간 보호 효능에 대한 분자 기전 연구 : Glutathione S-Transferase 조절 = The Study of Molecular Mechanism in hepatoprotective Effect : The Role in Glutathione S-Transferase A2 Gene Induction
간은 외인적 물질을 대사하는 주 기관으로 간 기능의 특성상 산화적 스트레스에 지속적으로 노출된다. 본 연구에서는 간독성 동물모델에서 후보 물질에 의한 독성 방어 효능을 평가하고, 간독성에 대한 주요 방어기전으로 제 2상 해독화 유전자인 glutathione S-transferase A2 (GSTA2)의 발현 및 그 분자기전을 규명하였다.
1장에서는 발암원성 벤조피렌 (BP) 간독성 모델과 해열진통제인 아세트아미노펜 (APAP)의 과용량 간독성 모델에서 인삼의 간독성 방어효능을 평가하였다. 동양의학에서 수 천년 간 사용된 강장제인 인삼은 건조한 백삼 (KWG)과, 증기로 찐 홍삼 (KRG)으로 나뉘어 진다. 제조공정이 다른 백삼과 홍삼의 간독성 방어효능을 동물에서 관찰하고, 간 대사효소인 GSTs의 역할을 규명하였다.
담배 연기나 탄 음식 등에 함유되어 있는 BP은 대사에 의해 반응 중간체를 생성하여 DNA를 공격함으로서 발암성을 나타내는 화학물질이다. BP를 동물에 투여하였을 때 혈장내 독성지표인 aspartate aminotransferase (AST), alanine aminotransferase (ALT) 및 lipid peroxide (LPO)를 현저하게 증가시켰다. KWG를 4주 동안 전투여하였을 때, BP 매개성 생화학적 독성지표인 AST, ALT 및 LPO 활성 증가는 유의적으로 감소되었다. 친전자성 반응 중간체의 포합과정에 요구되는 glutathione (GSH) 및 GST 활성은 BP에 의하여 고갈되었고, KWG 전처리에 의하여 정상화되었다. 이러한 KWG의 간독성 지표의 개선효과는 조직 병리학적 관찰에서도 동일하게 나타났다. KWG의 간보호 효능에 대한 분자기전을 관찰하기 위해, 간세포주인 H4IIE에서 BP 대사에 관여하는 효소 발현변화를 관찰하였다. BP는 제 1상 간 대사효소인 cytochrome P450 1A1 (CYP1A1)에 의하여 친전자 반응중간체인 BP diol epoxide (BPDE)로 전환되어 거대분자를 공격한다. KWG는 BP의 독성 반응중간체의 형성에 관여하는 CYP1A1의 mRNA 및 protein 발현을 용량 의존적으로 저해시켰다. 제 2상 간 해독화 효소인 GST는 외인적 반응중간체를 포합하여 배설시키는 역할을 함으로서 독성물질에 의한 공격을 감소시킨다. 간세포주에 KWG를 투여하였을 경우, Nrf2 단백질의 전사 활성화를 매개하여 GSTA2 유전자를 현저히 증가하였으며, GST 단백질 증가를 수반하였다. 본 연구에서 KWG가 BP의 대사활성화에 의한 친전자성 반응중간체 생성의 감소 및 해독화 효소 유도를 통한 포합 배설의 촉진을 통하여 BP에 의한 간독성을 억제함을 규명하였다. KWG의 대사효소 발현조절을 매개한 BP의 간독성 보호 효과는 발암원에 대한 화학 예방제제로서의 가능성을 제시한다.
해열 및 진통제로 널리 사용되고 있는 아세트아미노펜 (APAP)은 고용량에서 심각한 간 괴사를 수반한 사망을 유발한다. APAP 고용량 투여군은 53%의 사망률을 나타내었으나, KRG를 1주일 동안 전투여하였을 때 APAP에 의한 사망률이 현저히 감소하였다. 중등도의 간독성을 유발하는 농도의 APAP는 동물의 혈장내 독성지표인 AST 및 ALT를 현저히 증가시켰다. KRG를 5주 동안 저용량으로 전투여하였을 때 APAP에 의한 AST 및 ALT 활성증가를 유의적으로 감소시켰다. 이러한 KRG의 간독성 지표의 개선효과는 조직병리학적 관찰에서도 동일하게 나타났다. KRG의 간보호 효능에 대한 분자 기전을 관찰하기 위해 간세포주인 H4IIE에서 제 2상 해독화 유전자인 GSTA2의 발현 및 상위 신호를 관찰하였다. 간세포주에 KRG를 투여하였을 경우 전사인자인 Nrf2 및 C/EBP의 핵 내 이동 및 전사활성화를 매개하여 GSTA2 유전자를 현저히 증가시켰다. KRG의 GSTA2 발현 및 전사활성화는 세포내 주된 신호 경로인 JNK, PI3K 및 PKA의 다중 신호계의 조절을 받는다. 본 연구에서 APAP 독성에 대한 KRG의 사망률 개선 및 간독성 보호 효과는 APAP를 포함하는 독성물질의 노출에 의한 간독성을 KRG가 예방할 수 있을 것으로 사료된다.
