인삼(Panax ginseng C. A. Meyer)은 한 포장에서 오랜 기간 재배되는 생육특성으로 병충해 발생이 많아 이를 방제하기 위하여 다양한 화학농약이 사용되고 있으며, 인삼의 안전성을 확보하기 위하여 농약잔류허용기준을 설정하여 관리하고 있다. 그러나 살포된 농약은 인삼뿐만 아니라 낙하하여 토양 중에 잔류되기도 하며, 이는 인삼으로 흡수·이행되어 최종적으로 소비자의 안전에 위해를 가할 가능성이 있다. 잔류농약으로부터 인삼의 안전성 확보를 위한 체계적인 관리를 위하여 지속적인 잔류농약모니터링 및 잔류특성 연구와 인삼 중 잔류농약의 효율적인 분석법을 개선 및 개발하고, 이를 적용하는 연구가 필요하다. 이에 본 연구에서는 안전성 검사 시 부적합 빈도가 높은 tolclofos-methyl 및 endosulfan을 선정하여 인삼재배지 토양 중 인삼으로의 잔류특성을 구명하고, 국내 주요 인삼재배지 토양 및 인삼(잎·줄기, 뿌리)을 채취하여 잔류농약을 분석함으로써 농약의 잔류 여부 및 양상 등을 파악해 국내산 인삼의 잔류농약에 대한 안전성 확보에 기여하고자 하였다. 또한 primary secondary amine (PSA) 정제과정을 포함한 QuEChERS-EN법과 MWCNTs (multi-wall carbon nanotubes)를 이용한 변형된 분석방법들의 비교 평가를 통해 유효성 및 타당성을 검토하여 인삼 중 모니터링법으로써의 적합 여부 및 개선방향을 제시하고자 수행하였다.
토양 및 인삼(잎·줄기, 뿌리) 중 tolclofos-methyl 및 endosulfan의 잔류특성을 구명하기 위하여 제품 일정량을 5.0 mg kg-1 수준으로 토양에 혼화처리한 후, wagner pot에 채운 뒤 묘삼을 정식하였으며, 2년 동안(2013년 4월∼2014년 12월) 분기별로 시료를 채취하여 GC-MS로 분석하였다. 본 연구결과, 토양 중 tolclofos-methyl 및 endosulfan이 잔류할 경우 인삼의 생육기간 동안 인삼(잎‧줄기, 뿌리)으로 이행됨을 확인하였고, 초기 잔류량 대비 2014년 12월에 채취한 토양 시료 중 0.46%의 tolclofos-methyl과 42.3%의 endosulfan이 잔류됨을 확인하였다. 동일 시점에 채취한 인삼 뿌리 시료 중 두 농약의 인삼으로 흡수·이행된 잔류량은 최대 잔류량 대비 tolclofos-methyl의 경우 7.8%, endosulfan은 24.9%가 잔류되었다. 본 연구결과를 통하여 tolclofos-methyl의 경우 농약안전사용기준을 준수하지 않고, 인삼의 생육기간 동안 관행적으로 2회, 3회 살포한다면 잔류허용기준을 초과할 우려가 있을 것으로 판단되었다. 또한 endosulfan의 경우 토양뿐만 아니라 인삼에서도 잔류성이 큰 것을 확인하였고, 농업현장에서 사용되어 검출되기 보다는 endosulfan 농약의 특성상 이전에 사용한 농약이 토양환경 중 오래 잔류하기 때문에 부적합 인삼을 발생시키는 것으로 사료된다.
잔류농약 안전성 검사 시 인삼 중 부적합 비율이 높은 tolclofos-methyl 및 endosulfan의 잔류특성 구명에 이어 홍천, 철원, 풍기, 금산 지역의 토양 및 인삼(잎·줄기, 뿌리) 중 잔류농약 모니터링을 수행하여 농약의 잔류양상을 파악하고자 하였다. 분석대상 성분은 GC-MS/MS 74성분 및 LC-MS/MS 172성분으로 총 246성분이었으며, QuEChERS방법을 이용하여 토양 및 인삼(잎·줄기, 뿌리) 시료를 분석하였다. 2013년 인삼재배지 토양 및 인삼(잎·줄기, 뿌리) 중 잔류농약 모니터링 수행결과, 총 50종의 농약이 0.01∼15.01 mg kg-1 수준으로 검출되었으며, 2014년도에는 총 23종의 농약이 0.01∼2.00 mg kg-1 수준으로 검출되었다. 2년 동안 동일한 지역의 토양 및 인삼(잎·줄기, 뿌리) 중 잔류농약을 분석한 결과, 검출된 성분은 매우 다른 경향을 보였으며, 농가에서 실제 사용하였다고 한 농약의 성분과는 차이가 있었다. 이는 인삼재배 예정지로 밭을 관리하였더라도 전 면적의 토양을 분석·검증할 수 없어 결과에 차이를 보인 것으로 판단되며, 바람, 관수 등에 의하여 주변농가에서 사용한 농약이 영향을 주어 실제 사용 농약과 다른 성분이 검출된 경우도 배제할 수 없을 것으로 사료된다. 인삼에 검출된 농약 성분은 대부분 살균제로 사용이 많은 농약들이 검출되었다. 본 연구결과를 통하여 주요 인삼재배 지역의 토양 및 인삼 중 잔류농약의 양상을 파악하였으며, 지속적인 연구를 통해 인삼재배지 토양 및 인삼에 대한 관리영역의 확대가 필요할 것으로 사료된다.
