The present study was carried out to investigate the role of P.acnes on the evolution of non-inflamed comedon into inflamed lesion Anti-P.acnes antibody titers in the sera of patients who have severely inflamed acne lesions were checked using indirect immunofluorescence test, immunoelectrophoresis and dot blot immunoassay. The sera of the acne patients which showed high titers of anti-P.acnes antibody were incubated with cytosolic antigens and cell wall antigens of P.acnes. And then generation of superoxide anion by-normal human neutrophils and chemotactic effects were assessed.
The results were as follows:
1) In immunofluorescence test, anti-P.acnes antibody titers in sera were significantly increased in the acne patients.
2) Using immunoelectrophoresis, cytosolic antigens, cell wall antigens and culture supernatnat of P.acnes were reacted with acne patients' sera. Precipitating lines to cytosolic antigens, cell wall antigens and culture supernatant were differnt from each other.
3) In dot blot immunoassay. anti-P.acnes antibody titers of the acne patients were much higher than of the healthy normal controls.
4) The sera of 22 patients with severely inflamed acne lesion and 15 healthy controls without history of acnes lesion were incubated with cytosolic and cell wall antigens of P.acnes. Superoxide anion generation by normal human neutrophils were significantly increased in the acne patients. There was dose-related increment of superoxide anion generation. In comparison to cytosolic antigens, cell wall carbohydrate antigens were more stimulatory to neutrophils.
5) The sera of 15 acne patients were incubated with cytosolic antigens, cell wall carbohydrate antigens and culture supernatants. And It's effects on neutrophil chemotaxis were assessed. Cell wall antigens had most significant effect on chemotaxis in comparison to cytosolic antigens and culture supernatant.
With above results, in the presence of high levels of ant-P.acnes antibody, P.acnes is a greater stimulus for the induction of neutrophil chemotaxis and superoxide anion generation.
