Phenotypic transition of vascular smooth muscle cells and endothelial cells by miRNA-155-5p = NF-κB-의존적 micoRNA-155-5p에 의한 혈관평활근세포와 혈관내피세포의 표현형 변화

체내 염증성 사이토카인 생성의 증가는 다양한 심·혈관계 질환을 유도한다. 이러한 사이토카인 중에서 종양괴사인자(TNF-α)는 다양한 질환 발병의 핵심 생체분자로 보고되고 있으나, 정확한 분자 기전은 명료하게 밝혀진 바가 없다. 따라서 본 연구에서는 TNF-α에 의한 혈관구성세포인 혈관내피세포와 혈관평활근세포의 기능 및 항상성 조절 기전에 대한 연구를 수행하였으며, 다음과 같은 연구결과를 확보하였다.
첫째, TNF-α에 의한 NF-κB의 활성화는 miRNA-155-5p의 생성을 유도함으로써 혈관평활근세포에서 soluble guanylyl cyclase (sGC)의 발현을 억제하고, 이로 인하여 혈관내피세포-특이적인 일산화질소합성효소(eNOS)에 의해 생성되는 일산화질소(nitric oxide, NO)에 의한 cGMP의 생성을 억제하여 NO에 의한 셍체신호를 혈관평활근세포 내로 전달하는 기능을 차단함을 확인하였다. 이러한 현상은 수축성 혈관평활근세포의 표지 단백질들의 발현을 감소시킴으로써 혈관의 이완반응을 억제하고, 증식성 혈관평활근세포로의 표현형 전환을 유도하여 세포증식을 촉진하는 결과를 유발하였다. 따라서 혈관평활근세포에서 TNF-α에 의해 의존적으로 발현이 증가된 miRNA-155-5p는 sGC의 발현 억제를 통해 혈관이완을 저해할 뿐만 아니라 혈관평활근세포의 표현형 변화를 통해 세포증식을 촉진하여 혈관내경이 좁아지게 되고 혈압상승을 동반하는 동맥경화를 유발할 수 있음을 세포실험과 동물실험을 통해 확인하였다.
둘째, 고지방식이를 투여한 마우스에서 혈관의 노화현상이 촉진되고 혈압이 상승되었으며, 혈액 내에 염증성 사이토카인과 mRNA-155-5p의 발현이 증가이 증가되는 현상을 확인하였다. 한편, 혈관내피세포에서 TNF-α를 처리한 결과 NF-κB에 의존적으로 miRNA-155-5p의 발현이 증가되고 혈관내피세포의 노화가 촉진됨을 확인하였으며, 이러한 현상은 miRNA-155-5p의 발현 억제제를 처리한 결과 효과적으로 회복됨을 확인하였다. TNF-α에 의해 의존적으로 증가되는 miRNA-155-5p는 eNOS의 발현을 억제하여 NO의 생성을 억제함을 확인하였다. 뿐만 아니라 miRNA-155-5p는 PI3K의 촉매 단위체인 p110α의 발현을 억제함으로써 하부 신호전달 분자인 AKT와 MDM2의 활성화를 억제하여 p53의 안정화를 촉진하고 핵 내에 축적되는 현상을 확인하였다. 핵 내에 축적된 p53은 세포주기 억제제인 p21의 발현을 증가시켜 혈관내피세포의 노화를 촉진함을 확인하였다. 한편, miR-155가 결핍된 마우스에서는 비만으로 인한 혈관노화와 혈압상승이 유의하게 억제됨을 확인하였다. 이러한 결과는 비만으로 인하여 유발되는 만성염증은 NF-B에 의존적인 miR-155-5p의 발현 증가를 통해 PI3K/AKT/MDM2/p53의 경로를 조절하여 혈관내피세포의 노화 촉진을 유발하고 고혈압과 같은 혈관질환을 유발할 수 있음을 제시하고 있다.
이상의 연구결과를 종합해 볼 때, 염증으로 인한 NF-κB 의존적인 miRNA-155-5p의 발현의 증가는 혈관내피세포와 혈관평활근세포의 기능억제는 물론 이들 두 세포 간의 상호작용을 억제하여 다양한 심혈관계 질환의 발병원인이 될 수 있음을 제시하고 있으며, 이를 통해 혈관에서 발현되는 miRNA-155-5p는 심혈관계 질환의 치료용 표적분자로서의 활용가치가 있음을 밝혔다.
