골조직 배양에 있어서 골흡수에 미치는 Bovine Serum Albumin의 영향에 관한 연구 = EFFECTS OF BOVINE SERUM ALBUMIN ON BONE RESORPTION IN TISSUE CULTURE

This research was performed to elucidate the effect of bovine serum albumin (BSA) on
bone resorption in organ culture.
Two hundred μCi ^45CaCl_2 was injected subcutaneously to a mother rat on the 17th
day of gestation. Nineteen-day fetal rats prelabelled with ^45Ca were removed from a
mother rat and ulnae and radii were isolated from fetus with aseptic techniques. One
side of paired fetal ulnae and radii was employed as control group and the other side as experimental group.
While control group was cultivated in BFJb media, experimental group was cultivated in
BGJb media added with BSA in the final concentration of 0.1%, 0.2% and 0.4%.
After 24, 48 and 72 hours of culture, radioactivities of ^45Ca released into medium in
each period and radioactivities still remained in the bone which was cultivated for 72 hours were measured with liquid scientillation counter.
Bone resorption rate was calculated by the % ratio of radioactivity of ^45Ca released
into medium to total radioactivity of that was in the bone at the beginning of culture.
The effect of BSA on bone resorption was observed by the ratio of bone resorption of
experimental group to control.
The results were as follows.
1. At the concentration of 1㎎/ml BSA, bone resorption tended to be increased with time but showed no significance.
At the concentration of 2㎎/㎎ BSA, the tendency of bone resorption was similar to 1㎎/ml BSA group but showed somewhat greater effect than 1㎎/ml BSA group. At the concentration of 4㎎/ml BSA, the greatest and significant bone resorption was shown in all of this group.
2. The enhancing effect of BSA on bone resorption showed clearly the dose-dependent
tendency and the differences of the bone resorption rate between control and
experimental group were statistically sinificant in higher concentration of BSA medium.
