Background: Light-emitting diodes (LEDs) have been shown to promote hair growth, but the efficacy is not consistent across clinical trials, and in vitro studies on human dermal papilla cells (hDPCs) have rarely been performed. We have previously shown that 660 nm is the most potent wavelength of LED light in the effect on hDPC proliferation.
Objectives: we examined whether LED light could antagonize IFN-γ-induced cellular changes in hDPCs via the JAK-STAT pathway
Methods: Various wavelengths of LED light were irradiated on IFN-r treated hDPCs. Cell proliferation was determined using a MTT assay. IFN-r Rc, STAT1 phosphorylation were analyzed by western blot analysis. mRNA expression of IFN-r-related genes was measured by real-time RT-PCR.
Results: Treatment with 660 nm light counteracted the effect of IFN-γ, whereas 415, 525, and 830 nm had no significant effect. IFN-γ treatment transiently induced IFN-γR expression and increased STAT1 phosphorylation, which persisted longer than the induced. All four wavelengths strongly inhibited IFN-γ-mediated activation of IFNrR and STAT1 phosphorylation. IFN-γ treatment increased the expression of IFN-γ-related genes such as caspase-1, IL-1ß and IL-18. All tested wavelengths suppressed the effect of IFN-γ on caspase-1 and IL-18 expression, but only 660 and 830 nm inhibited IL-1ß expression.
Conclusion: Our results suggest that LED light therapy can be used to treat AA by regulating IFN-r-induced changes at the cellular and genetic levels.
