그래핀 전극을 이용한 신경세포의 인공적 신경돌기 분화 및 소듐 이온 검출

전기 자극 요법은 신경계 질환을 치료하는 유망한 방법이다. 이 방법은 생체 전극(Bio-electrode)을 통한 전기 자극의 직접 또는 간접 전달에 의해 신경 세포의 활동 및 분화를 유도한다. 통상적으로, 전기 자극을 위해 금속 또는 실리콘 전극이 사용되어왔지만 세포 및 조직에 독성이 있어 사망에 이를 수 있다. 이러한 이유로, 전도성 중합체 및 탄소 전극을 사용하는 다양한 접근법이 생체 전극으로서 사용되는 것이 시도되어 왔다. 이들 전극 중 그래핀 전극은 우수한 전기적 특성과 생체 적합성으로 인해 신경 공학 및 조직 공학 연구에 널리 사용되고 있으며, 생체 전극으로서의 잠재력에 대한 많은 연구가 보고 된 바 있다.
본 연구에서는 화학기상증착(chemical Vapor Deposition, CVD) 방식으로 제작된 단층(single-layer)의 그래핀 시트(graphene sheet)를 생체 전극으로써 사용하여 다음과 같은 연구를 수행하였다.
(1) CVD 그래핀의 항균성 평가 및 불소 플라즈마 기능화를 이용한 항균성 향상 실험 그리고 그래핀 전극의 항균 메커니즘을 규명하였다. CVD 그래핀 전극의 생체 적합성에 관한 많은 연구가 보고되었으나, 항균력과 메커니즘은 아직까지 불분명하다. 생체 전극의 항균 특성은 미생물의 흡착 및 성장을 억제하여 전극이 삽입되는 부위의 감염을 사전에 예방할 수 있다. 낙하 시험(drop-test)을 사용하여 항균성을 평가 한 결과, CVD 그래핀의 항균성은 88.9%로 나타났으며, 불소화에 의해 항균성이 92.6%로 향상되었다.
(2) 전기 자극을 이용한 인위적인 신경 분화 유도는 그래핀 전극상에서 두 종류의 신경세포(신경아종양세포(neuroblastoma, SH-SY5Y), 쥐 해마 뉴런(rat hippocampal neuron))에 대해 평가하였다. 그래핀을 통한 전기 자극은 신경세포의 신경 분화를 성공적으로 유도하였다. 순수 그래핀(Pristine graphene, PG)과 산소 플라즈마 처리 그래핀(Oxygen plasma treated graphene, OG)에서 2가지 유형의 신경세포의 신경 분화는 전기 자극에 의해 상이한 경향을 나타냈다. SH-SY5Y 세포의 경우, 분화된 신경돌기의 최대 길이는 전기 자극을 인가한 순수 그래핀(electrical stimulation with PG, PGST)에서 세포 배양판(cell culture plate, CCP) 대비 147.6%로 가장 길게 분화하였으며, 평균 길이는 전기 자극을 인가한 산소 처리된 그래핀(electrical stimulation with OG, OGST)에서 CCP 대비 171.5%로 가장 길게 분화하였다. 쥐 해마 뉴런의 경우, 분화된 신경돌기의 최대 길이와 평균 길이는 PGST에서 CCP에 비해 각각 107.5%와 110.9%로 가장 길게 분화하였다. 또한, 세포 당 분화된 신경돌기의 수는 OGST 조건에서 CCP에 비해 281.9%로 증가하였다. 따라서, PG를 이용한 전기 자극은 신경돌기의 길이 분화를 위해서 사용될 수 있으며, OG를 이용한 전기 자극은 신경돌기의 네트워크 형성을 위한 활용이 예상된다.
