신경영양인자는 신경 및 비신경 조직 모두에서 중요한 역할을 하는 인자이다. 포유동물의 신경영양인자 패밀리는 신경 성장 인자, 뇌 유래 신경 영양 인자, 신경영양인자-3 및 신경영양인자-4 (NT-4)로 구성된다. NT-4는 고친화성 트로포마이오신 수용체 키나제 B (TrkB)와 저친화성 범뉴로트로핀 수용체 (p75NTR) 간의 상호작용을 통해 난포 발달, 난모세포 성숙, 신경 세포 성장 및 발달에 관여하는 성장인자이다. 뇌와 난소는 모두 신경 분포가 높은 기관이기 때문에 다양한 신경영양인자가 신경의 성장과 발달을 촉진시킬 수 있다. 따라서, 신경영양인자가 신경 및 난소 발달에 미치는 영향을 조사하는 것은 매우 중요하고 가치 있는 연구가 될 것이다. 돼지의 생식 생물학 및 중개 의학 분야는 지난 수십 년 동안 연구되었지만 신경영양인자가 난소 생리학 및 돼지의 신경 발달에 미치는 영향에 대한 연구는 아직 충분하지 않다. 따라서, 본 연구는 돼지의 배아 발달 및 신경 분화에 대한 NT-4의 역할을 조사하는 것을 목표로 하였다.
CHAPTER Ⅰ의 연구 결과, 돼지 난모세포의 체외성숙 동안 NT-4의 첨가가 난모세포의 핵 및 세포질 성숙을 향상시키는 것으로 나타났다. 이 연구에서는 돼지 난소에서 NT-4, TrkB 및 p75NTR의 분포를 확인했다. 또한, 체외성숙 동안 10ng/mL의 NT-4를 첨가하면 단위발생 유래 돼지 배아의 후속 발달 능력이 향상됐다. 따라서, NT-4는 돼지의 난포 발달에 관여하고, 난모세포의 체외성숙에 필요하다.
CHAPTER Ⅱ에서는 돼지 난구-난모세포 복합체의 체외성숙 동안 NT-4의 생리학적 역할을 조사했다. 이 연구는 성숙한 돼지 난구-난모세포 복합체에서 NT-4, TrkB 및 p75NTR의 존재를 입증하여 NT-4 관련 신호 경로가 돼지 난모세포의 성숙 및 발달에 관여한다는 것을 증명했다. NT-4는 뉴로트로핀/p75NTR 신호 전달 경로를 통해 NFKB1 전사체를 상향 조절하여 난구세포 및 난모세포 복합체의 체외성숙을 촉진하고 GDF9, BMP15, CD9 및 DNMT3A 전사체를 상향 조절하여 난모세포 품질을 향상시켰다. 체외성숙 이후 난구세포 내에서 세포 외 신호 조절 키나아제 1/2 (ERK1/2)의 인산화 수준을 비교했을 때, 인산화된 ERK1/2 수준이 대조군보다 NT-4가 처리된 난구세포에서 유의하게 더 높았다. 반면, 성숙된 난모세포 내에서 전체 및 인산화된 ERK1/2 수준은 모두 대조군 보다 NT-4가 처리된 난모세포에서 유의하게 더 높았다. 또한, 체외성숙 시, NT-4의 처리는 체외수정 및 체세포 핵이식 이후 후속 배아 발달 능력을 향상시켰다. 종합하면, NT-4는 체외성숙 동안 돼지 난구-난모세포 복합체에서 ERK1/2 신호 전달 경로를 조절함으로써 난모세포 성숙과 난구세포의 확장을 촉진하는 역할을 한다.
CHAPTER Ⅲ에서는 돼지 단위발생 배아의 체외배양 중 NT-4 보충이 단위발생 후 배아의 발달 능력과 배반포의 품질을 향상시키는 것으로 나타났다. NT-4는 CDX2, PPAG3 및 GATA3 전사체를 상향 조절함으로써 돼지 착상 전 배아 발달에서 내부 세포 덩어리 계통보다는 영양외배엽 계통의 발달에 기여할 수 있다. 배반포 세포 계통 사양의 분석에서 체외배양 시, NT-4의 보충은 대조군과 비교하여 단위발생 유래 배반포에서 영양외배엽 마커인 CDX2를 발현하는 세포의 수를 유의하게 증가시켰다. 또한, 본 연구를 통해 NT-4는 돼지 착상 전 배아 발달 동안 Hippo-Yes-associated protein (Hippo-YAP) 및 mitogen-activated protein kinase (MAPK) 경로 관련 유전자를 조절할 수 있음을 확인했다. 본 연구를 통해 체외배양 시, NT-4 첨가가 단위발생 후 돼지 체외배양 동안 영양외배엽 계통 분화 관련 유전자의 전사를 조절함으로써 배반포 품질을 향상시킬 수 있음을 확인했다.
