Evaluation of skin sensitization potential is a major part of safety assessment of new ingredients in cosmetics and drugs to be applied topically. In recent years, it has been postulated that dendritic cells respond to a variety of chemical allergens by up-regulating expression of various co-stimulatory molecules. It might provide the basis for an in vitro alternative approach for the identification of contact sensitizers. Several studies have shown that contact sensitizers induce co-stimulatory molecules on human hematopoietic cell lines. This study attempts to investigate the utility of THP-1 cell line and their co-stimulatory molecule expressions as a predictive in vitro assay system for contact sensitization by chemicals. We investigated the expression of CD54 and CD86 on the THP-1 cells using flow cytometry after a 24 h exposure to known sensitizers such as 1-chloro-2,4-dinitrobenzene (DNCB), Hexylcinnamic aldehyde (HCA), Eugenol (Eu), Cobalt sulfate (Co) and known non-sensitizers such as Sodium lauryl sulfate (SLS), Benzalkonium chloride (BKC), Dimethyl sulfoxide (DMSO). The cytotoxicity of test materials was assessed with WST-1 assay to determine the appropriate dose and IC50 (50% inhibitory concentration) was obtained. Based on these data, three concentrations (0.1X, 0.5X, 1X of IC50) were selected. Relative fluorescence intensity (RFI) was used as an expression indicator. All allergens augmented CD54 and / or CD86 expression, while no augmentation was observed with non-allergens except SLS. From the expression of CD54 and / or CD86, we could classify the sensitizer / non-sensitizer. These data suggest that expression of CD54 and / or CD86 in THP-1 cells may be able to classify the potency of allergen, and this model can be a useful candidate for alternative test for skin sensitization.
