If the toxic inorgainc (heavy metals and others) and organic compounds (pesticides, industrial solvents, oil components, and detergents) from industry and other anthropogenic sources are relentlessly released into the environment including wastewater and soil without any prior treatment, they may contaminate the environment to a extent that the living ecology can be destructive and can not easily restore as original. Eventually, they may be taken up into the human body through a food web chain, resulting in a environmental disaster. Therefore, it is imperative to monitor the toxicity of environmental samples through the available bioassays which have been so far and will be further developed, so that we can safely protect ourselves from the haphazard. Once the results of toxicity test are obtained, the natural and engineered microorganisms can be applied to remediate the contaminated environment. In this paper, first of all, the kinds and requirements of the toxicity tests will be briefly introduced and explained. Equal mixture of cis- and trans-1,3-dichloropropene which is a fumigant for the agricultural purpose and golf course management in U.S.A. and other countries was used as model chemical for this paper Then, the actual experimental results obtained from the consecutive and extensive studies for the 1,3-D chemical will be presented to demonstrate the mechanism involved in the differential enhanced biodegradation of the cis- and trans-1,3-D in soil. The extensive differential enhanced degradation of 1,3-D was investigated and confirmed by Ou et al. (1995) and especially Chung et al. (1999). In addition, the involvement and isolation of microorganisms in the differential enhanced degradation of the 1,3-D were further investigated. Our experimental results showed that degradation of the cis- and trans-1,3-D was enhanced, with trans isomer being degraded faster than cis isomer, and the degradation rates were gradually increased with the consecutive applications of 1,3_D in field soil and faster in surface than subsurface soils. As expected, microorganisms (bacteria) were responsible for the enhanced degradation of the toxic chemicals. After many trials and errors, a mixed bacterial culture capable of degrading 1,3-D was initially isolated from an enhanced soil sample collected from a site treated with 1,3-D. Similar to the enhanced soil, the mixed culture degraded trans-1,3-D faster than cis-l,3-D. This mixed culture could not utilize cis- and trans-1,3-D as a sole source of carbon for growth. Rather, a variety of second substrates were evaluated to stimulate the differential enhanced degradation of the two isomers. As a result, the mixed culture degraded cis- and trans-1,3-D only in the presence of a suitable second substrate. Second substrates that had the capacity to stimulate the degradation included soil leachate, tryptone, tryptophan, and alanine. Other substrates tested, including soil extract, glucose, yeast extract, and indole failed to stimulate the degradation of the two isomers. Therefore, it appeared that the degradation of cis- and trans-1,3-D was a cometabolic process. The mixed culture was composed of at least four species of bacteria.
