Background: There are reports suggesting a possibility of relationship of Becker’s nevus (BN) to nerve distribution such as BN showing linear and segmental configuration. However, so far there are no studies regarding the expression of neuropeptides in lesions of BN. Objectives: To elucidate neuropeptide expression in Becker’s nevus. Methods: PCR array of 84 genes related to neuronal process was done on the lesion of BN and normal skin. Among the genes with 10 fold or more increase in lesional skin, real-time PCR was performed using both skin samples for neuropeptide Y (NPY), galanin and theirreceptors. Immunohistochemistry was done on both samples to look for the distribution and density of NPY, galanin and their receptors. Results: The results of PCR array revealed that 13 out of 84 genes related to neuronal process were increased by 10 fold or more in lesional skin of BN. In real-time PCR of NPY, galanin and their receptors, statistically significant increase of NPY, NPY2R and GAL1R was verified in lesional skin compared to normal skin. In immunohistochemistry, NPY, NPY1R and NPY2R wereall expressed in both lesional and normal skin. As for galanin, GAL1R and GAL2R, lesional skin showed higher degree of expression when compared to normal skin. Conclusion: NPY and galanin may play a role in the pathogenesis of BN. Further studies regarding molecular pathogenesis of these peptides and other possible factors influencing BN may provide clues in treating BN without recurrence.
