Influenza is a major problem in both humans and avian species. Annual epidemics of seasonal influenza in humans are estimated to result in about 3 to 5 million cases of severe illness, and about 290,000 to 650,000 deaths worldwide. Also, several hundreds and thousands of poultry are being mass culled after disease outbreak. It has been known that avian influenza overcome species barrier, infect and even induce death in humans. To prevent this, constant monitoring of viruses and development of effective vaccine should be conducted.
Besides vaccine antigen development, research on effective delivery method and adjuvant for vaccine should be carried out. As an effective method of delivering vaccine, it is known that intradermal injection such as micro-needle is more effective than intramuscular injection which is commonly used as a main inoculation route of inactivated vaccine, because intradermal injection could stimulate rich antigen presenting cells of dermis and epidermis. However, the adjuvant for enhancing the effect of the vaccine in the intramuscular inoculation method is well studied, whereas adjuvant for intradermal inoculation method needs to be studied. Because the adjuvant used for the intramuscular inoculation could induce side effect when administered in intradermal injection, there is an urgent need to develop additional adjuvant for intradermal injection.
In chapter I, pathogenicity and transmissibility of newly isolated H5N6 highly pathogenic avian influenza virus (HPAIV) in Korea were confirmed in ferret model, which is an animal model similar to human respiratory system for influenza study. The virus used in this study was confirmed that it did not propagate well in the ferret with no transmissibility, and also showed no resistance gene against the antiviral drug.
In chapter II, bivalent virus-like particle (VLP) vaccine that is composed of the hemagglutinin (HA) and matrix 1 (M1) proteins of the H5N1 HPAIV and fusion (F) protein of Newcastle disease virus (NDV) which cause high mortality in chickens similar to HPAI was developed. Cytoplasmic and transmembrane domains of F protein were changed to those of HPAIV HA protein to effectively fuse with the matrix protein. A single immunization of chickens with the recombinant VLP vaccine induced high levels of hemagglutination inhibition (HI) antibody titers against H5N1 HPAI virus and anti-NDV antibody detected in ELISA and protected chickens against subsequent lethal HPAIV and NDV infections and reduced viral shedding. Furthermore, DIVA tests using the commercial NP-cELISA tests against HPAIV and HI assay against NDV were easily performed. These results strongly suggest that utilization of chimeric VLP vaccine in poultry species may be a promising strategy for the better control of HPAI and ND simultaneously and reduce the cost of the recombinant protein vaccine.
In chapter III, effective adjuvants for the intradermal vaccination by using microneedles were investigated. In order to increase the vaccine efficacy and reduce the vaccine dose, there is a urgent need to find some adjuvants for the microneedle vaccination. Zymosan, which is the cell wall preparation of Saccharomyces cerevisiae as TLR2 and dectin-1 agonist, or poly (I:C) as TLR3 agonist, was coated on a microneedle with inactivated influenza virus, and immunized into BALB/c mouse to determine the immunogenicity including synergetic effect between two adjuvant and protectivity. As a result, the group injected with zymosan and vaccine antigen showed significantly stronger humoral response including HI titer, IgG and IgG subtype responses without any adverse effects, compared to the group immunized with the vaccine antigen alone. Also, cellular immune responses were also enhanced in the group received adjuvant with vaccine antigen. In addition, they showed superior protection and reduced lung viral dose after lethal viral challenge. This study confirmed that zymosan could be used as an immunostimulant for microneedle vaccination for influenza.
Taken together, in order to make better response to influenza outbreak, it is necessary to constantly monitor newly emerging viruses and develop high quality influenza vaccine through vaccine antigen development as well as effective delivery method with adequate adjuvant. By characterizing each virus, strains that vaccines are most urgently needed could be determined. VLP techology could be successfully adopted to highly immunogenic antigen development of those strains with simple serological test for DIVA and microneedle vaccination with zymosan could induce stronger immune response without any side-effect. These results strongly suggested that development and advancement of influenza vaccine may help of the better control of influenza.
인플루엔자는 사람 및 조류에서 큰 문제가 되고 있는 질병이다. 사람의 경우 세계적으로 매년 3-5백만명이 계절 독감에 감염되어 29-65만명의 사망자를 내는 것으로 추정되고 있다. 조류인플루엔자의 경우 발생 시 많은 가금류가 살처분되고 있으며, 이외에도 종간 장벽을 넘어 사람으로 전파되어 감염 및 사망하는 사례가 발생하고 있다. 이를 예방하기 위해서는 바이러스에 대한 꾸준한 모니터링과 함께 바이러스를 효율적으로 방어할 수 있도록 백신이 개발 및 고도화되어야 한다.
