小柴胡湯이 肥滿誘導白鼠의 肝 및 副睾丸脂肪組織과 血淸脂質의 變化에 미치는 影響 = Effects of Soshihotang(小柴胡湯) on the Histological Changes in Liver and Epididymal Fat and the Biochemical Changes of Lipid Metabolism in Obese Rats Induced by High Fat Diet
This experimental study was designed to investigate the effects of Soshihotang on the weight and lipid metabolism in obese rats induced by high fat diet. Total population of sixty male rats(Sprague-Dawley) were utilized for this investigation. All rats selected for this study were six weeks old with weight of 190±5g. The first group of twenty rats(Normal) were fed with normal diet(Samyang Co. Korea), second group of twenty rats(Control) were fed with high fat diet(Harlan. TD94095, USA), and the remaining twenty rats(Sample) fed with high fat diet and the Soshihotang. The measurement has been performed on (1) the weight of epididymal fat pad with electric weight gauge(Librorel-1200H, Shimadzu, Japan) (2) the average size and the number of epididymal fat cells with light microscope(Olympus, Japan) and personal computer(Image Analysis System. LECO 2001, Canada) (3) the area % of fat drops in hepatic lobule with Light microscope and Computer image analysis system (4) the quantity of total cholesterol in serum on enzyme method COD-POD system (5) the quantity of triglyceride in serum on enzyme method LPL-GK-GPO system (6) the quantity of free fatty acid in serum on enzyme method ACS-ACO system (7) the quantity of total lipid in serum on Sulfo-phosphovanillin method (8) the quantity or insulin in serum on Radioimmunoassay method.
The results are as follows :
1. It showed significant evidence that high fat diet conjunction with Soshihotang decreased the weight of rats compare to control group.
2. It showed significant evidence that high fat diet conjunction with Soshihotang decreased the epididymal fat pad weight, the average size of epididymal fat cells and the number of epididymal fat cells compare to control group.
3. It showed significant evidence that high fat diet conjunction with Soshihotang decreased the area % of rat drops in hepatic lobule compare to control group.
4. It showed significant evidence that high fat diet conjunction with Soshihotang decreased total cholesterol, triglyceride, free fatty acid and total lipid in serum compare to control group.
5. It showed significant evidence that high fat diet conjunction with Soshihotang decreased insulin level in serum to control group.
Based on these results, it is proved that Soshihotang decreased the weight of fat, the size of fat cells and the number of fat cells. So it is concluded that the clinical application of Soshihotang can help the treatment of obesity.