CHAPTER Ⅳ에서는 돼지 만능성줄기세포를 활용한 신경 분화 과정에 있어서 NT-4는 신경의 성숙을 촉진한다는 것을 확인했다. 먼저, 유전자 비통합적인 센다이 바이러스 접근법을 사용하여 돼지 유도만능줄기세포를 수립했다. 총 10개의 돼지 유도만능줄기세포주가 수립되었으며, 이를 유지하기 위한 최적의 배양 조건은 백혈병 억제 인자 (Parmar et al.)에 의존함을 확인했다. 수립된 돼지 유도만능줄기세포는 강한 알칼리성 인산분해효소 활성과 배아체 형성능을 보였고, 정상 핵형을 가지고 있었다. 또한, 면역형광 분석을 통해 수립된 돼지 유도만능줄기세포는 세포 내 만능성 마커인 OCT4, SOX2 및 NANOG와 세포 외 만능성 줄기 세포의 표면 마커인 SSEA4에 대해 양성인 것으로 나타났다. 돼지 유도만능줄기세포의 특성 분석 이후, 신경 유도는 SB431542 (TGF-β 억제제) 및 LDN193189 (BMP4 억제제)를 사용한 이중 SMAD 억제 (dSMADi) 방법을 사용해 진행되었다. 배측 중뇌 영역의 패터닝은 SHH 및 FGF8과 같은 인자를 사용하여 촉진되었다. 뉴런으로 추정되는 세포는 신경 유도 및 분화 후 17일째에 미성숙뉴런의 표지자인 TUJ1 및 성숙뉴런의 표지자인 MAP2와 같은 신경세포 특이적인 마커의 발현을 나타냈다. 흥미롭게도 분화 시, NT-4를 처리한 군에서 성숙한 뉴런의 비율이 대조군보다 유의하게 높았다. 또한, 신경 분화 17일째에 중뇌 도파민 뉴런의 특이적인 마커인 TH와 OTX2를 발현하는 뉴런도 확인했다. 추가적인 연구가 반드시 필요하지만 이 연구를 통해 돼지 유도만능줄기세포에서 유사 도파민 뉴런을 생성하기 위한 효율적인 플랫폼을 개발할 수 있었다.
최종적으로, 본 연구는 돼지의 초기 배아 발생 및 신경 발달뿐만 아니라 난모세포의 체외성숙에도 NT-4가 유익한 역할을 할 수 있다는 것에 대한 통찰력을 제공할 수 있다. NT-4는 돼지 난모세포의 체외성숙 동안 난구-난모세포 복합체에서 ERK1/2 신호 전달 경로를 조절함으로써 난모세포의 체외성숙을 촉진하고 난모세포의 품질 개선에 기여한다. 또한, NT-4는 착상 전 배아 발달 동안 영양외배엽 계통 분화에 관여한다. 마지막으로, 뇌 유래 신경영양인자, 신경교 세포주 유래 신경영양인자 및 NT-4와 같은 다양한 신경영양인자의 조합을 사용하여 돼지 유도만능줄기세포주를 유사 중뇌 도파민 뉴런으로 분화하는 새로운 방법을 제시했다. 따라서, 본 연구의 결과는 돼지의 배아 발달 및 초기 신경 분화에 대한 이해를 높이고 임상 적용의 기초 학문 자료를 제공하였다.
Neurotrophic factors play important roles in both neural and non-neural tissues. The mammalian neurotrophin family consists of four members: nerve growth factor, brain-derived neurotrophic factor, neurotrophin-3, and neurotrophin-4 (NT-4). NT-4 is a growth factor involved in follicle development, oocyte maturation, as well as neuronal growth and development through the interaction between high-affinity tropomyosin receptor kinase B (TrkB) and low-affinity pan-neurotrophin receptor (p75NTR). Since both the brain and ovary are highly innervated organs, various neurotrophic factors can promote nerve growth and improve neural development during neural and ovarian development. Therefore, it is important and valuable to determine how neurotrophic factors affect neural and ovarian development. Although porcine reproductive biology and translational research have been explored over the past several decades, there are not enough studies on the effects of neurotrophic factors on ovarian physiology and neural development in pigs. Therefore, this study aimed to investigate the roles of NT-4 on embryonic development and neural differentiation in pigs.
