환경스트레스는 작물의 생장과 생산성을 제한하는 주요 요인이다. 식물은 저온, 염분, 그리고 가뭄 등의 환경스트레스를 극복하기 위한 다양한 기작을 가지고 있다. 식물의 원형질막은 환경스트레스에 노출되는 첫 번째 세포의 방어막으로서 펌프, 채널, 교환형 수송체 등의 단백질들을 내재하고 있으며 생장 조절 뿐만 아니라 스트레스 방어 기작에 기여한다. 이러한 원형질막 단백질들은 서로 긴밀한 신호전달체계, 인산화, 그리고 상호작용 등을 통하여 세포의 항상성을 유지시킨다. Rare cold inducible 2(RCI2) 단백질은 원형질막에 위치하며 저온, NaCl, 그리고 가뭄 스트레스에 특이적으로 발현이 증가된다고 보고되었다. 또한, RCI2 단백질의 기능은 원형질막의 전위를 조절하여 식물의 환경스트레스 내성을 증진시킨다고 알려져 있다. 아쿠아포린 PIP2 는 수분수송채널 단백질로서 세포막을 가로질러 효율적으로 수분, 이온, 그리고 몇몇의 용질을 수송한다고 밝혀져 있다. PIP2의 활성도는 단백질 N말단 및 C말단 잔기의 인산화에 의해 조절된다. 이러한 PIP2 단백질의 기능은 세포 내 수분항상성 뿐만 아니라 NaCl 및 가뭄 스트레스에도 중요하다. 이 연구는 바이오에너지 작물 카멜리나에서 RCI2와 PIP2 단백질 사이의 상호작용과 NaCl 스트레스 방어기작 조사가 목표이며, 더불어 아직 명확히 밝혀지지 않은 RCI2 단백질의 기능을 규명하고, PIP2의 새로운 수분수송활성도 조절기작을 밝혀내어 내염성 작물 개발에 기여하고자 하였다. 카멜리나의 RCI2 단백질은 작은 크기의 소수성 단백질이며 RCI2A부터 H까지 총 8개의 단백질 서열이 밝혀져 있다. 단백질 구조 예측 분석 결과에 의하면 카멜리나의 RCI2 단백질은 공통적으로 2개의 transmembrane domain을 가지고 있으며, C말단의 친수성 꼬리의 유무에 따라 tail-type(CsRCI2A/B/C/H) 그리고 no-tail type(CsRCI2D/E/F/G)으로 분류된다. 담배(tobacco) 잎에서 CsRCI2-eYFP 융합 단백질의 일시적 발현 실험 결과 CsRCI2A/B/E/F 단백질들은 공통적으로 원형질막과 세포질 내 소낭에서 관측되었다. 효모의 PMP3(RCI2 homolog) 단백질이 기능하지 못하는 돌연변이 △pmp3 효모를 이용한 상보성 실험에서 no-tail type CsRCI2A, CsRCI2E△58, 그리고 CsRCI2F△58의 발현은 △pmp3 효모의 염분내성을 회복시켰지만 CsRCI2E 및 CsRCI2F와 같은 tail-type의 발현에서는 회복되지 않았다. 환경스트레스 유형별 CsRCI2 유전자들의 전사량 변화를 확인하기 위해 저온, NaCl, 그리고 mannitol을 카멜리나 1주된 묘에 처리하였을 때, CsRCI2A/B/E/F 유전자의 발현량이 현저히 증가함을 확인하였다. NaCl shock 및 NaCl acclimation 처리조건으로 CsRCI2E 및 CsRCI2F 단백질의 축적을 조사한 결과, 두 단백질 모두 축적량이 증가하였지만 NaCl 강도, 처리기간, 그리고 농도에 따라 단백질 축적 정도가 다양함을 확인 할 수 있었다. 아쿠아포린 CsPIP2;1의 경우 NaCl 농도 및 처리기간에 따른 전사량 변화가 대조군(control)에 비해 큰 차이를 보여주지 않았지만, CsPIP2 단백질의 축적량은 NaCl 처리 농도의 증가에 따라 감소함을 나타냈다. CsRCI2와 CsPIP2;1 사이의 상호작용을 조사하기 위해 gene ontology 분석을 실행하였으며, protein binding과 관련한 유사한 주석들(annotaions)을 확인하였다. Bimolecular fluorescence complementation (BiFC) 그리고 co-immunoprecipitation (Co-IP) 실험 결과, 일반조건 및 NaCl 처리조건 모두에서 CsRCI2E/F와 CsPIP2;1 사이의 binding이 확인되었다. Xenopus laevis (아프리카발톱개구리) oocyte를 이용한 swelling assay 실험에서, CsPIP2;1의 수분수송활성도는 CsRCI2A/B/E/F의 공동발현에 의해 감소되었다. 그리고 CsRCI2 단백질들의 공동발현은 CsPIP2;1의 수분 및 이온 수송에 의한 전류(current)의 변화를 감소시켰다. 한편, tail-type인 CsRCI2E/F의 공동발현이 no-tail type CsRCI2A/B에 비해 CsPIP2;1의 수분수송 및 전류변화를 더 감소시켰다. X. laevis oocyte에 발현된 CsPIP2;1 단백질 분포를 조사한 결과, CsRCI2E 또는 CsRCI2F를 CsPIP2;1과 함께 공동발현 시켰을 때 CsPIP2;1의 단백질 분포가 현저히 감소됨을 확인하였다. 이 결과를 통해 CsRCI2E/F 단백질의 발현은 원형질막으로부터 CsPIP2;1의 분포를 저해 함으로서 세포막 내 수분 및 이온 수송이 감소됨을 알 수 있다. PIP2;1은 NaCl 스트레스 하에서 원형질막으로부터 세포내부로 내재화되며, 이 과정에 clathrin 단백질이 관여한다고 알려져있다. 또한 RCI2 단백질의 발현도 세포질 내 vesicle 에서 확인 되었다. CsRCI2 단백질의 발현이 CsPIP2;1의 내재화와 관련이 있는지 확인하기위해 wild type(WT) 및 RCI2D/H 과발현 카멜리나에 지질특이적 염색시약 FM4-64와 vesicle 고정처리시약 brefeldinA를 처리하여 vesicle의 분포 변화를 조사하였다. 