This experiment was performed in an attempt to explore the stimulating effect of cultural filtrate, which was produced by Penicillium sp. No. 2 isolated from contaminants, upon the mycelial growth and sclerotial production of Sclerofium rolfsii Sacc, from Magnolia kobus. Mycelial growth and sclerotial production of the fungus were estimated by dry weight of mycelia and sclerotia grown or formed on the basal medium, which was a glucose-asparagine solution or plate, modified with NH4NO3 as a nitrogen source and amended with cultural filtrate, of Penicillium sp. or its fraction.
The experiment results are as follows:
1. The stimulating effect on the mycelial growth of fungus examined was shown in the cultural filtrate of Penicillium sp. It increased linearly with increase of filtrate concentrations to the degree of 6-15ml/50ml basal medium solution.
2. NH4NO3 as a nitrogen-source for mycelial growth of test-fungus was more effective than asparagioe regardless of cultural filtrate or stimulating substance added.
3. In a series of fractionation of the cultural filtrate, mycelial growth was shown only in un-volatile, ether insoluble, cation-adsorbed and anion-unadsorbed substance fractions, when rested in volatile, unvolatife, volatile acids, ether soluble organic adds, ether insoluble, cation-adsorbed, cation-unadsorbe, anion-adsorbed and anion-unadsorbed substance fractions. Among them, sclerotia wereproduced only on cation-adsoorbed fraction.
4. According, to the above resulte, it is suggested that substance for myoelial growth, substance for sclerotial production and inhibitor of sclerotial production are included in cultural filtrate but they are different with one another. It is further suggested that the former two substances are un-volatile, ether insoluble, and adsorbed to cation-exchange resin but not adsorbed to anioa, whereas the later one is unvolatile, ether insoluble, and not adsorbed to cation and anion exchange resin.
5. Seven amino acids, which are aspartic acid, cystine, glycine, histidine, lycine, tyrosine and dinitroaniline, were detected in fraction of substance adsorbed to cation-exchange resin by applying the paper chromatography improved with DNP-amino acids.