In the first study, NT-4 supplementation during in vitro maturation (IVM) enhanced nuclear and cytoplasmic maturation of porcine oocytes. This study involved the identification and localization of NT-4, TrkB, and p75NTR in porcine ovaries. Furthermore, NT-4 supplementation during IVM improved the developmental potency of parthenogenetic activation (PA)-derived porcine embryos. Therefore, NT-4 is required for follicle development and oocyte maturation in pigs.
In the second study, it was investigated the physiological roles of NT-4 during IVM of porcine cumulus-oocytes complexes (COCs). This study demonstrated the presence of NT-4, TrkB, and p75NTR in matured porcine COCs, suggesting that NT-4-related signaling pathways are involved in the maturation and development of porcine COCs. NT-4 promoted the maturation of COCs by upregulating NFKB1 transcript through the neurotrophin/p75NTR signaling pathway and enhanced oocyte quality by upregulating GDF9, BMP15, CD9, and DNMT3A transcripts. The level of phosphorylated extracellular signal-regulated kinase 1/2 (ERK1/2) was significantly higher in NT-4-treated cumulus cells than in untreated cells. Both total and phosphorylated ERK1/2 levels were significantly higher in NT-4-treated than in untreated oocytes. Moreover, NT-4 improved subsequent embryonic development after in vitro fertilization (IVF) and somatic cell nuclear transfer (SCNT). Collectively, NT-4 promoted oocyte maturation and cumulus expansion by regulating the ERK1/2 signaling pathway in porcine COCs during IVM.
In the third study, NT-4 supplementation during in vitro culture (IVC) improved the developmental potency of embryos and quality of blastocysts after PA. NT-4 may contribute to the development of a trophectoderm (TE) lineage rather than inner cell mass (ICM) in the porcine preimplantation embryonic development by upregulating CDX2, PPAG3, and GATA3 transcripts. In the analysis of blastocyst cell lineage specification, NT-4 supplementation significantly increased the number of cells expressing the TE marker CDX2 in PA-derived blastocysts compared with the control. In addition, NT-4 can regulate genes related to the Hippo-Yes-associated protein (Hippo-YAP) and mitogen-activated protein kinase (MAPK) pathways during porcine preimplantation embryonic development. This study, for the first time, showed that NT-4 supplementation improved blastocyst quality by regulating the transcription of trophectoderm lineage specification-related genes during porcine IVC after PA.
In the final study, it was investigated that NT-4 promoted neuronal maturation. Porcine induced pluripotent stem cells (iPSCs) from fibroblasts were generated by using an integration-free Sendai virus approach. A total of 10 porcine iPSC lines were established, and it was confirmed that the optimal culture conditions for maintaining porcine iPSCs were dependent on leukemia inhibitory factor (LIF). The porcine iPSCs showed strong alkaline phosphatase (AP) activities and embryoid body formation capacities with a normal karyotype. Furthermore, immunofluorescence analysis showed that porcine iPSCs were positive for the intracellular pluripotent markers OCT4, SOX2, and NANOG, as well as the extracellular pluripotent stem cell surface marker SSEA4. The neural induction proceeded with a dual SMAD inhibition (dSMADi) with SB431542 (TGF-β inhibitor) and LDN193189 (BMP4 inhibitor). The specification of the ventral midbrain region was promoted using regional factors, such as sonic hedgehog (SHH) and fibroblast growth factor 8 (FGF8). The putative midbrain dopaminergic (mDA) neurons showed the expression of neuronal markers, such as TUJ1 (immature neuron) and MAP2 (mature neuron), at day 17 after neural induction and differentiation. In addition, some neurons expressing TH and OTX2, specific markers of mDA neurons, were also identified at day 17 of neuronal differentiation. Interestingly, the proportion of mature neurons in the NT-4 treatment group was significantly higher than that in the control group during neural differentiation. Although further studies are required, this study developed an efficient platform for generating porcine putative neurons from porcine iPSCs.
In summary, the present study provided insight into the beneficial role of NT-4 in oocyte maturation as well as early embryonic and neural development in pigs. NT-4 promotes oocyte maturation and contributes to the improvement of oocyte quality by regulating the ERK1/2 signaling pathway in COCs during porcine IVM. NT-4 is also involved in the TE lineage specification during pre-implantation embryonic development. The final study presented a novel method for the differentiation of porcine iPSCs into putative mDA neurons using the combination of various neurotrophic factors, such as brain-derived neurotrophic factor, glial cell line-derived neurotrophic factor, and NT-4. Taken together, the findings of the present study enhance the understanding of embryonic and early neuronal development in pigs and provide the basis for clinical applications.
