Rice bran oil (RBO) was fractionated with acetone at a low temperature (-16℃) and the yield of fractionated solid (S-RBO) and liquid (L-RBO) phase were 66.5 and 33.5%, respectively. The L-RBO contained more unsaturated fatty acids (81.8%) than S-RBO (76.1%). Response surface methodology (RSM) was used to determine the effects of three variables (water activity of lipase, reaction temperature and time) on the lipase-catalyzed incorporation of CLA into L-RBO. We used CLA as the acyl due to its purported health benefits. The transesterified lipid (TL) contained palmitic (13.5%), oleic (37.5%), linoleic (26.6%) and conjugated lioleic acid (CLA) isomers (20.7%), respectively.
According to ridge analysis, optimal reaction conditions for water activity of lipase, reaction temperature, and time were 0.177, 55.66 ℃, and 29.25 h, respectively for producing TL with maximum incorporation of CLA. The coefficient of determination (R2 = 0.94) showed that the fitted model explained 94% of the observation. The TL was reproduced to confirm the experimental incorporation with the estimated value. The data showed that the experimental response was reasonably close to the estimated response.
Solid fat stock was produced from the fractionated rice bran oil (solid phase, S-RBO) and palm stearin (PS) through lipase-catalyzed reaction, in which conjugated linoleic acid (CLA) was intentionally incorporated. For optimizing the reaction, response surface methodology (RSM) was employed with four reaction variables such as water activity, reaction temperature, reaction time and mole ratio of S-RBO to PS. The predictive model was adequate due to no significant lack of fit and satisfactory level of coefficient of determination (R2=0.95). The melting point of solid fat stock was affected by reaction time and substrate mole ratio, whereas water activity and reaction temperature had no significant effect. Based on ridge analysis, the combination of Aw (X1; 0.32), reaction temperature (X2; 65.3 ℃), reaction time (X3; 28.9 h) and substrate mole ratio (X4; 1:1.1) was optimized for producing solid fat stock with target melting point of 43.8 ℃. The solid fat stock (SFS) contained 39.9% palmitic, 31.3% oleic, 13.2% linoleic acid, and 10.9% CLA isomers. Solid fat contents were 23.4, 10.9 and 2.5% at 20, 30 and 40 ℃, respectively.
Mustard oil (MO) was fractionated at low temperature with acetone. Among all fractions obtained from MO, -24L, i.e., the liquid fraction obtained at -24 °C, showed higher levels of oleic (24.7%) and linoleic (22.0%) acids, and lower level of erucic acid (24.5%) compared to other fractions. The results demonstrated that the lowest temperature (-24 °C) was most effective for lower levels of erucic acid through crystallization of triacylglycerol (TAG) molecules. From the economic point of view and reaction efficiency, the ideal mole ratio was 1:3 for all cases (32.9, 35.2, 37.9, 38.2 and 34.8%, respectively) when observed the effect of mole ratio on the incorporation of capric acid (CA) into fractions. The content of erucic acid (24.3, 28.3, 27.0, 27.6 and 19.0%, respectively) was found reasonable lower level for 24 h reaction time of all cases. One purpose of this study was to obtain erucic acid reduced fractions because erucic acid has undesirable effect on human health. The result suggested that erucic acid could be attained in the solid fraction rather than the liquid fraction and better reduction of erucic acid would be obtained at lower temperature.
Low-trans blend (LTB) was produced from the fractionated mustard oil (solid phase, S-MO) and palm stearin (PS) through lipase-catalyzed reaction, in which linoleic acid (LA) was intentionally incorporated. For optimizing the reaction condition, response surface methodology (RSM) was employed with three reaction variables such as substrate mole ratio of S-MO to PS (X1), reaction temperature (X2) and reaction time (X3). The predictive models were adequate and reproducible due to no significant lack of fit and the P-value of the model was very small (0.0037) for melting point and (0.0132) for ω6/ω3 ratio, and satisfactory level of coefficient of determination (R2 = 0.92) for melting point and (R2 = 0.89) for ω6/ω3 ratio. The melting point of LTB was affected by substrate mole ratio, whereas reaction temperature and reaction time had no significant effect. The ω6/ω3 ratio of LTB was affected by substrate mole ratio and reaction temperature but reaction time had no significant effect. Based on ridge analysis, the combination of 1:2 (X1), 65.17 ℃ (X2) and 21.46 h (X3) was optimized for producing low-trans blend with target melting point of 34.57 ℃. For considering the ω6/ω3 ratio, the optimum condition found 1:1.7 substrate mole ratio, 61.42 ℃ reaction temperature and 25.85 h reaction time. The LTB for melting point contained mainly 37.8% palmitic, 24.8% linoleic, 21.3% oleic and 9.7% erucic acid. Solid fat contents were 30.3 and 10.3% at 20 and 30 ℃, respectively. The results suggested that RSM can be used to optimize the lipase-catalyzed production of low-trans blend with specific melting point.
