구제역 (Foot and mouth disease) 은 포유동물의 질병 중에서 가장 심각한 질병이고 특히, 발굽이 둘로 갈라진 우제류 (소, 돼지, 양, 염소, 낙타 등)에서 않은 경제적 피해를 유발한다. 구제역 바이러스는 Picornaviridae, Aphthovitus에 소하며, 바이러스 입자는 60개의 단백질로 구성된 icosahedral 구조의 외피 (Capsid)와 한 가닥의 RNA genome으로 구성된다. 외피는 4종류 (VP1, VP2, VP3, VP4)의 바이러스 단백질로 구성되는데 외부에 노출된 단백질은 VP1- VP3이며, VP4는 작은 크기의 단백질이며 내부에 위치하고 있다. 구제역 바이러스는 7종 (0, A, C, Asia-1, SAT1, SAT2, SAT3)의 혈청형과 다양한 아혈청형이 존재하는 것으로 밝혀져 있다. 구제역 예방을 위해 불활화 바이러스 백신이 상용화되어 판매되고 있으며, 일반적으로 바이러스를 세포배양 후 ethyleneimine으로 불활화하고 adjuvant와 혼합하여 백신으로 사용한다. 구제역 백신은 발생 할 수 있는 다양한 혈청형의 바이러스에 대처하기 위해 다양한 바이러스 혈청형이 포함된 다 역가로 제조된다. 대부분의 지역 및 국가에서 구제역 발생 시 백신 접종으로 효과적으로 구제역을 통제하고 있지만, 모든 국가에서 백신을 생산하지는 않는다. 모든 국가에서 구제역 백신 제조가 제한되는 이유는 구제역 백신 생산 시 바이러스 유출에 의한 안전성 문제와 불활화후 바이러스가 잔존 할 수 있는 가능성 때문이다. 그래서, 안전하고 쉽게 생산 할 수 있는 백신개 발이 필요하다.
구제역 발생 후 축산물 수출을 위해서는 감염 및 감염이 의심되는 동물을 모두 살 처분해야만 한다. 살 처분에 후에 구제역 청정지역 및 국가임을 증명하기 위하여 방대한 양의 혈청을 검사를 실시하여야 하는데, 현재 구제역 항체 검사를 위한 표준 검사법은 바이러스 중화시험법으로 많은 실험과정이 필요하고, 생물학적 실험으로 숙련된 시험자가 필요하며, 바이러스 배양을 위해서는 특별한 생물학적 차페 시설이 필요한 단점이 있다. 대량의 혈청 검사를 위해서는 ELISA를 이용하고 있지만, 현재 OIE에서 인정받은 Liquid Phase Blocking (LPB) ELISA 는 않은 실험단계와 낮은 특이도와 대량의 시료 검사에는 적합하지 않은 것으로 알려져 있다. 이를 보완하기 위하여 Mackay 등에 의해 Solid Phase Blocking ELISA가 개발되어 특이도를 개선하였지안, 불활화 바이러스를 고정화 항원으로 사용하는 등 아직 대량검사에는 적합하지 않아, 안전하고, 신속하며, 구제역 바이러스 혈청형에 특이적인 항체를 검출 할 수 있는 기술 개발이 요구된다.
2002년 5월 OIE에서는 구제역백신 지역 및 국가에서는 구제역 백신 동물과 구제역 감염 동물을 감별할 수 있는 기술을 이용하여 구제역 감염동물이 없다는 것으로 증명하면 구제역 청정국 지위를 부여한다고 발표하였다. 구제역 백신 접종 동물과 감염동물을 감별하기 위해서는 FMDV Non Structural protein (NSP)에 대한 항체여부와 관련이 있다. NSP는 바이러스 복제과정에서는 발현되는 단백질로 구제역 백신 제조단계에서는 세포단백질과 NSP를 제거하는 정제공정으로 인해 감염된 동물에서만 NSP에 대한 항체가 형성된다. 현재, 3ABC와 3AB 단백질이 가장 우수한 진단 마커로 알려져 있으며, 일부 문헌에서는 과거 및 현재의 감염여부를 판정할 수 있는 것으로 보고되었다. 또한, NSP는 구조 단백질과 달리 동질성이 높아 바이러스 혈청형에 관계없이 적용할 수 있는 특징이 있다.
본 연구의 목적은 1) 곤충세포에서 유전자 재조합 FMD Virus Like Particle을 제작하여 면역학적 특성을 연구하여 안전한 백신의 가능성을 탐색하고, 2) 구제역 바이러스 0형에 대한 중화항체 검사를 위한 진단법을 VLP 항원 trapping ELISA를 이용하여 개발하고 평가하고자 하며, 3) Epitope-blocking ELISA를 개발하여 구제역 바이러스 종류 및 축종에 관계 없이 구제역 감염동물과 백신접종 동물을 감별할 수 있는 기술을 개발하여 유효성을 측정 하고자 한다.
제 1장. 배큘로바이러스에서 발현한 유전자 재조합 FMDV Like Particle 구조의 면역학적특성
구제역 바이러스가 P1단백질에서 성숙되기 위해서는 단백질분해효소 3C가 필요하다. 본연구에서는 구제역 바이러스의 P1과 3C 유전자를 각각의 프로모터를 이용하여 발현 할 수 있는 재조합 배큘로바이러스를 제작하였고, 곤충세포 (sf9)에 접종하여 pentamer 유사입자를 생산하였다. 3C 단백질에 의해 생산된 바이러스 유사입자는 western blot에 의해 확인되었고, 항원성은 간접 ELISA에 반응성이 입증되었다. CsCI 농도구배 초원심 분리법으로 확인한 결과 동일한 농도에서 구제역 바이러스 구조와 유사한 pentamer 입자가 관찰되었고, 전자현미경에서 관찰한 결과 바이러스 조립과장의 pentamer 크기인 7-8nm의 입자가 확인되어 곤충세포에서 바이러스 단백질이 조립되었다는 것을 알 수 있었다. 또한, pentamer 유사 입자를 마우스에 면역 후 2주 후에 높은 수준의 구제역 바이러스에 특이적인 항체와 중화항체가 유도되는 것으로 확인되었다. 이와 같이 비감염성 재조합단백질은 항체 진단법 개발 및 구제역 예방을 위한 백신 개발의 가능성을 증명하였다.