2장에서는 인삼의 간보호 효능에 있어 유효한 작용을 갖는 saponin의 구성성분을 규명하고, 나아가 천연물에서 기원한 prototype인 isothiocyanate (ITCs)계 화합물의 GSTA2 발현을 간세포주에서 비교하여 효능이 좋은 후보물질을 선별하고 그 분자기전을 연구하였다.
인삼의 생리활성 효능은 그 구성 성분인 saponin에 의해 조절된다. 간보호 효능에 있어서 GSTA2를 유도하는 KRG의 성분을 규명하기 위하여 KRG의 주요 구성 saponin에 의한 효소 유도능을 관찰하였다. 홍삼 제조과정에서 특이적으로 생성되는 ginsenosides Rg3는 GSTA2를 농도 의존적으로 증가시켰으며, 높은 효력을 나타내었다. Ginsenoside Rb1, Rc 및 Rg1에서는 GSTA2 발현 변화는 관찰되지 않았다. Ginsenoside Rg3의 GSTA2 발현은 JNK, PI3K 및 PKA의 신호 경로 조절을 매개한 Nrf2 및 C/EBP의 활성화에 의존하는 것으로 관찰되었다. 본 연구에서는 ginsenoside Rg3가 KRG의 GSTA2 유전자 발현에 대한 주요 성분임을 처음으로 규명하였다.
천연물에서 기원한 prototype인 isothiocyanates (ITCs)계 화합물은 독성물질에 대한 암 발생을 억제하는 화학예방제제이다. 본 연구는 7종의 ITCs 계열 화합물에 의한 GSTA2 유도 등을 비교한 결과 간세포에서 PEITC의 GSTA2 발현이 가장 현저하게 유도되었다. 또한, PEITC는 리포터 유전자 활성법을 통한 GSTA2 전사 활성도 증가시켰다. PEITC는 GSTA2 전사인자인 Nrf2 및 C/EBP의 핵 내 이동을 현저히 유도하였다. ARE 및 C/EBP binding site가 결핍된 plasmid vector의 형질도입에 의한 전사활성 억제는 PEITC의 GSTA2 유도가 전사인자의 전사활성화를 매개함을 증명하였다. PEITC의 GSTA2 유전자 발현 및 전사활성에 대한 상위 신호는 JNK, PI3K 및 PKA의 다중 신호계이다. 본 연구에서 도출된 PEITC의 GSTA2 유전자 발현 유도는 화학예방 활성에 있어 중요한 방어 기전이며, PEITC의 화학예방제제로서의 가능성을 제시한다.
종합하면, 본 연구에서는 후보물질들의 간독성 방어 효과를 대표적인 독성 유발 물질인 BP 및 APAP에 의한 간독성 모델에서 관찰하였다. 후보물질들은 동물모델에서 독성물질에 의한 심각한 간손상을 정상으로 회복시켰으며, 간세포에서 독성물질의 대사활성화를 억제하고 해독화에 기여하는 효소를 유도함으로써 독성물질의 대사를 조절하였다. 효소 발현의 상위 신호 및 전사활성화를 규명하여 후보물질의 세포내 분자기전을 증명하였다. 본 연구로부터 도출된 간 대사효소 발현의 제어 및 상위신호의 발굴은 간독성 방어에 중요한 약리학적 타겟을 제공하며, 천연 후보물질로부터 간보호 효능을 스크린 하는데 있어서 활용될 수 있는 간보호 활성 효능 검색계를 제공한다.
Liver plays a major role in metabolism of xenobiotics and is continuously exposed to oxidative stress. This study aimed to evaluate the hepatoprotective effects of natural substance on hepatotoxicity in vivo and molecular mechanism for hepatoprotection focused on metabolic phase II enzyme in vivo and in vitro.
Chaptor. 1. I evaluated the hepatoprotective effects of ginseng against benzo[a]pyrene (BP) and acetaminophen (APAP) induced liver toxicity in mice. Ginseng is a well-known medicinal herb in traditional Asian medicine and is considered as an adaptogen. Of the two kinds of ginseng, white ginseng is air-dried ginseng, and red ginseng is produce by steaming raw ginseng. First, I examined the hepatoprotective effect of Korea White Ginseng (KWG) and Korea Red Ginseng (KRG) in mice. Moreover, I evaluated the role of liver metabolic enzyme such as GSTs.
