Characterization of the binding partners of a transmembrane adaptor protein LIME in T cells

LIME 은 transmembrane adaptor 단백질로 Lck SH2 domain 에 결합하는 TCR signaling에 중요한 역할을 한다고 연구되어졌다.
기존 연구에서 LIME 에 결합하는 단백질을 찾기 위해 Yeast two-hybrid screening 이 수행되어졌고 그 결과 또 다른 결합 단백질인 Grb2 와 Lad를 찾아내었다. 이 논문에서는 이들 단백질과 LIME 과의 상관관계와 downstream signaling 기작에 대해 연구하였다.
먼저 LIME은 in vitro에서 Lck의 인산화에 의존하여Grb2와 결합하는 데 결합위치는 Grb2의 SH2 domain이 LIME의 137 tyrosine site에 결합했다. 또한 LIME은 293T 세포에서 또 다른 adaptor 단백질인 Lad와도 결합했다. 그 결합 형태 역시 Grb2 와 같이 Lck 의 인산화에 의존하여 LIME의 137tyrosine site에 Lad의 SH2 domain이 결합하는 형태였다.
더욱이 Jurkat T세포에서의 Lad와 LIME의 결합은 CD3 stimulation 시 6시간에서 9 시간에 결합을 하였고 그 이후에는 결합이 사라지는 것을 확인할 수 있었다. 이 결과로 보아 LIME 과 Lad 의 결합은 time-dependent한 결합으로 확인할 수 있었다. 더 나아가 T cell에서 Lad와 LIME의 위치변화를 확인하였는데 그 결과 Lad 와 LIME 이 T cells 과 APC conjugation 1 시간 이전에는 같이 위치하지 않다가 3 시간에서는 synapse에서 빠져 나가면서 같이 위치하였고 6 시간에는 synapse 의 반대방향에서 같이 위치하였다. 이것으로 미루어 보아 LIME 과 Lad 의 결합은 T cell activation후 early stage가 아닌 later stage에서 이루어 지고 그 단계에서 기능을 수행할 것으로 여겨진다.
비록 앞의 실험결과로 Lad와 LIME이 later stage에서 어떤 기능을 하는지는 확실히 알 수는 없지만 기존에 Lad가 chemokine과 integrin에 의해서 T cell migration 에 관여한다는 연구결과가 있었기 때문에 아마도 LIME이 이러한 기능을 하는 Lad를 recruit하여 더 늦은 단계에서 T cell migration에 관여할 것으로 여겨진다.
LIME is a transmembrane adaptor protein identified as a binding partner of the p56Lck SH2 (Src-homology 2) domain, and contains a short extracellular domain, a transmembrane domain and a cytoplasmic tail containing five tyrosine-based motifs. The protein is expressed in hematopoietic cells and lung. From the previously yeast two-hybrid screening with LIME as bait, Grb2 and Lad were identified as the binding partners of LIME. Previously Lad was shown to function as an adaptor molecule in the membrane proximal region by providing docking sites for many other signaling molecules, containing an SH2 domain, a proline-rich SH3 binding motif and four tyrosine-based motifs at C- terminus.
Here, based on these previous findings, we studied the downstream signaling pathways regulated by LIME. In the binding assay in 293T cells, Grb2 associated with tyrosine 137 of LIME in an Lck phosphorylation-dependent manner via its SH2 domain. Besides, in 293T cells, LIME ass0ciated with an another adaptor protein, Lad. Using various mutants of Lad and LIME, the binding motif was mapped to tyrosine 137 of LIME and the SH2 domain of Lad. Notably, in Jurkat T cells stably expressing CD8-LIME chimera, binding of Lad to LIME was inducible upon TCR stimulation. The binding was not detected until 3hr after TCR stimulation, but was induced at 6hr, maintained up to 9hr and vanished at 24hr. This result indicates that the binding of Lad to LIME is induced at the later stage of T cell activation. Combined with the fact that the expression of Lad and LIME is inducible in T cells, the binding pattern suggests that LIME may function at the later stage of T cell activation through recruitment of Lad.
To further understand the meaning of recruitment of Lad by LIME occurring at the later stage of T cell activation, we studied the localization of Lad and LIME in a time-dependent manner after T cell-APC (antigen presenting cell) conjugation. Under confocal microscopy, at 3-6hrs after conjugation, Lad and LIME co-localized at the opposite site of synapse in approximately 25-30% of the cells. Although, at this stage, we do not have explanation for a function of LIME-Lad interaction, one possible hypothesis is that by recruiting Lad at the later stage, LIME may regulates adhesion and migration of T cells. In relation to this, Lad was previously shown to regulate chemokine or integrin-dependent T cell migration. Perhaps, at around 6hr after APC-T cell conjugation, LIME and Lad translocates to the opposite side from synapse and regulates migration.
Currently, the function of LIME mediated through the inducible recruitment of Lad is under investigation.
