Transglutaminase (TG) catalyse acyl transfer reactions, resulting in intermolecular crosslinking, underlying a number of physiological processes, such as receptormediated endocytosis, aging, immune reaction, fertilization, differentiation, and malignant transformation of cells, etc..
Elucidation of basic mechanism of the wound healing process is important, because there exist great needs for modulation of wound healing process whether promotion or inhibition. In wound healing, we assumed that the cross-linking of the intra and intercellylar molecules would play the major role. For the cross-linking of the molecules, transglutaminases are the major candidate to carry-out such functions. In the present study, we designed the animal experiment to determine the role of transglutaminases in wound healing by monitoring the changes in actifity and amount of transglutaminase in the tissue of 3rd degree burned skin, autogenous and homogenous-full-thicknss-grafted-skin in the function of time after infliction of wound. The samples were homogenized and separated into cytosol an microsomal fraction by ultracentrifugation.
For each of the samples, we analyzed the transglutaminase activity by radiolabelled-putrescine incorporation and amount change of the enzyme by western blot and ELISA methods.
The results showed significant increase of the enzyme activity from the point (about 4th wk) when though to be the basic wound healing process is ended and the breaking strength of wound began to increase with high velocity. Thereafter, the enzyme activity maintained the manner of persistent increasing pattern, while the amount of enzyme increased at high level during the early wound healing process and decreased as time passed toward the wound maturation phase.
Cytosol and microsomal transglutaminase showed a little difference in changes of both enzyme amount and activity in burned and homograft group. But in changes of enzyme amount of autograft group, only microsomal transglutaminase was varied, while cystol transglutaminse maintained its extremely low level throughout the wound healing process.