BP, a prototypic PAH, is the most potent carcinogens found in cigarette smoke, charred foods and petroleum by products. The carcinogenicity of BP requires metabolic activation to generate electrophilic intermediates that covalently bind to critical DNA targets to initiate carcinogenesis. BP caused severe liver injury in rats, as indicated by elevated plasma ALT, AST and LPO levels. Pretreatment with KWG for 4 weeks completely abrogated increases in the ALT, AST and LPO levels when challenged with BP. BP resulted in decrease of the hepatic glutathione (GSH) and glutathione S-transferase (GST) activity. However, the pretreatment returned GST activity to control levels. These protective effects of KWG against BP-induced toxicity were consistent with the results of histopathological examinations. I next examined the effects of KWG on the gene expression of the enzymes that metabolize BP in H4IIE cells. The major activation pathway for the BP is mediated by the CYP1A1, ultimately leading to the highly mutagenic BP diol epoxide (BPDE); this compound is believed to be the ultimate carcinogenic metabolite of BP. KWG moderately inhibited BP-induced CYP1A1 mRNA and protein expression. The GSTs are a family of enzymes that assist in the excretion of carcinogens by making them hydrophilic via conjugation with GSH. The GSTA2 gene expression was significantly increased by KWG through the Nrf2/antioxidant responsive element pathway for enzyme induction.
These results suggest that KWG is efficacious in protecting against BP-induced hepatotoxicity as results of metabolic regulations through both the inhibition of metabolic enzyme activation and the enhancement of electrophilic detoxification, implying that KWG should be considered a potential chemopreventive agent.
Acetaminophen (APAP) is a broadly used analgesic in the global. When high doses of APAP treated in laboratory animals or non-alcoholic humans, causes centrilobular hepatic necrosis by acetaminophen. A single dose of APAP in mice caused severe liver injury leading to acute lethality of 53%. Pretreatment with KRG for 1 week significantly reduced APAP-induced mortality analyzed by Kaplan-Meier’s survival test. APAP exposure also markedly increased AST and ALT activities compared to control. Pretreatment of KRG for 5 weeks significantly abrogated the increases in AST activities by APAP. These protective effects of KRG against APAP were consistent with the results in histopathological examinations. To evaluate the molecular mechanism of the protective effect of KRG, we next determined the detoxifying enzyme expression and upstream signaling. H4IIE cells treated with KRG significantly increased hepatic GSTA2 mRNA levels and protein expressions in a dose-dependent manner. Exposure of KRG also increased the levels of nuclear factor-erythroid 2 related factor2 (Nrf2) and CCAAT/Enhancer Binding protein (C/EBP), which are critical transcriptional factors for the enzyme induction. These effects are downstream of multiple cellular signaling including PI3K, JNK or PKA. These results suggest that KRG has protective effect against APAP induced liver toxicity and mortality.
Chaptor. 2, I investigated of effective ginseng saponins in hepatoprotective effect and effective candidates on GSTA2 gene induction by ITCs compounds in H4IIE cells. Moreover, I examined the molecular mechanism of GSTA2 gene induction. Ginsenosides are components of ginseng saponin and they possess various physiological and pharmacological activities. The observation of a major compound of KRG for the GSTA2 gene induction among saponin revealed that ginsenoside Rg3 significantly increased GSTA2 protein expression, whereas Rb1, Rc and Rg1 failed to induce. Ginsenoside Rg3 also increased transcriptional activity of GSTA2 through Nrf2 and C/EBP downstream of PI3K, JNK or PKA, using ARE reporting analysis. Taken together, we clearly demonstrate that KRG is efficacious in protecting in APAP-induced hepatotoxicity through GST induction, and that Ginsenoside Rg3 is as a major component of KRG for the GST induction implying that Ginsenoside Rg3 should be considered a potential hepatoprotective agent.
Although isothiocyanates have been shown to inhibit carcinogen-induced tumorigenesis through the metabolic regulation, limited studies have been made to determine their potential for regulation of detoxifying enzyme. Therefore, the present study was designed to investigate the structure-activity relationships of these seven arylalkyl ITCs for their effects on detoxifying enzymes. In vivo experiment, PEITC among arylalkyl ITCs resulted in potent and sustained induction of GSTA2 mRNA by 8~12-fold. Similarly, PEITC is the most efficacious for the GSTA2 induction in the hepatocyte-derived H4IIE cells with BITC showing less induction. Therefore, we focused on the effect of PEITC to elucidate the molecular mechanism of GSTA2 induction. The cells treated with PEITC significantly increased hepatic GSTA2 mRNA levels and protein expressions in a dose-dependent manner. In addition, PETIC also potently increased promoter activity of GSTA2 using vector containing the -1.65 kb GSTA2 promoter region. PEITC resulted in nuclear translocation of NF-E2-related factor-2 (Nrf2) and CCAAT/enhancer binding protein (C/EBP). The cells transfected with deleted promoter construct pGL-ΔARE and pGL-ΔC/EBP fail to increase their activity by PEITC confirming the critical role of these transcriptional factors for the enzyme induction. Furthermore, we found that the multiple signaling of JNK, PI3K and PKA were involved for the induction and transactivation of GSTA2 by PEITC. Taken together, these results suggest that the induction of GST is an important mechanism for the chemopreventive activity of ITCs implying the most potent potential of PEITC among other ITC homologous.
Taken together, in this study, I evaluated the protective effect of natural compound. BP or APAP induced hepatotoxicity and digged out pharmacological targets through against the induction of detoxification enzyme and their upstream signal pathways. In addition, this study provides a screening system of hepatoprotective effect among natural candidate compounds.