제 2장. 구제역 바이러스 O형 중화항체 검출을 위한 유전자 재조합 VLP antigen trap
Foot and mouth disease (FMD) is the most contagious disease of mammals and has a great potential for causing severe economic loss in susceptible cloven-hoofed animals. FMDV belongs to the Aphthovirus genus of the family Picomaviridae. Its viral particle contains a single-stranded RNA genome within an icosahedral capsid consisting of 60 copies of each of the four proteins, VP1-VP4. The particle shell is made up from the three larger structural proteins, VP1-VP3, while the smaller VP4 is located internally. Seven serotypes of FMDV (types O, A, C, Asia 1, and the South African Territories [SAT] types 1-3) and a variety of subtypes are known. Inactivated virus vaccines of varying composition are available commercially. Typically, virus is used to infect a suspension or monolayer cell culture and the resulting preparation is clarified, inactivated with ethyleneimine and blended with adjuvant. Many FMD vaccines are multivalent to provide cover against the different serotypes likely to be encountered in a given field situation. Although vaccination against FMDV by using inactivated virus during outbreak is effective, this kind of vaccines is not produced in many FMD-free countries. The restriction is because of the safety concerns about the likelihood of outbreaks due to the release of virus from vaccine production plants or the presence of residual live virus in chemically inactivated vaccine. Therefore, development of a safe and easily adaptable vaccine would be of major interest. Eradication by culling infected and suspected herds is ruthless but necessary to reopen export markets. Large scale serological testing is necessary in order to evaluate the effectiveness of eradication programs. The international standard test for FMD antibody detection is the virus neutralization test (VNT), but this test is laborious, results may be variable due to its biological nature and must be performed in specialized high containment facilities. When large numbers of sera need to be tested, a high throughput can be managed by first screening the samples in an ELISA. The liquid phase blocking ELISA (LPBE) is approved for this purpose by the Office International des Epizooties (OIE). However, the LPBE, due to its many reaction steps, low specificity and low testing capacity, is not suitable for testing large amounts of samples. The solid phase competition ELISA developed by Mackay et al. has improved specificity but retains other characteristics that are less than ideal for mass serology. Therefore, the development of quick, safety and simple serotype specific FMDV antibody ELISAs was warranted, In May 2002, the OIE updated its International Animal Health Code in light of advances in diagnostic tests, allowing countries that vaccinate in the face of an outbreak of FMD to regain disease-free status after 6 months if they can differentiate vaccinated from infected or convalescing animals. Differentiation of convalescing or infected animals is based on identifying antibodies to the NSPs of FMDV. The NSPs are expressed only by replicating viruses. Inactivated vaccines are purified to remove cellular proteins and NSPs, and therefore only animals that have been infected with live virus should develop antibodies to these proteins, Currently, the polyproteins 3ABC and 3AB appear to be most promising as diagnostic antigens. Tests for antibodies to some NSPs of FMD virus are useful in providing evidence of previous or current viral replication in the host, irrespective of vaccination status. NSPs, unlike structural proteins, are highly conserved and therefore are not serotype specific and as a consequence, the detection of these antibodies is not serotype restricted.
The aims of this thesis are as follow; 1) To understand the immunological characteristics of recombinant FMD virus-like-particle (VLP) structure expressed by baculovirus for developing a safe and easily adaptable vaccine. 2) The development of recombinant VLP antigen trapping ELISA for detection against FMDV O serotype neutralizing antibody and the validation of for the serological detection of antibody to serotype O foot-and-mouth disease. 3) An epitope-blocking ELISA (EB-ELISA) is developed to distinguish animals infected with FMDV from those immunized with commercial vaccines independent FMDV serotype and animal species.
CHAPTER Ⅰ:
Immunological Characteristics of Recombinant FMD Virus-Like-Particle Structures Expressed by Baculovirus
The assembly of Foot-and-mouth disease virus (FMDV) requires the cleavage of P1 polyprotein by protease 3C into individual structural proteins, In this study, the recombinant pentamer-like structures were produced in insect (Sf9) cells that were inoculated with recombinant baculoviruses that expressed, simultaneously, the genes for the P1 and 3C proteins of FMDV from individual promoters. The FMDV pentamer-like structures were processed by viral 3C protease, as shown in Western blots, and were antigenic, as revealed by their reactivities in an indirect ELISA. Analysis by CsCI gradient centrifugation showed that the pentamer-like structures were similar to authentic pentameric subunits from FMDV in terms of sedimentation velocity. Furthermore, the pentamer-like structures induced high levels of FMDV-specific antibodies in mice following immunization, Observations made under the electron microscope revealed that the pentamer-like structures expressed by insect cells selfassembled to form pentameric subunits of 7-8 nm in diameter, which resemble the authentic FMDV (23±2 nm in diameter). Furthermore, the empty capsid-like particles or intermediates induced high levels of FMDV-specific antibodies in mice following immunization and neutralization antibodies were induced in the second week after vaccination. The potential utilities of these recombinant, noninfectious, FMDV empty capsids for diagnostic and vaccine purposes are promising.