(3) 전기 자극 용 그래핀 전극은 이온 선택성 전계 효과 트랜지스터(ion-selective field effect transistor, ISFET)로 동작 가능하다. ISFET 동작을 위해 기준전극(reference electrode)은 필수적이며 Ag/AgCl 기준전극이 일반적으로 사용된다. 그러나, Ag/AgCl 기준전극은 구조 및 크기로 인해 소형화에 한계가 있다. 따라서, 본 연구에서는 기존에 사용되는 Ag/AgCl 기준전극을 대체하기 위해 불소 기능화한 그래핀 기준전극을 개발하였다. 불소 기능화 한 그래핀 기준전극은 H+, Na+, K+ 및 Ca2+ 이온에 대하여 감도를 나타내지 않았다. 또한, 6시간 동안 측정된 장시간 사용 안정성 평가에서 측정된 전위 변화량(∆VGS)은 1.65 mV/h로 매우 낮았다. 그래핀-ISFET의 Na+ 이온에 대한 감도는 상용화 기준전극인 Ag/AgCl 기준전극으로 측정 시 43.5 mV/dec이었으며, 그래핀 기준전극으로 측정 시 감도는 59.2 mV/dec이었다.
Electrical stimulation therapy is a promising method for treating neurological diseases. This method induces the activity and differentiation of nerve cells by the direct or indirect transmission of an electrical stimulation through bio-electrodes. Conventionally, metal or silicon electrodes have been used for electrical stimulation, but these electrodes can be toxic to cells and tissue. For this reason, various approaches using conductive polymers and carbon electrodes have been attempted to be used as the bio-electrodes. On the other hands, graphene is widely used in neuroscience and tissue engineering research due to its excellent electrical properties and biocompatibility. Many studies have been reported on graphene as a bio-electrode.
In this study, a single-layer graphene sheet manufactured by chemical vapor deposition (CVD) was used as a bio-electrode for applying electrical stimulation. The following studies were conducted to utilize the CVD graphene sheet as a bioelectrode.
(1) The antibacterial activity of CVD graphene, the mechanism of antibacterial activity on graphene electrode, and the functionalization method to increase antibacterial activity were investigated. Although many studies have been reported on the biocompatibility of CVD graphene sheets, the antibacterial activity and antibacterial mechanism are still unclear. The antibacterial activities of the bio-electrode inhibit the adsorption and growth of microorganisms, thereby preventing infection of the area where the electrode is inserted. As the result of evaluating the antibacterial activities using the drop-test method, the antibacterial activity of CVD graphene was 88.9%, and the antibacterial activity was improved to 92.6% by fluorination.
(2) Induction of artificial neuronal differentiation using electrical stimulation on graphene electrode was evaluated for two types of neurons, neuroblastoma cells (SH-SY5Y) and rat hippocampal neurons. Electrical stimulation through graphene successfully induced the neuronal differentiation of neurons. Neuronal differentiation of two types of neurons in pristine graphene (PG) and oxygen plasma-treated graphene (OG) showed different trends by electrical stimulation. In the case of SH-SY5Y, the longest length of differentiated neurites on electrical stimulation with pristine graphene (PGST) was increased to 147.6% compared to cell culture plate (CCP), and the average length of differentiated neurites on electrical stimulation with oxygenated graphene (OGST) was increased to 171.5% compared to CCP. In the case of rat hippocampal neurons, the longest and average lengths of differentiated neurites on PGST were increased by 107.5% and 110.9%, respectively. In addition, the number of differentiated neurites per cell was found to significantly increase in OGST to 281.9% compared to CCP. Therefore, electrical stimulation using PG can be utilized for length differentiation of neurites, and electrical stimulation using OG is expected to be used for network formation of neurites.
(3) Graphene electrodes for electrical stimulation can be operated by ion-sensitive field effect transistor (ISFET). A reference electrode is essential for ISFET operation, and an Ag/AgCl reference electrode is generally used. However, the Ag/AgCl reference electrode has limitations in miniaturization due to its structure and size. Therefore, in this study, a fluorine-functionalized graphene electrode was developed to replace the existing Ag/AgCl reference electrode. The fluorine-functionalized graphene reference electrode was insensitive to H+, Na+, K+ and Ca2+ ions. Furthermore, the potential change measured in the real-time and long-term stability evaluation was very low (1.65 mV/h). The sensitivity of Na+ ion on the graphene-ISFET was 43.5 mV/dec when measured by Ag/AgCl reference electrode, and the sensitivity when measured by fluorinated graphene reference electrode was 59.2 mV/dec.
