밀원판별의 기준이 될 수 있는지를 조사하기 위하여 아카시아 꿀 7개, 잡화 꿀 9개, 밤 꿀 5개, 토종꿀 5개, 총 26개의 시료로 기존의 물리화학적 방법과 SDS-polyacrylamide gel electrophoresis pattern을 분석하였다. 꿀의 수분 함량은 밀원에 따른 차이를 나타내지 않았고 당조성분 중 sucrose는 pure honey와 adulterated honey를 구별하는 신뢰할 수 있는 기준이 될 수 없었으며 밀원별차이도 나타내지 않았으나 벌꿀의 숙성정도와 다른 당의 혼입여부를 판단하는 기준은 될 수 있었다. Fructose/glucose ratio도 현저한 유의적 차이를 나타내지 않았다. 총 산도는 밀원이나 진위여부의 판단 기준이기보다 비 이상적인 발효의 발생 여부를 판단하는 꿀의 품질기준으로서의 가치가 있었다. 설탕의 원료인 사탕무(C3식물)의 carbon isotope ratio는 C4식물(옥수수, 사탕수수)의 carbon isotope ratio보다 낮으며 벌꿀의 밀원이 되는 대부분의 꽃 꿀의 carbon isotope ratio와 유사한 값을 나타내므로 C3식물인 사탕무 당의 오입을 구별하지 못했다. 또한 stable carbon isotope ratio만으로는 밀원의 종류를 사탕무를 포함한 C3식물과 C4식물로는 구별 할 수 있을 뿐이었다.
HMF는 밀원에 일정한 기준 없이 다양하게 나타났으며 채밀 후 시간이 경과함에 따라 모든 꿀에서 점차적으로 증가하므로 특정 밀원을 구별하는 기준이 되지 못했다. 함유하고 있는 무기물의 조성과 함량은 꿀의 종류에 따라 현저하게 차이가 있었다.
아카시아 꿀 시료들의 Pb 함량은 0.5 mg/kg 이하였으며 밤 꿀 시료들의 Pb 함량은 1.25 mg/kg 이상으로 나타났다. 토종꿀 1은 10.64±0.02 mg/kg로 나타났으며 잡화 꿀 3, 5, 6은 평균 0.5 mg/kg 이상으로 나타났다. Pb의 함량이 높다는 것은 밀원 인 꽃이 생장하는 지역의 환경이 오염되었을 의미한다.
SDS-polyacrylamide gel electrophoresis pattern에 의한 꿀 단백질의 분자량 분석에서 양봉 꿀의 특이적인 59 kDa 단백질과 토종꿀의 특이적인 56 kDa단백질이 각 벌꿀 시료마다 약간의 차이는 있었으나 존재함을 알 수 있었다. 서양 꿀벌의 대표적인 소화액 효소류인 72 kDa-β-glucosidase의 단백질 band도 밤 꿀 3과 밤 꿀 5, 토종꿀 5에서 확인 하였다. 밤 꿀 1과 밤 꿀 2, 밤 꿀 3, 토종꿀 4, 토종꿀 5에서 밤 꿀의 특징적인 단백질 band가 존재하였으므로 꿀 속의 단백질에는 소화액 기원의 효소류 이외에 밀원식물 기원 단백질도 존재하는 것으로 나타났다.
This study was carried out to analyze the physicochemical properties and SDS-polyacrylamide gel electrophoresis patterns of twenty six kinds of honey products in order to identify factors potentially useful for distinguishing honey sources. The moisture content of all samples was not related to the source plant's floral type. Sucrose, a major ingredient of honey, did not present any reliable criterion that could be used to distinguish pure from adulterated honey, nor was it useful for identifying the sources of honey products. However, this study showed that sucrose content could be used to identify the mixing of honey with other sugar sources, as well as the degree of honey maturity. The fructose/glucose ratio did not differ significantly between samples. However, total acidity was useful for determining whether or not the honey was fermented abnormally, although it was not a useful criterion for identifying the source plant's floral type or the genuineness of the honey. This study did not address the adulteration of honey with sugar beet (a C3 plant) because the stable carbon isotope ratio of sugar beet - the main ingredient of sugar syrup - is lower than the carbon isotope ratio of C4 plants (including cane and corn sugar), and similar to nectar from source plants of honey. However, the stable carbon isotope ratio can be used to determine if the source plant for honey production is a C3 or C4 plant. After collecting a honey sample, the HMF level could not be used to discriminate among different source plants because it exhibited too much variation and increased gradually over time. The mineral content and chemical composition of honey samples did not show any differences associated with floral sources, however they did vary among samples. The Pb contents of acacia honeys and chestnut honeys were below 0.5 mg/kg and above 1.25 mg/kg, respectively. The Pb content of native bee honey sample 1 was 10.64±0.02 mg/kg and those of poly floral honey samples 3, 5, and 6 were above 0.5 mg/kg. These results indicate that the floral sources and associated soils were contaminated with Pb.
Based on SDS-PAGE analysis of honey samples, the molecular weights of the major proteins in native and foreign bee honeys were 56 kDa and 59 kDa, respectively. In the case of honey samples 3, 5, and native honey, β-glucosidase was a specific factor associated with western bee honey detected at 72 kDa. In addition, the range of 31.9-33.5 kDa bands could be used to identify honey as chestnut-specific, since chestnut honey samples 1, 2, 3 and native honey samples 4, and 5 exhibited characteristic protein bands of 32.1 kDa, 31.9 kDa, 33.5 kDa, 32.3 kDa, and 32.5 kDa. Therefore, it appears that SDS-PAGE analysis can reveal specific protein patterns associated with enzymes from the honey bees as well as botanical sources.
