Confirmation of stereotactic brain tumor model for drug efficacy using multimodal non-invasive molecular imaging

The preclinical use of imaging tools has potential to research and evaluate the drug development. Especially for the anticancer drug, the value of molecular imaging is rapidly emerging with many advantages. Orthotopic brain tumors proliferate rapidly and the degree of growth varies depending on the location of tumor implantation, making it more difficult to select a model suitable for drug efficacy evaluation. In this study, after U87MG cell line expressing Firefly luciferase stereotactic transplantation based on bregma, optical and MRI images were acquired. Photon values (Total and average) and tumor size for each individual were observed. As a result of comparing photon values for each object using optical images, four animals (12%) with an average photon value of 1x10^6 or lower were estimated to have not been established as tumors, and six animals (18%) with an average photon value of 1x10^7 or less were estimated to have small tumors. Out of a total of 33 animals, 70% were confirmed to have been established as models suitable for efficacy evaluation using optical imaging. The size and location of the tumor were confirmed as a result of the MRI image. Tumors were divided into those with a size of 5 mm^3 or less, 5-20 mm^3, and over 20 mm^3. No tumors could be identified by MRI images in 20% (7 out of 33). 9% less than 5 mm^3, 18% with 5-20 mm^3, and 53% with more than 20 mm^3. 70% with tumors of 5 mm^3 or larger are suitable for use in drug efficacy evaluation. Based on these results, we established a stereotactic brain tumor model, and presented a model validation method that can be used for drug efficacy evaluation using the presence or absence of tumor formation using BLI and the location and actual size of the tumor using MRI.
