Differentiation of mesenchymal stem cells into vascular cellss under mechanical stimulation for bioengineered blood vessels = Bioengineered 血管을 위한 力學的 刺戟을 利用한 中間曄 줄기細胞의 血管細胞로의 分化
For successful outcomes in tissue engineering studies for bio-blood vessels, three major factors must coincide: scaffolds, cells, and biomimetic environments. Especially, a blood vessel has a layered structure, and a different cell type resides in each layer. Among the different cell types, endothelial cells (ECs) and smooth muscle cells (SMCs), which play important roles in functionalities of blood vessels, are the current focus in many related research areas. Compared to SMCs, ECs are known to be difficult to culture in vitro, and even when they are apparently cultured in vitro, they easily lose the original functions that they had in the human body because they are cultured in totally different environments compared to those in the human body. Futhermore, it is widely known that ECs orientation is needed on tubular type scaffolds for the successful regeneration, and this orientation affects original functions of blood vessels. To induce cellular orientation, mechanical stimuli have been applied for regeneration of blood vessels. Most related research have been focused on shearing stress because the shearing stress affects MSCs differentiation into ECs and induce cellular orientation.
In this study, based on the theory that types and magnitudes of initial mechanical stimuli play important roles in MSCs' fate, we studied the early MSCs' fate under various types of shear stress along with biochemical reagents of EC-related growth factors. For these studies, we developed a flow engaging bioreactor system to produce mechanical stimuli for the vascular regeneration. Two types of shear stress, high shear stress (10 dyne/cm2) and low shear stress (2.5 dyne/cm2) were applied on MSCs for 24 hours and we observed the effects of types of shear stress along with chemical stimulation (endothelial cell growth medium; EGM-2) on early MSC differentiation into EC like cells. From the results, markers related to SMC differentiation were up-regulated under high shear stress condition and low shear stress condition induces EC related markers. Based on these results, we confirmed that low shear stress may be more efficient to determine MSC's early fate to the EC lineage, and tried to determine more efficient duration of the stimulation. All mRNA levels related to EC makers were significantly up-regulated at day 1, and in the results of immunofluorescence staining, vWF, a mid-EC marker, was highly expressed at day 2 and kept their levels for the culture time.
Based on these results, we suggested MSC's fate into EC like cells might be determined within 24 hours under low shear stress condition, and we tried to induce SMC differentiation of MSCs onto outer layer of the scaffolds applying flow induced stretch after 1 day low shear stress. 3% stretch for 3 days and 5% stretch for 4 days were applied consequently. From the results, the mRNA levels were not increased under 3% stretch, but the levels were significantly up regulated with 5% stretch. Flow cytometry analysis also confirmed that 5% stretch for 24 hours induced expressions of EC and SMC markers. Therefore, we suggest that combined conditions of mechanical stimulation (2.5 dyne/cm2 shear stress for 1 day + 3% stretch for 3 days + 5% stretch for 1 day) may be more efficient for EC and SMC differentiations of MSCs onto layered tubular type scaffolds.