그 결과, 100 mM NaCl 처리 유무와 무관하게 CsRCI2D 및 CsRCI2H 과발현 카멜리나가 WT에 비해 더 많은 세포내 vesicle을 생성하였다. CsRCI2의 발현이 clathrin-mediated endocytosis(CME) 와 연관이 있는지 확인하기 위하여, CME inhibitor인 TyrA23을 처리하여 vesicle 분포 변화를 관찰하였다. 그 결과, TyrA23의 처리에도 불구하고 WT 및 CsRCI2 과발현 카멜리나에서 vesicle 형성을 관찰 할 수 있었다. 따라서 CsRCI2의 발현에 의해 유도되는 내포작용은 CME와 무관한 pathway를 가지고 있을 것으로 추측된다. 결론적으로 CsRCI2와 CsPIP2;1의 상호작용은 CsPIP2;1의 세포내 내재화에 관여함으로써 NaCl 스트레스하에서 식물 세포가 수분 및 이온 항상성을 유지하는데 기여함을 밝혀냈다. 이러한 결과들은 환경스트레스 저항성에 대한 PM 단백질들의 재배치의 미세 조정을 이해하는데 도움이 될 것이다.
더보기Abiotic stresses are major limiting factors for crop growth and productivity. Plants show various adaptations to overcome or avoid abiotic stresses such as cold, salinity, drought, and metal ions. The plasma membrane (PM) is the first protective barrier for cells and contains various proteins, including pump-, channel-, and exchanger-type transporters for growth regulation and stress defense mechanisms. These PM proteins are reported to be tightly regulated to maintain cell homeostasis via various mechanisms involving signal transduction, phosphorylation, and protein interactions. Rare cold inducible 2 (RCI2) proteins are localized at the PM and are significantly induced by cold, salt, and drought stresses. The RCI2s are believed to enhance abiotic stress tolerance by regulating the membrane potential. Aquaporin PIP2 is a water transport channel protein that facilitates the transport of water, ions, and several solutes across the PM. The activity of PIP2s is regulated by phosphorylation of its N- and C-terminal residues. PIP2s are known to be important for water homeostasis under NaCl and drought stress. In this study, I focused on the functions of RCI2s and their interactions with aquaporin PIP2;1, which is important for cell water regulation under NaCl stress in the bioenergy crop Camelina sativa L. CsRCI2s have two transmembrane domains and can be divided into two types based on the absence (CsRCI2A/B/C/H, no-tail type) or presence (CsRCI2D/E/F/G, tail-type) of hydrophilic C-terminal tail, analyzed by amino acid sequence hydropathy and protein structure prediction. Subcellular localization analysis revealed the presence of eYFP-fused CsRCI2A/B/E/F proteins in the PM and subcellular vesicles. In RCI2 homolog △pmp3 mutant yeast, NaCl tolerance can be recovered by the expression of no-tail type CsRCI2A, CsRCI2E△58, and CsRCI2F△58, but not by CsRCI2E and CsRCI2F. To characterize the transcriptional changes of CsRCI2s under abiotic stresses, their expression levels were determined under various physiological conditions such as cold, high salt, and high osmolarity. The results showed that the transcription of CsRCI2A, CsRCI2B, CsRCI2E, and CsRCI2F genes significantly increased under NaCl, cold, and mannitol stress. Under NaCl