쌀겨기름(RBO)을 -16 °C의 저온에서 아세톤으로 분별한 결과, 고형 쌀겨기름(S-RBO)과 액체 쌀겨기름(L-RBO)의 수율은 각각 66.5%, 33.5% 이었다. L-RBO는 불포화 지방산(81.8%)을 S-RBO (76.1%)보다 더 많이 함유하였다. 공액 리놀레산(CLA)을 L-RBO에 리파아제 촉매 결합시키는데 대한 3가지 변수(리파아제의 수분활성도, 반응 온도 및 시간)의 영향을 조사하기 위하여 response surface methodology (RSM)을 이용하였다. CLA가 인체에 대한 기능성 때문에 CLA를 아실기로 사용하였다. 재구성 지질 (transesterified structured lipid, SL)은 팔미트산 13.5%, 올레산 37.5%, 리놀레산 26.6%, 공액 리놀레산(CLA) 20.7%를 함유하였다. Ridge 해석 결과, CLA와 최대한 결합한 SL을 생산하기 위한 최적 반응조건은 리파아제 수분활성도 0.177, 반응온도 55.66 °C, 반응시간 29.25 시간이었다. 상관계수 R2=0.94는 예측모델이 측정값과 94% 일치함을 의미한다. CLA 실험 결과와 예측 값을 확인하기 위하여 SL을 반복 생산하였다. 데이터는 실험 값이 예측 값과 합리적 수준으로 근사함을 보였다.
공액 리놀레산을 의도적으로 혼합한 리파아제 촉매 반응에 의하여 분류 쌀겨기름의 고체상(S-RBO)과 팜 스테아린(PS)으로부터 고형 지방을 생산하였다. 반응을 최적화하기 위하여 4가지 변수인 수분활성도, 반응온도, 반응시간, PS 대 S-RBO의 몰비에 대하여 RSM을 이용하였다. 고형지방층의 녹는점은 반응시간과 기질의 몰비에 따라 영향을 받는 반면, 수분활성도와 반응시간은 큰 영향을 미치지 않았다. Ridge 해석법으로 43.8 °C의 목표 녹는점을 가진 고형 지질을 생산하기 위한 조건을 Aw(X1; 0.32), 반응온도 (X2; 65.3 °C), 반응시간 (X3; 28.9시간), 기질 몰비 (X4; 1:1.1)로 최적화하였다. 합성된 SL인 고형지질은 팔미트산 39.9%, 올레산 31.3%, 리놀레산 13.2%, CLA 이성체 10.9%를 함유하였다. 고형 지방 함량은 20, 30, 40 °C에서 각각 23.4, 10.9, 2.5 %이었다.
겨자유 (MO)는 저온에서 아세톤으로 분류하였다. 겨자유로부터의 분별물 중에서 -24 °C에서 얻은 액상이 다른 분별물에 비해서 올레산(24.7%)과 리놀레산(22.0%)의 함량이 높고, erucic acid (24.5%)의 함량이 낮았다. Triacylglycerol (TAG) 분자의 결정화로 erucic acid의 함량을 낮추는데 가장 낮은 온도 -24 °C가 가장 효과적이었다. 경제성과 반응효율의 관점에서는 카프르산을 분별물에 결합 시 몰비 영향을 고려할 때, 모든 경우 (32.9, 35.2, 37.9, 38.2, 34.8 %)에 대하여 이상적인 몰비는 1:3 이었다. Erucic acid 함량 (24.3, 28.3, 27.0, 27.6, 19.0%)은 모든 경우에 대하여 24시간 반응 시 낮은 수준임을 확인하였다. Erucic acid가 인체에 부정적인 영향을 미치기 때문에 본 연구의 목적 중 하나는 erucic acid 함량을 낮추는 것이었다. 결론적으로 erucic acid는 액상보다는 고형분별물에 분포하고 아세톤을 이용한 저온분별에서 감소시킬 수 있었다.
리놀레산을 의도적으로 혼합한 리파아제 촉매반응에 의하여 분별 겨자유 (고체상, S-MO)와 팜 스테아린으로부터 low-trans 고체지를 생산하였다. 반응조건을 최적화하기 위하여 S-MO 대 PS의 기질 몰비 (X1), 반응온도 (X2), 반응시간 (X3)와 같은 3개의 반응변수에 대하여 RSM을 적용하였다. 예측모델은 재현성이 우수하였으며 모델의 p 값은 녹는점이 0.0037, ω6/ω3이 0.0132로 매우 낮고 상관계수는 녹는점이 R2=0.92, ω6/ω3이, R2=0.89로 만족스러운 수준이었다. 반응온도와 반응시간은 큰 영향이 없는 반면, low-trans 고체지의 녹는점은 기질 몰비에 따라 영향을 받았다. Ridge 해석법을 이용함으로써 34.57 °C의 목표 녹는점에서 low-trans 고체지를 생산하기 위한 조건을 1:2 (X1), 65.17 °C (X2), 21.46시간 (X3)으로 최적화하였다. 오메가 지방산(ω6/ω3 )비를 고려 시, 최적조건은 기질 몰비 1:1.7, 반응온도 61.42 °C, 반응시간 25.85시간 이었다. low-trans 고체지는 녹는점에서 팔미트산 37.8%, 리놀레산 24.8%, 올레산 21.3%, erucic acid 9.7%를 함유하였다. 고형지 함유량은 20 °C에서 30.3%, 30 °C에서 10.3% 이었다. 결론적으로 특정 녹는점에서 리파아제 촉매 반응에 의한 low-trans 고체지 생산을 최적화하기 위하여 RSM을 효과적으로 이용할 수 있었다.
