Soil serves as a habitat of various fungi and keratinophilic fungi, such as Trichophyton(T.) ajelloi, Microsporium(M.) gypseum, Chrysosporium sp., M. cookei, are an important group of fungi that live in soil. These fungi are becoming a major study to trace the source of infection of the dermatophytes since it identified in infectious lesions. Hence, we collected sand from the highly populated areas in Haeundae beaches, Busan to identify the keratinophilic fungi. In mid-June 2015, We collected approximately 150g of each samples from the Haeundae beaches and seawater in sterile polyethylene bags. To remove external contamination, we scraped away the top 1 cm of the soil surface for typical sunny days during the experiment. For isolation, hair bait technique of Vanbreuseghem was used. Sterile Petri dishes were half filled with the soil samples and moistened with water and baited with keratin by burying sterile human hairs in the soil. The dishes were incubated at 25°C and fungal growth was examined daily for 4 weeks. Isolated fungi were stained with lactophenol cotton blue and the hyphae, microconidia and macroconidia were observed it under the phase contrast microscope. For the identification of fungi, microscopy and fungal culture on Sabouraud‘s dextrose agar was performed. From the 100 soil samples of Haeundae beaches, the keratinophilic fungi (KPF) were isolated in 21 cases. The most common of fungi in soil were M. gypseum (43%), Chrysosporium sp.(35%), T. ajelloi (13%), M. cookei(9%). KPF has been mainly isolated from dry sand than from wet sands in seawater and highly populated areas. M. gypseum was isolated from the soil samples of over all area, Chrysosporium spp was isolated from dry sand in heavily populated areas. T. ajelloi was isolated on the outside of beach in sand area and M. cookei was isolated between wet and dry sand. In this study, we found that the isolation rate of the KPF depend on place and the KPF isolated area is high densely populated areas that are dry sand than wet sand. Our results provide a basic understanding of the distribution of keratinophilic fungi.