지속적인 잔류농약 모니터링 및 안전성 검사를 수행함과 더불어 인삼 중 잔류농약을 효율적으로 분석할 수 있는 최적의 분석법을 개선 및 확립하는 연구도 병행되어야 한다. 이에 QuEChERS-EN법과 MWCNTs를 이용한 변형된 분석법들의 비교 평가를 통하여 인삼 중 모니터링법으로써의 적합 여부 및 개선방향을 제시하고자 수행하였다. 본 연구의 분석대상 농약은 총 335성분, 323품목으로 GC-MS/MS 분석성분 123성분(114품목)과 LC-MS/MS 분석성분 212성분(209품목)을 선정하였다. QuEChERS-EN법과 MWCNTs (5 mg, 10 mg, 15 mg, 20 mg) 및 m-PFC를 활용한 분석방법을 이용하여 직선성(linearity), matrix effect (ME%), 검출한계(LOD) 및 정량한계(LOQ), 회수율에 대한 유효성을 검증하였다. 각 분석방법들의 직선성을 비교 평가한 결과, 분석성분에 대하여 대부분 0.97 이상의 값을 나타내어 전반적으로 안정적인 직선성을 확인하였으며, matrix effect (ME%)를 산출하여 매질의 개선효과를 판단하였을 경우 GC-MS/MS 분석 시 MWCNTs 20 mg을 사용하였을 때 가장 우수한 효과를 확인할 수 있었다. 반면 LC-MS/MS 분석 시 QuEChERS-EN법에서의 매질의 개선효과가 더 높았으나 MWCNTs를 이용한 분석방법과 크게 차이나지 않았으며, 각 분석방법들의 검출한계(LOD) 및 정량한계(LOQ)를 비교 평가한 결과, 10 μg kg-1 이내의 범위를 대부분 만족하였다. GC-MS/MS 및 LC-MS/MS를 이용한 회수율시험 결과, 각 분석방법들에 대하여 76∼97%의 성분들이 70∼130% 및 RSD 30% 이하의 범위를 만족함을 확인할 수 있었다. 본 연구결과, MWCNTs 5 mg 및 m-PFC를 활용한 분석방법이 개선효과가 가장 뚜렷한 것으로 나타나 인삼에 적용할 수 있는 새로운 유형의 d-SPE 흡착제임을 입증하였다. 이에 MWCNTs를 이용한 개선 분석방법이 인삼 중 모니터링법으로써 적합한 것으로 사료되었으며, 인삼뿐만 아니라 다양한 농산물에도 적용 가능할 것으로 판단된다.
인삼재배지 토양 및 인삼 중 잔류농약 안전관리 연구결과, 인삼 경작지에 오래 잔류되어 인삼에 영향을 줄 수 있는 농약에 대한 지속적인 모니터링 및 잔류특성 연구의 필요성을 시사하였다. 또한 부적합 인삼의 발생을 줄이기 위하여 인삼재배지 토양 및 인삼재배 예정 지역의 토양에 대한 잔류농약 모니터링 연구뿐만 아니라 인삼 중 잔류농약의 효율적인 분석을 위하여 최적의 분석방법을 확립하는 연구도 이루어져야 할 것이다. 본 연구결과를 통하여 농업인들이 좋은 품질의 인삼을 안정적으로 재배·생산할 수 있는 안전체계를 구축할 수 있을 것으로 사료되며, 우리나라 인삼의 국제적 경쟁력이 강화됨과 동시에 인삼 생산 및 수출 활성화에 기여할 수 있는 방안의 밑거름이 될 것으로 기대된다.
The ginseng (Panax Ginseng C. A. Meyer) that usually grows in a field for 4 to 6 years tends to get often damaged by disease and insect pest. Therefore, the application of various chemical pesticides in the cultivation of ginseng would be unavoidable. In this situation, the Pesticide MRLs (maximum residue limits) is established and imposes limitations to secure the safety. However, the sprayed pesticides can remain onto ginseng as well as into the soil, being fallen down on the ground, which could be translocated into the roots of ginseng. In the end, it could be harmful to consumer’s safety.