인플루엔자에 대해 효과적인 백신 개발을 위해서는 백신 항원 제작 이외에도 백신에 대한 효과적인 전달 방법 및 어쥬번트 개선에 대한 연구가 진행되어야 한다. 백신의 효과적인 전달법으로는 불활화 백신의 주요 접종경로로 사용되는 근육 내 접종법에 비해 마이크로니들과 같이 항원 제시 세포가 많은 피내에 백신을 접종하는 것이 효과가 좋은 것으로 알려져 있다. 다만 근육내 접종법의 경우 백신의 효과를 강화시키기 위한 어쥬번트에 대한 연구가 잘 되어 있는 반면, 마이크로니들과 같은 피내 접종법의 경우 근육 접종 시 사용하는 어쥬번트의 부작용으로 인해 추가적인 어쥬번트의 개발이 필요한 상황이다.
본 논문의 제 1장에서는 국내에서 2016년에 신규 분리된 H5N6 바이러스에 대해 사람으로의 전파 가능성을 측정하기 위한 동물 모델인 페럿 모델에서의 병원성 및 전파능을 확인하였다. 실험에 사용된 바이러스의 경우 페럿에서 바이러스가 잘 증식하지 못하고 전파능이 없는 것이 확인되었으며, 항바이러스제에 대한 내성도 관찰되지 않았다.
본 논문의 제 2장에서는 H5N1 고병원성 조류인플루엔자 바이러스의 HA 및 M1 단백질과 고병원성 조류인플루엔자와 유사하게 조류에 큰 피해를 일으키는 뉴캣슬병의 F 단백질에 대한 항원을 동시 발현시킨 바이러스 유사 입자 백신을 개발하였다. F 단백질의 경우 고병원성 조류인플루엔자의 M 단백질과 효과적으로 결합할 수 있도록 세포막 부위 및 세포내 부위를 고병원성 조류인플루엔자의 HA 단백질로 치환하였다. 개발된 바이러스 유사 입자 백신은 1회 접종만으로도 높은 수준의 고병원성 조류인플루엔자에 대한 HI 항체 역가 및 ND에 대한 ELISA 항체 역가 유도에 성공하였으며, 각각 고병원성 조류인플루엔자 및 뉴캣슬병 바이러스를 공격접종하였을 때도 방어능을 나타내었으며 바이러스 배출을 줄였다. 또한 시중에 판매되어 쉽게 구할 수 있는 인플루엔자 NP-cELISA 키트와 뉴캣슬병 진단에 흔히 사용되는 HI 항체 검출법을 이용하여, 백신에 포함되지 않은 고병원성 조류인플루엔자 NP 단백질 및 뉴캣슬병 HN 단백질에 대한 항체 생성 여부를 관찰하여 백신 개체와 백신 후 감염 개체의 구분에 성공하였다. 개발된 백신의 경우 높은 효과와 함께 DIVA가 적용 가능하여 고병원성 조류인플루엔자 및 뉴캣슬병 방역 정책에 효율적으로 사용될 수 있으며, 2가지 질병에 대해 동시에 백신을 실시함으로써 조류 차세대 백신에 문제점이 되고 있는 가격적인 측면을 낮출 수 있어 백신으로 활용될 수 있는 가능성이 높다.
본 논문의 제 3장에서는 마이크로니들을 사용한 백신의 피내 접종시 효과적인 어쥬번트를 탐색하였다. 마이크로니들 백신 시 백신의 효과를 높이고 항원량을 절감하기 위해 어쥬번트 개발에 대한 필요성이 높은 상태이다. TLR2 및 dectin-1 agonist로 알려진 Saccharomyces cerevisiae 세포벽 유래 물질인 zymosan과 TLR3 agonist로 알려진 poly (I:C)를 각각 또는 함께 인플루엔자 불활화 항원과 마이크로니들에 코팅하여 BALB/c 마우스에 접종 후 면역원성, 방어능 및 두 어쥬번트의 상승 효과를 관찰하였다. 실험 결과, zymosan과 인플루엔자 불활화 항원을 함께 접종한 그룹에서는 불활화 항원 단독 접종 그룹에 비해 부작용 없이 유의적으로 강한 IgG 반응, HI 역가 상승이 확인되었다. 또한 세포성 면역에 대해서도 항원 단독 그룹에 비해 강한 면역 반응을 나타내었다. 이에 따라 바이러스 공격 접종 이후에도 적은 체중 감소 효과와 함께 높은 방어능을 나타냈으며, 폐내 바이러스 양 측정 결과에서도 바이러스 증식 억제 효과가 관찰되었다. 종합적피내로 전달한 결과, 항원 단독을 전달했을 때와 마찬가지로 부작용이 나타나지 않았다. 결과적으로 zymosan은 마이크로니들을 통한 인플루엔자 백신에 효과적인 면역증강제로 사용될 수 있음이 확인되었다.
