목적: The purpose of the present study was to evaluate the effects of Ecabet Sodium(ES) on neomycin-induced apoptosis in hair cell of zebrafish. 방법: Transgenic zebrafish embryos were produced by paired matings of adult fish raised at 28℃. Zebrafishs were maintained with live paramecia and plant-based food for 5 days in embryo medium (1 mM MgSO4, 120 uM KH2PO4, 74 uM Na2HPO4, 1 mM CaCl2, 500 uM KCl, 15 uM NaCl, and 500 uM NaHCO3 in distilled water). Five day post-fertilization zebrafish larvae were exposed to ecabet sodium in each concentration of 0, 5, 10, 20, 40, and 80 μg/mL. After washing with embryo medium three times, neomycin sulfate was added to the embryo medium at final concentration of 125 uM for 1 hour. Fishes were anesthetized with Tricane for 5 min.Transgenic zebrafish (Brn3C: EGFP) naturally presents green-colored neuromasts which are visible using fluorescence microscopy without any staining. Hair cells were counted within neuromasts of the supraorbital (SO1 and SO2), otic (O1), and occipital (OC1) positions of lateral line using fluorescence microscopy and confocal microscopy (LSM5 PASCAL; Carl Zeiss, Jena, Germany).Cell apoptosis was examined by the deoxynucleotidyl transferase (TdT)-mediated dUTP-biotin nick end labeling (TUNEL) assay. Mitochondrial structure was observed after the fluorescent dye 2-[4-(dimethylamino)styryl]-N-ethylpyridinium iodine (DASPEI; Invitrogen, Carlsbad, CA, USA) stain. Ultrastructural analysis was performed with scanning electron microscopy, using a model S-4700 microscope (Hitachi) 결과: In our analysis, we demonstrated that ES (40 ug/Ml) ameliorated neomycin induced hair cell damage in zebrafish. 결론: Neomycin induces apoptosis of hair cells in zebrafish. Ecabet sodium attenuated neomycin-induced hair cell loss.Apoptotic cell death may be prevented by treatment with ecabet sodium.