This study is designed (Ⅰ) to identify the residual characteristics of tolclofos-methyl and endosulfan remaining in ginseng and in soil, which were confirmed through the analysis of ginseng (leaf·stem and root) that was collected quarterly in accordance with a growth phase of the seedling of ginseng. This study also aims (Ⅱ) to monitor residual pesticides in soil and ginseng (leaf·stem and root) from major ginseng farms, and (Ⅲ) to improve and develop efficient monitoring methods in cultivating ginseng. This study also attempts to draw a comparison between the MWCNTs (multi-wall carbon nanotubes) method and the QuEChERS-EN method, and contributes to improving direction and appropriate methods of monitoring in cultivating ginseng.
Ginseng was transplanted in pots and around 5.0 mg kg-1 of tolclofos-methyl and endosulfan were incorporated into soil. The samples (soil, leaf·stem and root of ginseng) were collected during April, June, September, and December over two years (2013-2014), and the pesticides residues in the samples were analyzed at the end of each month, using GC-MS. The results of this study shows that when tolclofos-methyl and endosulfan remained in soil were transferred to ginseng (leaf·stem, root) during the growth duration. It can be also seen that as for the samples of soil which were collected in December 2014, the amount of tolclofos-methyl and endosulfan remained 0.46% and 42.3% respectively. On the other hand, as for the samples of the roots of ginseng that were collected in the same period, the amount of tolclofos-methyl and endosulfan analyzed 7.8% and 24.9% respectively, based on the maximum residual amount. From the results, it can be indicated that if the tolclofos-methyl is sprayed twice or three times during the growth period of ginseng, and the safe use guideline is not observed, it may exceed the MRL. In the case of endosulfan, it was considered that it is highly likely to remain in soil as well as in ginseng. However, this remaining endosulfan pesticide is not detected only in the fields where it is used. Rather, it would be better to consider that because the previously used endosulfan pesticide tends to remain in soil for a long time, it could lead to inadequate ginseng.
To identify the residual patterns of pesticides, investigated were vegetative part of ginseng (leaf·stem, root) and soil samples that were collected from major ginseng cultivation regions in Korea, such as Hongcheon, Cheorwon, Punggi, and Geumsan. A total of 246 components was analyzed, by using the QuEChERS methods. 74 components of them were by the GC-MS/MS, and the other 172 components were by the LC-MS/MS. The outcome of monitoring of residual pesticides in soil and in vegetative part of ginseng revealed that 50 different kinds of pesticides were detected as 0.01∼15.01 mg kg-1 in season 2013, whereas 23 kinds of pesticides were detected as 0.01∼2.00 mg kg-1 in season 2014. The 2-year-long analysis of pesticide residues both in vegetative part of ginseng and in soil shows a quite different tendency, and actually the detected ingredients of pesticides were different from ones that was claimed to had used in the field.
This study was conducted to verify the suitability of monitoring methods for ginseng cultivation by comparing the QuEChERS-EN method and the modified method of MWCNTs. 123 components (from 114 items) were analyzed by the GC-MS/MS, and 212 components (from 209 items) were analyzed by the LC-MS/MS among a total of 335 and 323 pesticides, all of which were analyzed in this study. The validity of each method for linearity, matrix effect (ME%), the limit of detection (LOD), the limit of quantitative (LOQ), and recovery were investigated. As for the results of this study regarding the validation of linearity, it indicates that most of the analytical components showed a value of 0.97 or more, and excellent linearity values in GC-MS/MS and LC-MS/MS analysis. The effect of improvement was evaluated by the matrix effect (ME%), and the best effect was obtained when it used 20 mg of MWCNTs in GC-MS/MS analysis, while in the LC-MS/MS analysis confirmed improvement on the matrix effect in the QuEChERS-EN method, but there were not many significant differences from the MWCNTs analysis methods. The LOD and LOQ its values were confirmed within the range of 10 μg kg-1. As a result of the recovery tests for all methods using GC-MS/MS and LC-MS/MS, it was found that 76% to 97% of the component satisfied the effective recovery 70% to 130%, and less than 30% RSD. The results of this study illustrate that the analytical methods using 5 mg of MWCNTs and mPFC were the most effective to improve, which assures that they are the new types of d-SPE adsorbent that is applicable to ginseng as well as other various agricultural products.
To conclude, the result of the research on the safety management for residual pesticides in ginseng and in soil, suggests the need of continuous monitoring and studying of characterization of residual pesticides that may remain in soil for a long time. Furthermore, this study also implies that it is necessary to continue establishing effective methods to analyze the residual pesticides in ginseng. Finally, from the outcome, this study expects to contribute to establishing a safety management system in which farmers can produce good quality ginseng in a suitable environment, and consequently strengthening the international competitiveness of Korean ginseng, and vitalizing the production and the export of Korean ginseng in the future.
