Humoral Immune Response of the Squirrel Monkey after Immunization with Porphyromonas gingivalis and Prevotella intermedia

The colonization of the subgingival area by Porphyromonas gingivalis and Prevotella intermedia in the squirrel monkey has been inhibited by systemic immunization with whole P. gingivalis and P. intermedia. This paper reports the results of Western blot experiments directed towards identification of the predominant P. gingivalis and P. intermedia immunogens for the squirrel monkey. Hyperimmune sera associated with the delayed and diminished colonization of the gingival crevicular area by P. gingivalis and P. intermedia developed a large number of bands. Major antigenic banding was observed at 40 kDa for P. gingivalis: more complicated pattern of bands ranging from 30 kDa and 95 kDa were observed in P. intermedia. Sodium metaperiodate treatment of P. intermedia antigens revealed predominate protein antigens with MW 53 kDa and 31 kDa. Sera from animals that were naturally infected with P. gingivalis and P. intermedia and the sham-immunized animals after ligation and inoculation with P. gingivalis and P. intermedia all developed some of the same major bands as did the hyperimmunized animal sera. The predominate P. gingivalis and P. intermedia antigens were immunogenic by active immunization as well as in natural infection. This fact allows a comparison of the antibody functions and the effect of antibody quantity on the protective potential of specific antibodies after active immunization and natural infection. The resemblance between immune responsiveness of the squirrel monkey and human to similar P. gingivalis antigens further supports the squirrel monkey as a relevant model to study immune modulation of periodontal pathogen colonization.
