Effect of Immunization against Periodontal Pathogen in Squirrel Monkey

Periodontal diseases are inflammatory responses thought to be triggered by specific microorganisms colonizing in the gingival crevice. Theoretically periodontal diseases could be prevented if the etiologic organisms were not allowed to colonize the subgingival area. The humoral immune response is one mechanism which may modulate bacterial colonization in the gingival crevice. Recent findings of our studies have proven the usefulness of squirrel monkey as an in vivo model for studying colonization by putative pathogens. The study was undertaken to determine if the squirrel monkkey (Saimiri scuireus) is indigenously colonized with black-pigmented Porphyromonas and Prevotella species (BP/P) resembling human P. gingivalis and P. intermedia. The presence of BP/P was generally associated with an increased age. increased gingival index(GI). the presence of calculus. and increased levels of serum antibody. These data indicated that the squirrel monkey may be a good model for studying the parameters of natural infection of the gingival crevice with suspected periodontopathogenic BP/P microorganisms. It was also found that immunization with whole cells of P gingivalis and P. intermedia were able to modulate the colonization of the ginigival crevice by these bacteria. Squirrel monkeys(Saimiri scuireus) were immunized with putative periodontal pathogen Porphyromonas gingivalis and Prevotella intermedia. Immunization resulted in an increased level of lgG anti-P. gingivalis and anti-P intermedia sera and was associated with a strong trend toward a statistically significant reduction in BP/P colonization of the gingival crevice. These findings strongly indicate that squirrel monkey is a valid animal model for studying human periodontal diseases, which in most cases are inflammatory responses involving initial colonization by specific bacteria and immune modulation by host. In this paper the findings from squirrel monkey studies involving 1) natural occurrence of BP/P and 2) immune modulation of BP/P colonization by immunization with BP/P are reviewed. Also their impact on the development of possible vaccine for periodontal diseases is described.