The increase in the production of systemic inflammatory cytokines induces various cardiovascular diseases. Among these cytokines, tumor necrosis factor-alpha (TNF-α) is a major risk factor of various diseases including vascular disease, however, the underlying molecular mechanism is not clearly elucidated. In this study, I examined the mechanism by which inflammation-responsive microRNA (miR)-155-5p is critically involved in regulation of vascular function and homeostasis.
Part I. NF-κB-responsive miR-155-5p induces functional impairment of vascular smooth muscle cells by downregulating soluble guanylyl cyclase
Vascular smooth muscle cells (VSMCs) play an important role in maintaining vascular function. Inflammation-mediated VSMC dysfunction leads to atherosclerotic intimal hyperplasia and preeclamptic hypertension; however, the underlying mechanisms are not clearly understood. I analyzed the expression levels of miR-155-5p in cultured VSMCs, mouse vessels, and clinical specimens and then assessed its role in VSMC function. Treatment with tumor necrosis factor-α (TNF-α) elevated miR-155-5p biogenesis in cultured VSMCs and vessel segments, which was prevented by nuclear factor-kappa B (NF-κB) inhibition. MiR-155-5p expression was also increased in high-fat diet-fed ApoE-/- mice and in patients with atherosclerosis and preeclampsia. The miR-155-5p levels were inversely correlated with soluble guanylyl cyclase β1 (sGCβ1) expression and nitric oxide (NO)-dependent cGMP production through targeting the sGCβ1 transcript. TNF-α-induced miR-155-5p caused VSMC phenotypic switching, which was confirmed by the downregulation of VSMC-specific marker genes, suppression of cell proliferation and migration, alterations in cell morphology, and NO-induced vasorelaxation. These events were mitigated by miR-155-5p inhibition. Moreover, TNF-α did not cause VSMC phenotypic modulation and limit NO-induced vasodilation in aortic vessels of miR-155-/- mice. These findings suggest that NF-κB-induced miR-155-5p impairs the VSMC contractile phenotype and NO-mediated vasorelaxation by downregulating sGCβ1 expression. These data suggest that NF-κB-responsive miR-155-5p is a novel negative regulator of VSMC functions by impairing the sGC/cGMP pathway, which is essential for maintaining the VSMC contractile phenotype and vasorelaxation, offering a new therapeutic target for the treatment of atherosclerosis and preeclampsia.
Part II. inflammation-responsive miR-155-5p promotes endothelial senescence through the PI3K/AKT/MDM2/p53 and eNOS/NO pathways.
Endothelial senescence is a key risk factor in several cardiovascular diseases and can be induced by a variety of factors including obesity-induced chronic inflammation; however, the underlying molecular mechanisms are not clearly understood. I examined a functional role of inflammation-responsive miR-155-5p in vascular senescence and dysfunction in cultured endothelial cells (ECs) and high fat diet (HFD)-fed obese mice. Obese mice induced vascular senescence, inflammation, and miR-155-5p biogenesis, along with increased blood pressure. Treatment of ECs with TNF-α also upregulated miR-155-5p biogenesis and EC senescence, whose events were suppressed by an NF-κB inhibitor and an miR-155-5p inhibitor, suggesting that NF-κB-dependent miR-155-5p is responsible for inflammation-associated EC senescence and dysfunction. Moreover, NF-κB-responsive miR-155-5p biogenesis downregulated the PI3K subunit p110α expression, subsequently resulting in inhibition of AKT-dependent mouse double minute 2 homolog (MDM2) phosphorylation, prevention of ubiquitination-dependent p53 degradation, and upregulation of p21 expression in cultured ECs and HFD-induced obese mice. HFD-induced vascular senescence and dysfunction were significantly suppressed in miR-155-/- mice. These results suggest that NF-κB-responsive miR-155-5p plays an important role in EC senescence and vascular dysfunction by regulating the PI3K/AKT/MDM2/p53 axis.