shock or NaCl acclimation, the accumulation of CsRCI2E and CsRCI2F proteins increased to various degrees, depending on the duration and intensity of NaCl concentrations. These results indicate that each member of CsRCI2s has different characteristics under NaCl stress. In the case of aquaporin CsPIP2;1, the transcription levels were less than those of the control under NaCl stress and varied in a dose- and time-dependent manner. However, the protein accumulation of CsPIP2 at the PM decreased by NaCl stress. To understand the relationship between CsRCI2s and CsPIP2;1, similar annotations of protein binding were confirmed using gene ontology analysis. In bimolecular fluorescence complementation (BiFC) and co-immunoprecipitation (Co-IP) test, interactions between CsRCI2E/F and CsPIP2;1 were observed under normal conditions as well as NaCl stress. The water transport activity of CsPIP2;1 was decreased by co-expression with CsRCI2A/B/E/F in Xenopus laevis oocytes. Moreover, CsPIP2;1-dependent current changes by water and ion transport were decreased upon co-expression with CsRCI2s. However, the tail-type proteins CsRCI2E and CsRCI2F showed a higher inhibition of PM potential change than the no-tail type proteins CsRCI2A and CsRCI2B. The protein abundance of CsPIP2;1 expressed in X. laevis oocyte was significantly decreased by co-expression with CsRCI2E and CsRCI2F. These results suggest that removal of CsPIP2;1 protein in PM by CsRCI2s expression contributes to a reduction in water and ion transport under NaCl stress. On the other hand, the PIP2 proteins can be internalized as a vesicle in the intracellular space by clathrin-mediated endocytosis (CME) and also RCI2s can be observed in intracellular vesicle trafficking. To test the possibility of CsPIP2;1 internalization upon CsRCI2s expression, changes in the abundance of vesicles bound with lipid-specific dye (FM4-64) was observed in WT and transgenic Camelina overexpressing RCI2D (tail-type) and RCI2H (no-tail type). In CsRCI2D and CsRCI2H over-expressing Camelina, the abundance of FM4-64 stained vesicles was significantly increased not only under normal conditions, but also under 100 mM NaCl treatment. However, the increased vesicular trafficking upon CsRCI2D and CsRCI2H over-expression was not affected by CME inhibitor TyrA23. In conclusion, differential expression of CsRCI2s under various abiotic stresses might be a strategy to face stressful environments. The interactions between CsRCI2s and CsPIP2;1 contribute to maintaining the water and ion homeostasis of cell under NaCl stress through the internalization process. These results should be helpful in understanding the fine-tuning of PM protein reorganization for abiotic stress tolerance.
