약 10-20% 증가하였다. 이것은 성상세포와의 공배양 환경과 유체환경이 모세혈관내피세포의 밀착연접을 향상시키는 것을 나타내는 것을 확인하였다. 이러한 연구 결과를 바탕으로 제작된 뇌혈관장벽 미세유체소자는 향후 퇴행성 뇌 질환을 치료하기 위한 신약개발 및 뇌혈관장벽 연구에 유용하게 사용될 수 있다는 가능성을 제시하였다.
