트롬빈(Thrombin, TB)은 체내 출혈이 일어났을 때 그 양이 급격하게 증가하는 단백질의 하나로, 뇌출혈 등의 조기진단에 있어서 가장 유용한 지표 단백질로 최근 전기화학바이오센서의 개발을 통해 활발히 연구되고 있는 단백질이며 본 연구는 이러한 TB의 조기진단을 목표로 높은 감도의, 경쟁력 있는 전기화학 TB aptamer 센서의 개발에 목적을 두고 있다.
특히 이 바이오센서개발을 위해서 사용한 바이오 폴리머(Poly dopamine, PD)와 Aptamer는 생물의학 응용분야, 특히 바이오센서 분야에서 전도유망한 물질이지만 PD의 경우, 자신의 낮은 전도성(Conductivity)으로 인해, 전기 바이오센서 개발에서의 응용이 제한적인 단점이 있고, 이에 반해 Aptamer는 그 제조 방법이 기존의 Antibody보다 비교적 간단하여 경제적인 이점과 함께 대상 단백질과의 선택적 결합성이 뛰어나 대상 단백질의 검출센서 개발에 적극 활용되고 있는 추세이다.
그러므로 센서 개발을 위해서는 바이오센서에서의 PD 필름의 전도성의 증가가 필수적이다. 이 고분자 필름의 전도성을 증가시키기 위해, 다양한 금속과 금속 산화물 나노 입자를 고분자 필름에 사용하였다. 나노 입자를 포함하거나 나노 입자 층이 만들어진 PD 필름은 대상 분자의 고정에 활용할 수 있기 때문에 이러한 PD 필름을 준비하는데 있어 독특한 물리적, 화학적 특성을 가진 골드 나노 입자(AuNPs)를 PD 필름에 섞고, 층을 만들어 TB aptamer 센서의 제조를 위한 플랫폼으로 사용하였다.
TB aptamer 센서는 AuNPs와 Thiolated TB aptamer(TBA)의 결합반응을 통하여 고정화하여 TB/AuNPs/TBA 센서 플랫폼을 만들었고, 이 센서의 표면은 EIS, AFM, XPS와 SEM 기술을 사용하여 검증하였다.
트롬빈 검출을 위한 전기화학 신호 생성을 위한 TB와 결합시킨 메틸렌 블루(MB)는 산화, 환원 신호 표지로 사용되었다. 신호 감도의 증폭을 위해, TB과 결합된 AuNPs와 복합 MB를 이용하였고 이 TB/AuNPs/MBs의 결합 분자의 검증은 TEM, DLS, NMR 그리고 UV/VIS를 활용하였다.
PD와 PD/AuNPs 필름과 TB/AuNPs/MBs의 전기화학센서에 적용여부를 판단하기 위하여 전기 산화, 환원 반응을 측정하였고, 순환 전압전류법(Cyclic voltammetry, CV)를 사용하여 특성화 하였다.
최종적으로 TB의 검출에는 기존의 CV보다 높은 민감도와 검출 능력을 가진 사각 전압전류법(Square voltammetry, SWV)를 사용하였고, TB 검출은 aptamer의 결합 부분의 대한 TB/AuNPs/MBs 결합분자와 Free TB 사이의 경쟁적인 결합을 기반으로 측정되었다. 고정된 양의 TB/AuNPs/MBs 결합분자의 존재 하에 Free TB의 농도가 증가할 시에 전체적인 전류의 측정값이 감소가 되고, SWV의 전류 측정결과가 TB의 농도에 따라 비례적으로 감소함을 확인할 수 있었다. 최적화 된 실험 환경에서 선형 동적 영역(linear dynamic range)은 2pM ~ 10nM로 검출한계는 0.97 pM로 측정되었다. 또한 실질적인 검출 테스트를 위하여 TB를 포함하고 있는 인간 혈청 샘플에 대한 시험을 통하여 만족스런 검출 결과를 얻을 수 있었다.
Thrombin (TB) is a coagulation and serine protease that converts soluble fibrinogen to insoluble strands of fibrin as well as catalyzing many other coagulation-related reactions responsible for blood clotting. It plays an important role in thrombosis and haemostasis and its detection is very useful for early diagnosis of intracerebral hemorrhage and pulmonary metastasis. It is not present in blood under normal condition but can be elevated at low-micromolar concentrations during coagulation process. Thus, the development of a high sensitive aptamer sensor for the detection of thrombin would be of great interest.
Recently, for TB detection, many kinds of electrochemical sensors were developed with various novel bio materials and detection strategies. Among those novel materials, aptamers and the bio-inspired polymer, poly-dopmaine (PD) holds great promise for their use in biomedical applications, particularly in biosensor applications. Aptamers has
advantageous than antibodies because of their simple making process and target affinity. Otherwise, due to PD’s low conductivity, their application in electrochemical biosensors development is limited. Thus, it needs to increase the conductivity of PD film for biosensor applications. To increase the conductivity, various metal and metal oxide nanoparticles can be encapsulated into the polymer film. The nanoparticles-doped/deposited PD film can be used for further modification or biomolecules immobilization.
The aim of the present study is to prepare a nanoparticles-doped/deposited PD film that can be used for the fabrication of a high sensitive competitive electrochemical TB aptamer sensor. Gold nanoparticles (AuNPs), which have unique physical and chemical properties were doped/deposited into/onto the PD film and was used as a platform for the fabrication of a TB aptamer sensor. The TB aptamer sensor was fabricated by immobilizing a thiolated TB-binding aptamer (TBA) onto the AuNPs doped/deposited poly-dopamine/ film (PD/AuNPs). The PD/AuNPs surface was characterized using UV-Vis spectroscopy, AFM, XPS and SEM techniques.
For the electrochemical signal generation, TB-conjugated methylene blue (MB) was used as nonenzymatic- redox labels. The detection strategy in order to amplify the signal sensitivity, AuNPs-supported TB-conjugated multiple MB molecules (TB/AuNPs/MBs) were prepared. The TB/AuNPs/MBs conjugate was characterized by TEM, DLS, and UV/Vis spectroscopy. The electrochemical activities of the PD and PD/AuNPs films, and MB in the TB/AuNPs/MB conjugate were studied using cyclic voltammetry. Since, the reduction process of MB+ to MB was more pronounce than oxidation process, reduction process was used for analytical detection. For the high sensitive detection, high sensitive square wave voltammetry (SWV) was used. The TB detection was based on the competitive binding between multiple MBs-conjugated-TB and free-TB for the aptamer binding sites. When the concentration of free-TB increased in binding solutions in the presence of fixed amount of conjugate, the more amount of free-TB bonded with TBA. As a result, the current response was found to deceased. SWV responses revealed that the difference in current response (?I) between before and after competitive binding directly proportional to the concentration of free-TB. This result suggested that TB can be successfully detected with the TB aptamer sensor. Under the optimized condition, the linear dynamic range and the detection limit were determined to be 2 pM ~ 10 nM and 0.97 pM, respectively. The applicability of the proposed TB aptamer sensor was examined by applying it to human serum samples for the detection of TB and satisfactory results were obtained.
